Pyrogen,often as a contaminant,is a key indicator affecting the safety of almost all parenteral drugs(including biologicals,chemicals,traditional Chinese medicines and medical devices).It has become a goal to completely replace the in vivo rabbit pyrogen test by using the in vitro pyrogen test based on the promoted‘reduction,replacement and refinement’principle,which has been highly considered by regulatory agencies from different *** used NF-κB,a central signalling molecule mediating inflammatory responses,as a pyrogenic marker and the monocyte line THP-1 transfected with a luciferase reporter gene regulated by NF-κB as an in vitro model to detect pyrogens by measuring the intensity of a fluorescence ***,we show that this test can quantitatively and sensitively detect endotoxin(lipopolysaccharide from different strains)and nonendotoxin(lipoteichoic acid,zymosan,peptidoglycan,lectin and glucan),has good stability in terms of NF-κB activity and cell phenotypes at 39 cell passages and can be applied to detect pyrogens in biologicals(group A&C meningococcal polysaccharide vaccine;basiliximab;rabies vaccine(Vero cells)for human use,freeze-dried;Japanese encephalitis vaccine(Vero cells),inactivated;insulin aspart injection;human albumin;recombinant human erythropoietin injection(CHO Cell)).The within-laboratory reproducibility of the test in three independent laboratories was 85%,80%and 80%and the interlaboratory reproducibility among laboratories was 83.3%,95.6%and 86.7%.The sensitivity(true positive rate)and specificity(true negative rate)of the test were 89.9%and 90.9%,*** summary,the test provides a novel alternative for pyrogen detection.

针对沸腾床渣油加氢裂化工艺存在温度和压力交变的特点，设计了一种加氢裂化装置高温高压耐磨球阀。首先阐述其整体设计理论，充分考虑该球阀的密封、启闭性能及制造工艺的要求，与常规的大口径硬密封球阀整体结构进行了对比；其次基于该球阀的结构特点，将其中腔密封结构和阀座密封结构与常规硬密封球阀密封结构进行了对比及原理分析，并对该球阀的重要零部件进行设计计算与校核，证明了阀门结构设计的合理性；最后采用有限元法对整体温度场及密封副受力进行了分析，确定支架的温度，以确保该球阀执行机构的正常运行。通过对该球阀的研究，其性能具有全通径、流量大、流阻小、结构简单、密封可靠、不易卡塞、开关灵活和维修方便等优点，为促进加氢裂化装置高效、经济、长期运行提供保障。

The Chinese tree shrew(Tupaia belangeri chinensis),a member of the mammalian order Scandentia,exhibits considerable similarities with primates,including humans,in aspects of its nervous,immune,and metabolic *** similarities have established the tree shrew as a promising experimental model for biomedical research on cancer,infectious diseases,metabolic disorders,and mental health ***,we used metatranscriptomic sequencing to analyze plasma,as well as oral and anal swab samples,from 105 healthy asymptomatic tree shrews to identify the presence of potential zoonotic *** total,eight mammalian viruses with complete genomes were identified,belonging to six viral families,including Flaviviridae,Hepeviridae,Parvovirinae,Picornaviridae,Sedoreoviridae,and ***,the presence of rotavirus was recorded in tree shrews for the first *** viruses-hepacivirus 1,parvovirus,and picornavirus-exhibited low genetic similarity(94%)to known viral *** analyses also revealed that the rotavirus and mammalian orthoreovirus identified in this study may be novel *** findings provide insights into the diverse viral spectrum present in captive Chinese tree shrews,highlighting the necessity for further research into their potential for crossspecies transmission.

目的探讨动力相关蛋白1(Drp-1)、过氧化物酶体增殖物激活受体γ共激活因子1-α(PGC-1α)在胎粪吸入综合征(MAS)新生大鼠肺组织中的作用及其机制。方法50只2~3周龄SD新生大鼠随机分为5组(n=10):对照组、模型组与SN50低、中、高浓度组。对照组气管暴露后,气管内注入2ml/kg生理盐水,其余各组气管内注入2ml/kg的胎粪悬浮液;24h后对照组、模型组不予处理,SN50低、中、高浓度组腹腔注射10、30、60μg/ml浓度SN50各100μl。6h后处死各组大鼠,检测X线胸片、肺大体观、肺湿/干重比(W/D),HE染色观察肺组织病理变化,Westernblotting检测新生大鼠肺组织中NF-κB(p65)、p-NF-κBp65(p-p65)、Drp-1、PGC-1α蛋白的表达变化,免疫组织化学方法观察新生大鼠肺组织中p65、Drp-1、PGC-1α等相关蛋白表达。结果与对照组比较,模型组胸片及大体观可见炎性浸润,W/D、肺损伤病理评分明显升高(P0.05),而SN50中、高浓度组PGC-1α蛋白表达水平明显升高(P0.05)。免疫组化检测结果显示,与对照组比较,模型组p65、Drp-1蛋白表达水平明显升高(P0.05),而SN50中、高浓度组Drp-1蛋白表达水平明显降低(P<0.05),PGC-1α蛋白表达水平明显升高(P<0.05)。结论胎粪吸入可诱导新生大鼠肺组织炎症,其机制可能与增强氧化应激、促进线粒体功能障碍、激活Drp-1/NF-κB信号通路,以及抑制PGC-1α蛋白表达有关。

在当今人工智能投资快速增长的大背景下,现有研究对全球性长期产能过剩问题的关注不足,缺乏政治经济学的分析视角。本文将马克思两大部类分析视角拓展到三大部类,采用非竞争型投入产出表估算了世界主要国家三大部类的价值构成和一般利润率,并借鉴新卡莱茨基模型测算2000一2019年各国三大部类的产能利用率,选用PVAR模型分析了人工智能及一般利润率对三大部类产能利用率的影响。研究发现:发展中国家的三大部类产能利用率均值都高于发达国家,且两者都呈现明显的下降趋势,2008年金融危机带来的“创造性破坏”并未扭转全球范围内的长期产能过剩趋势。一般利润率变动会对产能利用率变动产生负向影响,且其作用力度在发展中国家更大;同时,人工智能增长对一般利润率变动的正向作用也是在发展中国家比发达国家更为显著。人工智能的增长在发达国家倾向于拓展潜在产出而相对拉低其产能利用率,而在发展中国家则短期内倾向于提升实际产出而相对提高其产能利用率。本文的研究结果有助于认清发达国家与发展中国家之间的产能过剩程度,推进全球范围内的长期产能过剩治理,为中国推动高新技术生产资料部类发展提供实践指导。
