Characteristics of escape mutations from occult hepatitis B virus infected patients with hematological malignancies in South Egypt

AIm To investigate the prevalence and virological characteristics of occult hepatitis B virus(HBV) infections in patients with hematological malignancies in South *** Serum samples were collected from 165 patients with hematological malignancies to monitor titers of HBV dNA, hepatitis B surface antigen(HBs Ag), and antibodies to HBV core(anti-HBc) and surface antigens. Serum samples negative for HBs Ag and positive for anti-HBc were subjected to nucleic acid extraction and HBV dNA detection by real-time polymerase chain reaction. dNA sequences spanning the S region were analyzed in cases with occult HBV infection. In vitro comparative study of constructed 1.24-fold wild type and S protein mutant HBV genotype d clones was further performed. RESULTS HBV dNA was detected in 23(42.6%) of 54 patients with hematological malignancies who were HBsA g negative, but anti-HBc positive, suggesting the presence of occult HBV infection. The complete HBV genome was retrieved from 6 occult HBV patients, and P120 T and S143 L were detected in 3 and 2 cases, respectively. Site directed mutagenesis was done to produce 1.24-fold genotype d clones with amino acid mutations T120 and L143. The in vitro analyses revealed that a lower level of extracellular HBsA g was detected by chemiluminescence enzyme immunoassay(CLEIA) with the clone containing T120 mutation, compared with the wild type or the clone with S143 L mutation despite the similar levels of extracellular and intracellular HBs Ag detected by Western blot. Southern blot experiments showed that the levels of intracellular HBV dNA were not different between these clones. CONCLUSION Occult HBV infection is common in patients with hematological malignancies and associated with P120 T and S143 L mutations. 120 T mutation impairs the detection of HBsA g by CLEIA.

Iterative computational approach to the solution of the hamilton-Jacobi-Bellman-lsaacs equation in nonlinear optimal control

In this paper, iterative or successive approximation methods for the hamilton-Jacobi-Bellman-lsaacs equations （HJBIEs） arising in both deterministic and stochastic optimal control for affine nonlinear systems are developed. Convergence of the methods are established under fairly mild assumptions, and examples are solved to demonstrate the effectiveness of the methods. However, the results presented in the paper are preliminary, and do not yet imply in anyway that the solutions computed will be stabilizing. more improvements and experimentation will be required before a satisfactory algorithm is developed.

The anticancer activity of stevenleaf(SV)on the basis of cell viability,cell cycle,and apoptosis induction in HepG2 cancer cells were *** controlled the growth of HepG2 cells with IC50 of 139.82μmol/L for 24 h,IC50 of 119.12μmol/L for 48 h and cell cycle arrested at G0/G1 phase,induced cell apoptosis and enhanced intracellular ROS *** cell cycle arrest,the mRNA expression levels of p21,p27 and p53 were up-regulated,while the expression levels of Cyclin A,Cyclin d1,Cyclin E and CdK1/2 were *** efficiently up-regulated TNF R1,TRAdd1 and FAdd and down-regulated Caspase8 for cell death receptors;similarly,up-regulated Bax,Bak,Cytc,Apaf1,Caspase3 and Caspase9,and down-regulated Bcl2,Bcl xl and Bad for mitochondrial signal *** induced the mTOR-mediated cell apoptosis in HepG2 cells via activation of Akt and *** mechanistic explanation for the anticancer activity of SV as functional food can be derived from above results.

