Expression and reconstitution of the bioluminescent Ca2+reporter aequorin in human embryonic stem cells,and exploration of the presence of functional IP3 and ryanodine receptors during the early stages of their differentiation into cardiomyocytes

In order to develop a novel method of visualizing possible Ca2＋ signaling during the early differentiation of hEsCs into cardi- omyocytes and avoid some of the inherent problems associated with using fluorescent reporters, we expressed the biolumines- cent Ca2＋ reporter, apo-aequorin, in HEs2 cells and then reconstituted active holo-aequorin by incubation withf-coelenterazine. The temporal nature of the Ca2＋ signals generated by the holo-f-aequorin-expressing HEs2 cells during the earliest stages of differentiation into cardiomyocytes was then investigated. Our data show that no endogenous Ca2＋ transients （generated by re- lease from intracellular stores） were detected in 1-12-day-old cardiospheres but transients were generated in cardiospheres following stimulation with KC1 or CaC12, indicating that holo-f-aequorin was functional in these cells. Furthermore, following the addition of exogenous ATP, an inositol trisphosphate receptor （IP3R） agonist, small Ca2＋transients were generated from day 1 onward. That ATP was inducing Ca2＋ release from functional IP3Rs was demonstrated by treatment with 2-APB, a known IP3R antagonist. In contrast, following treatment with caffeine, a ryanodine receptor （RyR） agonist, a minima/Ca2＋ response was observed at day 8 of differentiation only. Thus, our data indicate that unlike RyRs, IP3Rs are present and continually functional at these early stages of cardiomyocyte differentiation.

Preoperative Feeding in single Ventricle Neonates is Predictive of shorter Time to Goal Feed

Urban Park soil and Vegetation: Effects of Natural and Anthropogenic Factors

Various soil surface components, such as trees, shrubs and biological crusts, and human recreational activities, e.g., barbecues and trampling by visitors, may divide the area of the urban park into smaller fragments/mieroenvironments, differentiated by their microenvironmental conditions, which may differ in soil and vegetation characteristics. The spatial changes in the soil and vegetation characteristics and their causes were investigated in an urban park located south of Tel Aviv-Jaffa, Israel. The area of the park is 0.5 km^2, including groves, a lake, lawns, and rest areas. soil was sampled in nine microenvironments, of which seven were within the park： under Ceratonia siliqua trees （CsU）, under Ficus sycomorus trees （FLU）, rest area between tables under F. sycomorus （FIB）, rest area under tables under F. sycomorus （FIT）, open area with bare soil （Oas）, open area with biological crust cover （OaC）, and open area covered by herbaceous vegetation （OaV）. Two more microenvironments, planar and sloping open areas （CoP and Cos, respectively） in the vicinity of the park, were used as the controls. Electrical conductivity, concentrations of soluble ions （Mg^2＋, Ca^2＋, Na^＋, K^＋ and HCO3）, pH, contents of organic carbon, calcium carbonate and moisture, and grain size distribution were determined. In addition, herbaceous vegetation cover, number and diversity of herbaceous vegetation species were measured. It was found that soil properties and herbaceous vegetation characteristics varied within the park. soil organic carbon, electrical conductivity, soluble salts, penetration depth, and vegetation characteristics were affected by human activities, mainly in the rest area between the tables. In contrast, the above characteristics were affected by natural factors mainly in the rest of the microenvironments, which were subjected to low levels of anthropogenic intervention. Do females use their sexual status to gain resource access？ Investigating food-for-sex in wolves and dogs

While food sharing among related individuals can be explained by kin selection, food sharing be- tween unrelated individuals has been more of an evolutionary puzzle. The food-for-sex hypothesis provides an explanation for the occurrence of food sharing among nonkin. However, little is known about the socio-ecological factors that can promote such a commodity exchange. A species mating system is a factor potentially influencing food-for-sex patterns of behavior. Here, we compared wolves, which form pair-bonds, with dogs, which are typically promiscuous in freeranging contexts, to investigate the effect of reproductive stages on the behavior around a food source in 2 different contexts. Furthermore, we considered the roles of both the males and the fe- males in the potential food-for-sex exchange. Results indicate that in both species and for both sexes the breeding period promotes decreased aggression. Additionally, females were more per- sistent in their attempts to access the food and were able to monopolize the resource more when in heat as compared to outside the breeding period. Finally, in dogs, but not wolves, females spent more time in proximity to the male＇s bone and had a shorter latency to start eating it when in heat. Overall, this study demonstrates that the food-for-sex hypothesis plays a part in intersexual food sharing in canids, and highlights the role of females in the interaction. These effects were especially the case in dogs, suggesting a potential effect of mating system on food-for-sex responses

What are college students saying about psychiatric medication?

