Preparation of human decellularized peripheral nerve allograft using amphoteric detergent and nuclease

Animal studies have shown that amphoteric detergent and nuclease(DNase I and ribonuclease A) is the most reliable decellularization method of the peripheral nerve. However, the optimal combination of chemical reagents for decellularization of human nerve allograft needs further investigation. To find the optimal protocol to remove the immunogenic cellular components of the nerve tissue and preserve the basal lamina and extracellular matrix and whether the optimal protocol can be applied to larger-diameter human peripheral nerves, in this study, we decellularized the median and sural nerves from the cadavers with two different methods: nonionic and anionic detergents(Triton X-100 and sodium deoxycholate) and amphoteric detergent and nuclease(3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate(cHAPS), deoxyribonuclease I, and ribonuclease A). All cellular components were successfully removed from the median and sural nerves by amphoteric detergent and nuclease. Not all cellular components were removed from the median nerve by nonionic and anionic detergent. Both median and sural nerves treated with amphoteric detergent and nuclease maintained a completely intact extracellular matrix. Treatment with nonionic and anionic detergent decreased collagen content in both median and sural nerves, while the amphoteric detergent and nuclease treatment did not reduce collagen content. In addition, a contact cytotoxicity assay revealed that the nerves decellularized by amphoteric detergent and nuclease was biocompatible. Strength failure testing demonstrated that the biomechanical properties of nerves decellularized with amphoteric detergent and nuclease were comparable to those of fresh controls. Decellularization with amphoteric detergent and nuclease better remove cellular components and better preserve extracellular matrix than decellularization with nonionic and anionic detergents, even in large-diameter human peripheral nerves. changes on optical coherence tomography angiography and fluorescein angiography in eyes with neovascular age-related macular degeneration

AIM:lo evaluate the changes on optical coherence tomography angiography(OcTA) and fluorescein angiography(FA) and their correlation in neovascular agerelated macular degeneration(nAMD) before and after intravitreal aflibercept injections(IAIs).METHODS:In 43 treatment-na?ve patients with nAMD,choroidal neovascularization(cNV) in OcTA were morphologically and quantitatively analyzed before and after IAIs to determine whether they are correlated with leakage on FA or *** combining cNV in OcTA and leakage in FA,lesions were characterized as three types:l+c+(with both cNV and leakage),l-c+(with cNV but without leakage),or l+c-lesion(with leakage outside cNV).RESUlTS:Before IAI,while 27 eyes had l+c+lesion only,16 eyes had both l+c+and l-c+lesions *** capillaries and anastomosis in cNV were more developed in l+c+lesion,at 86.0% and58.1%,respectively,relative to 9.3% and 9.3% in l-c+lesions(Pcapillaries and anastomosis were decreased in the lesions with cessation of leakage on FA(Pctively).In quantitative analysis,neovascularization length and numbers of junctions and endpoints were also significantly ***:leakage on FA is associated with cNV morphology in OcTA and remained so after ***,by carefully assessing the morphological and quantitative changes of cNV in OcTA before and after treatment,activity of nAMD is expected even though cNV on OcTA is not completely matched with fluorescein leakage.

Pollen tube growth is essential for successful double fertilization, which is critical for grain yield in crop plants. Rapid alkalinization factors(RAlFs) function as ligands for signal transduction during fertilization. However, functional studies on RAlF in monocot plants are ***, we functionally characterized two pollen-specific RAlFs in rice(Oryza sativa) using multiple clustered regularly interspaced palindromic repeats(cRISPR)/cRISPR-associated protein 9-induced loss-of-function mutants,peptide treatment, expression analyses, and tag reporter lines. Among the 41 RAlF members in rice, OsRAlF17 was specifically expressed at the highest level in pollen and pollen *** applied OsRAlF17 or OsRAlF19peptide inhibited pollen tube germination and elongation at high concentrations but enhanced tube elongation at low concentrations, indicating growth regulation. Double mutants of OsRAlF17 and OsRAlF19(ralf17/19) exhibited almost full male sterility with defects in pollen hydration, germination, and tube elongation,which was partially recovered by exogenous treatment with OsRAlF17 peptide. This study revealed that two partially functionally redundant OsRAlF17 and OsRAlF19 bind to Oryza sativa male-gene transfer defective 2(OsMTD2) and transmit reactive oxygen species signals for pollen tube germination and integrity maintenance in rice. Transcriptomic analysis confirmed their common downstream genes, in osmtd2 and ralf17/19. This study provides new insights into the role of RAlF, expanding our knowledge of the biological role of RAlF in regulating rice fertilization.

