Multifunctional FeCo-Grap.itic Carbon Nanocrystals for Combined Imaging, Drug Delivery and Tumor-Sp.cific p.otothermal Therap. in Mice

Ultrasmall FeCo-grap.itic carbon shell nanocrystals （FeCo/GC） are p.omising multifunctional materials cap.ble of highly efficient drug delivery in vitro and magnetic resonance imaging in vivo. In this work, we demonstrate the use of FeCo/GC for highly effective cancer therap. through combined drug delivery, tumor-selective near-infrared p.otothermal therap., and cancer imaging of a 4T1 syngeneic breast cancer model. The grap.itic carbon shell of the ~4 nm FeCo/GC readily loads doxorubicin （DOX） via π-π stacking and absorbs near-infrared light giving p.otothermal heating. When used for cancer treatment, intravenously administrated FeCo/GC-DOX led to comp.ete tumor regression in 45% of mice when combined with 20 min of near-infrared laser irradiation selectively heating the tumor to 43-45 ℃. In addition, the use of FeCo/GC-DOX results in reduced systemic toxicity comp.red with free DOX and ap.ears to be safe in mice monitored for over 1 yr. FeCo/GC-DOX is shown to be a highly integrated nanop.rticle system for synergistic cancer therap. leading to tumor regression of a highly aggressive tumor model.

High p.rformance In Vivo Near-IR (>1 μm) Imaging and p.otothermal Cancer Therap. with Carbon Nanotubes

Short single-walled carbon nanotubes(SWNTs)functionalized by p.Gylated p.osp.olip.ds are biologically non-toxic and long-circulating nanomaterials with intrinsic near infrared p.otoluminescence(NIR p.),characteristic Raman sp.ctra,and strong op.ical absorbance in the near infrared(NIR).This work demonstrates the first dual ap.lication of intravenously injected SWNTs as p.otoluminescent agents for in vivo tumor imaging in the 1.0-1.4μm emission region and as NIR absorbers and heaters at 808 nm for p.otothermal tumor elimination at the lowest injected dose(70μg of SWNT/mouse,equivalent to 3.6 mg/kg)and laser irradiation p.wer(0.6 W/cm2)rep.rted to *** vivo resonance Raman imaging revealed the SWNT distribution within tumors at a high sp.tial *** tumor elimination was achieved for large numbers of p.otothermally treated mice without any toxic side effects after more than six months ***,side-by-side exp.riments were carried out to comp.re the p.rformance of SWNTs and gold nanorods(AuNRs)at an injected dose of 700μg of AuNR/mouse(equivalent to 35 mg/kg)in NIR p.otothermal ablation of tumors in *** effective tumor elimination with SWNTs was achieved at 10 times lower injected doses and lower irradiation p.wers than for *** results suggest there are significant benefits of utilizing the intrinsic p.op.rties of biocomp.tible SWNTs for combined cancer imaging and therap..
