despite a significant increase in utilization over the past decade,the number of donation after circulatory death(DCD)organs that are procured and transplanted in the United States(US)remains well below its *** is still room for expansion,as utilizing DCD organs to the fullest extent is currently the most viable solution to the persistent mismatch between supply and demand in *** convened a multidisciplinary transplantation summit to examine various aspects of DCD,with faculty members from around the world with clinical and academic interest in DCD donation and transplantation,including abdominal and cardiothoracic surgeons,organ procurement organization directors,hepatologists,and *** conference focused on identifying barriers to DCD organ utilization and strategies to overcome these *** divide the barriers to DCD utilization into three mains categories:(Ⅰ)policy and process variation;(II)logistical and transportation challenges;and(Ⅲ)higher risk perceptions related to DCD *** each barrier,we proposed a variety of solutions,providing an overview of the status of DCD donation in the US and suggestions on how to increase the use of *** is a specific focus on ex situ machine perfusion,normothermic regional perfusion,and other opportunities to expand DCD utilization without negatively impacting recipient outcomes.

Reviewing Self-Injuries Behavior in Macaques: The Role of Early Experience and Genetics in an Integrated Non-Human Model

Genetic differences in non-human primates and the context where they were reared are important in the determination of their behaviors. Some studies suggest that Self-Injurious Behavior results from stress or anxiety, but some macaques appear to be more vulnerable to acquiring Self-Injurious Behavior. Vulnerability and risk for socially reared macaques of developing Self-Injurious Behavior increased when they experienced early adverse events and suffered from consequent stress. Stressful events can cause alterations in the neuroendocrine and neuropeptide systems associated with the regulation of stress and anxiety. Dysregulation in these systems contributes to the occurrence of anxious episodes that lead to self-harm. This could be used by those animals to counteract the effects of anxiety, causing euphoria by releasing endogenous opioids. In developing an integrative approach of Self-Injurious Behavior, we propose the diathesis-stress model to illustrate how the environment and genetics are influenced as well, taking rhesus monkeys as an example. Rhesus macaques and humans have essentially the same gene 5-HTT as well as functional polymorphism. The allelic variation in the region promoting the serotonin transporter gene (5-HTT) was associated with significant differences in the number of depressive symptoms and contributions to social reward and punishment. Growing up without the mother and the rearing environment are important risk factors for developing abnormal behaviors. This work focuses on a general class of nonlinear process systems controlled by a two-tier networked control system integrating a local control system using continuous sensing/actuation with a networked control system using asynchronous sensing/actuation. To deal with control actuator faults that may occur in the closed-loop system and eliminate the ability of the two-tier networked control system to stabilize the process, a networked fault detection and isolation（fDI）and fault-tolerant control（fTC） system is designed which detects and isolates actuator faults and determines how to reconfigure the two-tier networked control system to handle the actuator faults and ensure closed-loop stability. The method is demonstrated using a reactor-separator process example.

Multitoxin Bt-crops expressing insecticidal toxins with different modes of action, for example, Cry and Vip, are expected to improve resistance management in target pests. While Cry1A resistance has been relatively well characterized in some insect species, this is not the case for Vip3A, for which no mechanism of resistance has yet been identified. Here we applied HT-SuperSAGE to analyze the transcriptome of the gut tissue of tobacco budworm Heliothis virescens (f.) laboratory-selected for Vip3Aa resistance. from a total of 1 324 252 sequence reads, 5 895 126-bp tags were obtained representing 17 751 nonsingleton unique transcripts (UniTags) from genetically similar Vip3Aa-resistant (Vip- Sel) and susceptible control (Vip-Unsel) strains. Differential expression was significant (≥2.5 fold or ≤0.4;P for 1989 sequences (11.2% of total UniTags), where 420 represented overexpressed (OE) and 1569 underexpressed (UE) genes in Vip-Sel. BlaSTN searches mapped 419 UniTags to H. virescens sequence contigs, of which, 416 (106 OE and 310 UE) were unambiguously annotated to proteins in NCBI nonredundant protein databases. Gene Ontology distributed 345 of annotated UniTags in 14 functional categories with metabolism (including serine-type hydrolases) and translation/ribosome biogenesis being the most prevalent. A UniTag homologous to a particular member of the REsponse to PAThogen (REPAT) family was found among most overexpressed, while UniTags related to the putative Vip3Aa-binding ribosomal protein S2 (RpS2) were underexpressed. qRT-PCR of a subset of UniTags validated the HT-SuperSAGE data. This study is the first providing lepidopteran gut transcriptome associated with Vip3Aa resistance and a foundation for future attempts to elucidate the resistance mechanism.