Rapid technical advances in the field of computeropened new avenues in animal behavior *** （cA） and virtual reality （VR） haveAnimated stimuli are powerful tools as theyoffer standardization, repeatability, and complete control over the stimulus presented, thereby＂reducing＂ and ＂replacing＂ the animals used, and ＂refining＂ the experimental design in line withthe 3Rs. However, appropriate use of these technologies raises conceptual and technical *** this review, we offer guidelines for common technical and conceptual considerations related tothe use of animated stimuli in animal behavior research. Following the steps required to create ananimated stimulus, we discuss （I） the creation, （11） the presentation, and （111） the validation of cAsand VRs. Although our review is geared toward computer-graphically designed stimuli, consider-ations on presentation and validation also apply to video playbacks. cA and VR allow both new be-havioral questions to be addressed and existing questions to be addressed in new ways, thus weexpect a rich future for these methods in both ultimate and proximate studies of animal behavior.

Dear Editor, The lATERAl ORGAN BOUNDARIES DOMAIN （lBD）/ ASYMMETRIclEAVES2-lIKE （ASl） genes （hereafter referred to as lBD） encode proteins containing a conserved plant-specific lOB domain and play roles in lateral organ development （Iwakawa et al., 2002; Shuai et al., 2002; Majer and Hochholdinger, 2011）. The lOB domain is approximately 100 amino acids in length and contains a conserved four-cys motif （cX2cX6cX3c）, the Gly-Ala-Ser block （GAS）, and the leucine-zipper-like coiled-coil motif （lX6lX3lX6l）（Shuai et al., 2002）. The leucine-zipper-like coiled-coil motif in the lOB domain is predicted to function in protein dimerization. lOB, AS2, and lBD4 preferentially bind unique DNA sequences in electrophoretic mobility shift assays （EMSAs）（Husbands et al., 2007）. Modulatory effects of the fruits of Tribulus terrestris l. on the function of atopic dermatitis-related calcium channels,Orai1 and TRPV3

Objective: To examine the effects of Tribulus terrestris l.(T. terrestris) extract on the modulation of calcium channels to evaluate its use in topical agents for treatment of atopic ***: The 70% methanol extract of T. terrestris was prepared. Human HEK293 T cells with over-expressed calcium release-activated calcium channel protein 1(Orai1),transient receptor potential vanilloid 1, or transient receptor potential vanilloid 3(TRPV3)were treated with T. terrestris extract. Modulation of ion channels was measured using a conventional whole-cell patch-clamp ***: T. terrestris extract(100 mg/m l) significantly inhibited Orai1 activity in Orai1-stromal interaction molecule 1 co-overexpressed HEK293 T cells. In addition, T. terrestris extract significantly increased the TRPV3 activity compared with 2-Aminoethyl diphenylborinate(100 mmol/l), which induces the full activation of ***: Our results suggest that T. terrestris extract may have a therapeutic potential for recovery of abnormal skin barrier pathologies in atopic dermatitis through modulating the activities of calcium ion channels, Orai1 and TRPV3. This is the first study to report the modulatory effect of a medicinal plant on the function of ion channels in skin barrier.