Spectral Properties of [(Ni0.45Co0.2Zn0.35F2O4)1–x(Sodium Acetylacetonate)x] Nanocomposite

The pure spinel phase of Ni0.45Co0.2Zn0.35F2O4 was prepared by coprecipitation method and ceramic method. The nanocomposite [(NCZF)1–x(Na(***))x] (x = 0%, 20%, 40%, 60%, 80%, 100%) was prepared by mixing two phases, Ni0.45Co0.2Zn0.35F2O4 with coprecipitation method [NCZF] and sodium acetylacetonate [Na(***)]. The TEm photographs of the as prepared ferrite samples by coprecipitation method at different ratios of Na(***) show that the crystalline cubic nanoparticles diameter were ranged from 26 - 36 nm and the particle size of Na(***) is greater than NCZF. The same behavior has also confirmed by XRd pat-terns indicating polycrystalline nature of prepared sample. The comparison between the SEm micrographs for NCZF nanoparticles prepared by a) ceramic method and b) co-precipitation method was done to show the effect of preparation condition on the microstructure. IR analysis confirms the composites formation.

Quaternion algebra has been used to apply the fractional Fourier transform(FrFT)to color images in a comprehensive ***,the discrete fractional random transform(dFRNT)with adequate basic randomness remains to be *** paper presents a novel multistage privacy system for color medical images based on discrete quaternion fractional Fourier transform(dQFrFT)watermarking and three-dimensional chaotic logistic map(3d-CLm)***,we describe quaternion dFRNT(QdFRNT),which generalizes dFRNT to handle quaternion signals effectively,and then use QdFRNT to perform color medical image adaptive *** efficiently evaluate QdFRNT,this study derives the relationship between the QdFRNT of a quaternion signal and the four components of the dFRNT ***,it uses the human vision system's(HVS)masking qualities of edge,texture,and color tone immediately from the color host image to adaptively modify the watermark strength for each block in the color medical image using the QdFRNT-based adaptive watermarking and support vector machine(SVm)*** limitations of watermark embedding are also explained to conserve watermarking ***,3d-CLm encryption is employed to improve the system's security and efficiency,allowing it to be used as a multistage privacy *** proposed security system is effective against many types of channel noise attacks,according to simulation results.

Electric and magnetic Properties of [(NCZF)1-x(Na(***))x] Nanocomposite

The pure spinel phase of Ni0.45Co0.2Zn0.35F2O4[NCZF] was prepared by co-precipitation method. The nanocomposite [(NCZF)1-x(Na(***))x] (x = 0%, 20%, 40%, 60%, 80%, 100%) was prepared by mixing two phases: ferrite phase and ferroelectric phase. The magnetic characters of compo-sites performed using vibrating sample magnitude (VSm) at room temper-ature with maximum applied field up to 2 kG. For smaller particle size 30.89 nm. The saturation magnetization ms decreases linearly with increasing particle size. The effect of temperature on the electrical resistivity of the different composites samples was studied. It is noted that, the resistivity decreases with rising temperature to have a minimum value at ferroelectric transition temperature, above this value the resistivity increases. The dielectric constant increases with increasing temperature for all composites. The dielectric loss tan (δ) as a function of temperature sharply with temperature reached to maximum value at ferroelectric phase transition then decrease again.

Efficiency of combining pomegranate juice with low-doses of cisplatin and taxotere on A549 human lung adenocarcinoma cells

Objective: To test the coalescence effect of two chemotherapy drugs at low effective dose(cisplatin and taxotere) combined with pomegranate juice on A549 cancer cells. methods: Infrared spectroscopy method is a qualitative test that was performed to ensure the existence of the phytochemicals providing the antioxidant activity through the presence of the hydroxyl group(-OH). The viability of A549 cell line and normal mCs was tested using the neutral red uptake, Clonogenic survival, XTT and Cell migration assays. Results: Our results showed that this combination firstly led to a greater decrease in the viability of cells comparing to those treated with chemotherapy drugs alone, and secondly led to a significant reduction in cell migration. Conclusions: These data suggest a synergistic effect between the pomegranate and cisplatin which makes probably this combination a powerful option for treating lung adenocarcinoma and in parallel minimizing the systemic side effects.