Immunohistochemical evaluation of vitamin D receptor(VDR) expression in cutaneous melanoma tissues and four VDR gene polymorphisms

The system proposed in this paper consists of humidity and temperature control during the food maturation process by using a dehumidifier equipped with a desiccant rotor containing silica *** this system the condensation/drainage stage is omitted since the humid room air is directed out of the cold store(process air)and the dried air is introduced via the dehumidifier inside the cold *** allows separate control of the humidity and temperature inside the cold *** experimental results are obtained in real conditions;the weight loss of the products was determined and it was,also,proved that dried products such as cheeses treated by this method retain their fresh color,texture and other organoleptic properties to ensure their final quality.

A gain-of-function mutation in the fibroblast growth factor receptor 3 gene(fGfR3)results in achondroplasia(ACH),the most frequent form of *** activation of fGfR3 impairs bone formation and elongation and many signal transduction *** of new and relevant compounds targeting the fGfR3 signaling pathway is of broad importance for the treatment of ACH,and natural plant compounds are prime drug candidate ***,we found that the phenolic compound(-)-epicatechin,isolated from Theobroma cacao,effectively inhibited fGfR3’s downstream signaling *** analysis in an fgfr3 mouse model showed that ciliary mRNA expression was modified and influenced significantly by the Indian hedgehog and PKA pathways.(-)-Epicatechin is able to rescue mRNA expression impairments that control both the structural organization of the primary cilium and ciliogenesis-related *** femurs isolated from a mouse model(fgfr3^(Y367C/+))of ACH,we showed that(-)-epicatechin eliminated bone growth impairment during 6 days of ex vivo *** vivo,we confirmed that daily subcutaneous injections of(-)-epicatechin to fgfr3^(Y367C/+) mice increased bone elongation and rescued the primary cilium defects observed in *** modification to the primary cilia promoted the typical columnar arrangement of flat proliferative chondrocytes and thus enhanced bone *** results of the present proof-of-principle study support(-)-epicatechin as a potential drug for the treatment of ACH.

【目的】苜蓿皂苷作为一类天然生物活性物质,具有提高畜禽机体免疫力、促生长及调节胃肠道功能等多方面效用。试验旨在探究苜蓿皂苷提取物对断奶湖羊生长性能、免疫功能、抗氧化能力和瘤胃微生物区系的影响。【方法】选取45日龄、体重相近(15.67 kg±1.18 kg)的断奶湖羊32只,随机分为2组,每组4个重复,每个重复4只羊。对照组(CON)湖羊饲喂基础饲粮,皂苷组(AS)湖羊在基础饲粮中添加2 g/kg(饲粮干物质基础)苜蓿皂苷提取物,预试期7 d,正试期60 d。试验结束后收集断奶湖羊瘤胃液测定微生物组成,采集颈静脉血用于测定血清免疫和抗氧化性能相关指标。【结果】(1)两组湖羊平均日增重(ADG)和料重比(f/G)均无显著差异(P>0.05)。(2)与对照组相比,皂苷组湖羊血清中免疫球蛋白(IgA、IgM)、抗炎因子白介素-10(IL-10)、CD4^(+)T细胞和CD4/CD8水平极显著或显著升高(Pf-α)、IL-6含量均极显著降低(PfA)浓度均无显著差异(P>0.05)。(5)两组间湖羊Shannon指数、Simpson指数和主坐标分析(PCoA)均无明显差异;在属水平上,皂苷组湖羊瘤胃液中硫酸盐还原菌脱硫弧菌属(desulfovibrio)和有益菌罗氏菌属(Roseburia)、凸腹真杆菌群(Eubacterium_ventriosum_group)等的相对丰度均显著高于对照组(P<0.05)。【结论】饲粮中添加苜蓿皂苷提取物可以影响断奶湖羊瘤胃微生物的组成,提高机体免疫和抗氧化能力,且一定程度上有利于机体健康。