Ethanol Extract of lycopodium serratum Thunb. Attenuates lipopolysaccharide-Induced c6 Glioma cells Migration via Matrix Metalloproteinase-9 Expression

Objectives： To elucidate how ethanol extract of l. serratum（ElS） could exert anti-migratory effects on glioma with the suppression of nuclear factor kappa B（NF-κB） downstream pathway. Methods： cell viability of ElS on c6 glioma was detected by 3-（4,5-dimethylthiazol-2-yl）-2,5-diphenyltetrazolium bromide（MTT） assay. Nitric oxide（NO） assay and 2＇,7＇-dichlorofluorescin diacetate（DcFH-DA） assay were applied to measure NO production and reactive oxygen species（ROS） generation on lipopolysaccharide（lPS）-induced c6 glioma cells. NF-κB, mitogen-activated protein kinase（MAPK）, inducible nictric oxide synthase（i NOS） and cyclooxygenase-2（cOX-2） protein were determined by Western blot. Wound healing assay was used to investigate the inhibitory effect of ElS on fetal bovine serum（FBS）-induced migration and matrix metalloproteinase（MMP）-9 and-2 activity was examined by zymography. Results： ElS suppressed lPS-induced phosphorylation of extracellular signal-regulated kinase（ERK）, c-Jun N-terminal kinase（JNK）, and p38 through inhibiting the expression of chemokine ccl2（or monocyte chemoattractant protein-1, McP-1）. In addition, ElS inhibited the expression of i NOS, cOX-2, and the production of NO by lPS in c6 glioma cells. ElS also significantly decreased serum-induced migration of c6 glioma cells in scratch wound healing in a dose-dependent manner（P〈0.01）. The activity of MMP-9 and-2 were also significantly attenuated by ElS with lPS treatment（P〈0.01）. conclusion： Our results suggest that downregulation of MMP-9 gene expression might be involved in the anti-migration effect of ElS against lPS-induced c6 glioma cells.

Biphasic Nano-Domains of Planar lipid Bilayer complexed with Fluorogenic Polymer Reporter Tailored for Antimicrobial Detection

We have fabricated an unexpected type of supported planar bilayer composed of receptor phospholipids and single-chained diacetylenes as fluorogenic reporters using protruded anchor moieties with a positive terminal *** topographical and surface thermodynamic analyses,as well as molecular dynamics simulations,revealed the coexistence of well-dispersed liquid-condensed(l_(c))domains forming nano-islands and liquid-expanded(l_(e))region in the planar bilayer,enhancing sensitivity against a prototype of ubiquitous membrane-associated antimicrobial peptides,*** l_(e)regions,acting as target receptors,enabled sensitive detection as the melittin adsorbed and inserted into these regions due to strong hydrophobic interactions between phospholipids and *** l_(c)domains,serving as signal reporters,enabled diacetylenes to assemble,polymerize,and fluoresce in response to the insertion of melittin into the l_(e)***,biphasic nanodomains of the planar lipid bilayer finally endowed this sensor system with a detection range of 100μMto 50 nM and a limit of detection(lOD)of∼37 nM for *** exceeded the operational performance of the colorimetric polydiacetylene vesicle solution 45 times,which reportedly ranged from 100 to 4μM with an lOD of∼1.7μM.

Advancements in diagnostic systems for metastatic cancer over the last few decades have played a significant role in providing patients with effective treatment by evaluating the characteristics of cancer *** the progress made in cancer prognosis,we still rely on the visual analysis of tissues or cells from histopathologists,where the subjectivity of traditional manual interpretation *** paper presents the development of a dual diagnosis and treatment tool using an in vitro acoustic tweezers platform with a 50MHz ultrasonic transducer for label-free trapping and bursting of human breast cancer *** cancer cell detection and classification,the mechanical properties of a single cancer cell were quantified by single-beam acoustic tweezers(SBAT),a noncontact assessment tool using a focused acoustic ***-mimicking phantoms and agarose hydrogel spheres(AHSs)served to standardize the biomechanical characteristics of the *** on the analytical comparison of deformability levels between the cells and the AHSs,the mechanical properties of the cells could be indirectly measured by interpolating the Young’s moduli of the *** a result,the calculated Young’s moduli,i.e.,1.527kPa for MDA-MB-231(highly invasive breast cancer cells),2.650kPa for McF-7(weakly invasive breast cancer cells),and 2.772 kPa for SKBR-3(weakly invasive breast cancer cells),indicate that highly invasive cancer cells exhibited a lower Young’s moduli than weakly invasive cells,which indicates a higher deformability of highly invasive cancer cells,leading to a higher metastasis ***-cell treatment may also be carried out by bursting a highly invasive cell with high-intensity,focused ultrasound.