Synthesis, Characterization and In VitroAntitumor Evaluation of New Pyrazolo[3,4-d]Pyrimidine derivatives

A new series of 3-(methylthio)-1-phenyl-1H-pyrazolo[3,4-d]pyrimidine derivatives was synthesized. The structures of the new derivatives were confirmed by the spectral data and elemental analyses. The antitumor activity of this series against human breast adenocarcinoma cell line mCF7 was evaluated. Out of twenty new derivatives, ten were revealed mild to moderate activity compared with doxorubicin as a reference antitumor. Among this new series N-(2-chlorophenyl)-2-(3-(methylthio)-4-oxo-1-phenyl-1H-pyrazolo[3,4-d]pyrimidin-5(4H)-yl)acetamide (13a) was found the most active one with IC50 equal to 23 μm.

绞股蓝Gynostemma pentaphyllum富含多种营养成分,具有多种功能,例如治疗血尿、咽和颈部水肿、抗肿瘤等。天然产物的结构不同,它们表现出不同的抗菌和抗肿瘤机制。本论文中,我们调查了绞股蓝皂苷对人肝癌细胞Hep G2、绞股蓝皂苷ⅩⅦ对人宫颈癌Hela细胞的抑制作用,并进一步研究了其分子机制。主要研究结果如下:（1）绞股蓝皂苷对人肝癌细胞Hep G2的抑制作用及其分子机制。在细胞存活率、细胞周期和诱导Hep G2癌细胞凋亡的基础上,评估了Stevenleaf（SV）的抗癌活性。SV处理诱导Hep G2停滞在G0/G1期,24h和48h的IC50值分别139.82μm和119.12μm;SV处理诱导诱导细胞凋亡并增强细胞内ROS的产生。对于细胞周期相关基因,SV上调p21、p-27、p-53的m RNA表达,下调Cyclin-A、Cyclin d1、Cyclin-E和CdK1/2。SV有效促进细胞受体相关TNF-R1、TRAdd1和FAdd的表达、下调Caspase-8基因的表达;SV处理后,线粒体途径的Bax、Bak、Cyt c、Apaf1、Caspase 3和Caspase9基因表达量上调,Bcl2、Bcl-xl和Bad基因的表达量下调。SV通过Akt和AmPK激活在Hep G2细胞中诱导m TOR介导的细胞凋亡。（2）绞股蓝皂苷ⅩⅦ对人宫颈癌Hela细胞的抑制作用及其分子机制。mTT法用于评估gypenoside ⅩⅦ对Hela细胞的抑制作用,通过Hoechst 33342染色和Annexin V-PI染色鉴定凋亡细胞死亡和凋亡,细胞周期分析用于鉴定细胞四个阶段（G0/G1,S和G2/m期）中的细胞比例。gypenoside ⅩⅦ处理后,细胞的抑制作用随浓度的增加而增加,细胞明显凋亡。与正常细胞相比,G0/G1期的细胞比例增加,而S和G2/m期的细胞比例降低。此外,gypenoside ⅩⅦ处理有效调控死亡受体途径和线粒体途径的基因表达。通过Caspase-8和caspase-9蛋白抑制剂的作用,进一步证明了gypenoside ⅩⅦ诱导Hela细胞的凋亡。上述结果表明绞股蓝皂苷和绞股蓝皂苷ⅩⅦ对癌症细胞具有显著的抑制作用,具有进一步开发为抗癌药或功能性食品的价值。

On the mHd squeeze flow between two parallel disks with suction or injection via ham and HPm

An analysis has been performed to study the problem of magneto-hydrodynamic （mHd） squeeze flow of an electrically conducting fluid between two infinite, parallel disks. The analytical methods called Homotopy Analysis method （ham） and Homotopy Perturbation method （HPm） have been used to solve nonlinear differential equations. It has been attempted to show the capabilities and wide-range applications of the proposed methods in comparison with a type of numerical analysis as Boundary Value Problem （BVP） in solving this problem. Also, the velocity fields have been computed and shown graphically for various values of physical parameters. The objective of the present work is to investigate the effect of squeeze Reynolds number, Hartmann number and the suction/injection parameter on the velocity field. Furthermore, the results reveal that ham and HPm are very effective and convenient.