The number of college students who take psychiatric medication has dramatically increased. These students may be at risk for negative mental health outcomes because research shows that mental illness can delay the attainment of developmental milestones critical to adulthood. This article explores college students’ experience with psychiatric medication and how it impacts functioning and stigma. Perceptions of medication treatment could be crucial to understanding the factors that enable college students with mental illness to thrive in a university setting. seventeen undergraduate college students in a private, Midwestern university who had a psychiatric illness and were taking prescribed psychiatric medication, were enrolled. A semi-structured interview queried college students about their perceptions of taking psychiatric medications and how the use of medication influences their functioning. Authors conducted thematic analysis by using the constant comparative method for coding data and sorting in-vivo codes by shared theme. Respondents generally reported positive attitudes toward medication and minimal stigma. Particular themes included: higher functioning;mitigation of symptoms;willingness to disclose;and positive long-term outlook regarding the use of medication. students were empowered by their treatment because it positively impacted functioning and integration into the college setting. However, in contrast to the majority of study participants, one minority student reported experiencing significant external and internal stigma due to her use of psychiatric medication. Although the study’s qualitative nature, small sample size,and lack of ethnic diversity of respondents limit generalizability, important preliminary findings indicate that some college students are benefiting from the use of psychiatric medication with minimal stigma. Beef heifer fertility: importance of management practices and technological advancements

The development of replacement heifers is at the core of cow-calf beef production systems. In 2020, the UsDA,National Agricultural statistics service reported 5.771 million beef heifers, 500 pounds and over, are under development for cow replacement. A compilation of data from several studies indicate that between 85% and 95% of these heifers will become pregnant in their first breeding season. several thousands of heifers being raised for replacement may not deliver a calf on their first breeding season and result in economic losses to cow-calf producers. Many management procedures have been developed to maximize the reproductive potential of beef heifers. such approaches include, but are not limited to the following: nutritional management for controlled weight gain, identification of reproductive maturity by physiological and morphological indicators, and the implementation of an estrous synchronization *** implementation of management strategies has important positive impact(s) on the reproductive efficiency of *** are limitations, however, because some heifers deemed ready to enter their first breeding season do not become pregnant. In parallel, genetic selection for fertility-related traits in beef heifers have not promoted major genetic gains on this particular area, most likely due to low heritability of female fertility traits in cattle. Technologies such as antral follicle counting, DNA genotyping and RNA profiling are being investigated as a means to aid in the identification of heifers of low fertility potential. To date, many polymorphisms have been associated with heifer fertility, but no DNA markers have been identified across herds. Antral follicle count is an indication of the ovarian reserve and is an indicator of the reproductive health of a heifer. Exploring the impact of COVID-19 on the movement behaviors of children and youth:A scoping review of evidence after the first year

Purpose:The objective of this scoping review was to summarize systematically the available literature investigating the relationships between the coronavirus disease 2019(COVID-19)pandemic and movement behaviors(physical activity,sedentary behavior,and sleep)of school-aged children(aged 511 years)and youth(aged 1217 years)in the first year of the COVID-19 ***:searches for published literature were conducted across 6 databases on 2 separate search dates(November 25,2020,and January 27,2021).Results were screened and extracted by 2 reviewers(DCP and KR)independently,using *** numeric analysis and content analysis were undertaken to present thematically the findings of included studies according to the associated impact on each movement ***:A total of 1486 records were extracted from database searches;of those,150 met inclusion criteria and were included for *** 150 articles,110 were empirical studies examining physical activity(n=77),sedentary behavior/screen time(n=58),and sleep(n=55).Results consistently reported declines in physical-activity time,increases in screen time and total sedentary behavior,shifts to later bed and wake times,and increases in sleep *** reported impacts on movement behaviors were greater for youth than for ***:The COVID-19 pandemic is related to changes in the quantity and nature of physical activity,sedentary behavior,and sleep among children and *** is an urgent need for policy makers,practitioners,and researchers to develop solutions for attenuating adverse changes in physical activity and screen time among children and youth.

Dementia is predominantly a disease of aging with millions of people suffering from dementia and Alzheimer＇s disease （AD） and in recent years little has happened to change and improve the cognitive functions of elderly people （2015 Alzheimer＇s disease facts and figures）. Every 4 seconds there is a new case of dementia in the world and with 7.7 mil- lion cases annually, the estimated global cost of AD and demen- tia disease is $604 billion.
