AIM: to examine the effects of Kampo medicine, Keishi-ka-Shakuyaku-to (tJ-60) on the signal transduction in diabetic gastrointestinal dysfunction. MEtHODS: Experimental diabetic models were prepared using streptozotocin (StZ)-treated Wistar rats. Randomly selected StZ rats were treated with insulin (12 U·kg-1·d-1) or tJ-60 (1% of food intake). Diacylglycerol (DG) and DG kinase activities were quantified in isolated aortic smooth muscle tissue. RESULtS: One of the key element of the Pi-turnover, DG kinase activity in resting state in gastric smooth muscle was significantly increased compared to the control value, and carbachol (CCh)-induced response was not detectable, but it was detected in the control rats. On the other hand resting activity in ileum did not differ from the control, but the CCh-induced responses were suppressed. treatment with tJ-60 indicated resistant effects for the alteration of DG kinase activities in diabetic intestinal tissues. Nitric oxide （NO） is increasingly being used in medical treatments of high blood pressure, acute respiratory distress syndrome and other illnesses related to the lungs. Currently a NO inhalation system consists of a gas cylinder of N2 mixed with a high concentration of NO. this arrangement is potentially risky due to the possibility of an accidental leak of NO from the cylinder. the presence of NO in the air leads to the formation of nitric dioxide （NO2）, which is toxic to the lungs. therefore, an on-site generator of NO would be highly desirable for medical doctors to use with patients with lung disease. to develop the NO inhalation system without a gas cylinder, which would include a high concentration of NO, NAMIHIRA et al have recently reported on the production of NO from room air using a pulsed arc discharge. In the present work, the temperature of the pulsed arc discharge plasma used to generate NO was measured to optimize the discharge condition. the results of the temperature measurements showed the temperature of the pulsed arc discharge plasma reached about 10,000 K immediately after discharge initiation and gradually decreased over tens of microseconds. In addition, it was found that NO was formed in a discharge plasma having temperatures higher than 9,000 K and a smaller input energy into the discharge plasma generates NO more efficiently than a larger one.

近年来,汽车发动机都要求较高使用性能来说,阀座的使用环境日益严酷。一般而言,设计的阀座材料的耐磨性,都是用实际发动机耐久试验评定的,试验时间都需要许多小时。在这项研究中,为柴油发动机排气阀座选择的材料制定了一个评定方法。这种评定方法是根据模拟试验和发动机耐久试验间的关系研制出的。通过对硬度与磨损间关系的多变量分析,开发出了一种耐磨性优异的、成本低廉的阀座材料。

In the outer periclinal cytoplasm of leaf epidermal cells of an aquatic angiosperm Vallisneria, blue light induces ＂chloroplast de-anchoring＂, a rapid decline in the resistance of chloroplasts against centrifugal force. Chloroplast de- anchoring is known induced within lmin of irradiation with high-fluence-rate blue light specifically, preceding the com- mencement of chloroplasts migration toward the anticlinal cytoplasm. However, its regulatory mechanism has remained elusive, although pharmacological analysis suggested that a calcium release from intracellular calcium stores is necessary for the response, in search of the responsible photoreceptors, immunoblotting analysis using antibodies against phototropins demonstrated that cross-reactive polypeptides of 120-kDa exist in the plasma-membrane fraction prepared from the leaves. In vitro phosphorylation analysis revealed that 120-kDa polypeptides were phosphorylated by exposure to blue light in a fluence-dependent manner. the blue-light-induced phosphorylation activity was sensitive to a Ser/thr kinase inhibitor, staurosporine, and unusually was retained at a high level for a long time in darkness. Furthermore, phototropin gene homologs （Vallisneria PHOtOtROPINI and PHOtOtROPIN2） expressed in leaves were isolated. We propose that calcium- regulated chloroplast de-anchoring, possibly mediated by phototropins, is an initial process of the blue-light-induced avoidance response of chloroplasts in Vallisneria.

BACKGROUND Secondary sclerosing cholangitis,characterized by biliary obstruction,can be caused by drugs such as immune checkpoint inhibitors(ICIs).While there a few reports of sclerosing cholangitis after immune checkpoint inhibitor administration,no case has been reported after discontinuation of such *** SUMMARY A 68-year-old man who underwent chemotherapy for lung adenocarcinoma with bone metastasis presented with abdominal pain and fever 4 mo after the final administration of *** tomography revealed thickening of the gallbladder wall and dilatation of the common bile *** retrograde cholangiopancreatography revealed an irregularly narrowed intrahepatic bile *** of the bile duct demonstrated that CD8+t cells were predominant over CD4+t *** biopsy showed dominant infiltration of CD8+t in the portal tract,but onion-skin lesions were not *** patient was diagnosed with immune-related sclerosing cholangitis induced by *** of methylprednisolone and endoscopic nasobiliary drainage were performed,but the cholangiography and laboratory test findings did not *** further treatment was administered due to disease progression,and the patient was referred for palliative *** Immune-related sclerosing cholangitis may have a late onset,and such cases occurring after discontinuation of ICIs should be carefully managed.

AIM: to investigate the mechanisms of liver growth and atrophy after portal vein ligation (PVL) and its effects on tumor growth. MEtHODS: Mice were subjected to PVL, partial hepatectomy, or sham surgery. the morphological alterations, activation of transcription factors, and expression of cytokines and growth factors involved in liver regeneration were evaluated. In a separate set of experiments, murine colorectal carcinoma cells were injected via the portal vein and the effect of each operation on liver tumor growth was studied. RESULtS: Liver regeneration after PVL and partial hepatectomy were very similar. In ligated lobes, various cytokines, transcription factors and regulatory factors were signifi cantly upregulated compared to nonligated lobes after PVL. Atrophy in ligated lobes was a result of early necrosis followed by later apoptosis. tumor growth was signifi cantly accelerated in ligated compared to non-ligated ***: tumor growth was accelerated in ligated liver lobes and appeared to be a result of increased growth factor expression.

AIM: to analyze the effect of bipolar electrocoagula-tion and argon plasma coagulation on fresh specimens of gastrointestinal tract. MEtHODS: An experimental evaluation was performed at Hospital das Clinicas of the University of So Paulo, on 31 fresh surgical specimens using argon plasma coagulation and bipolar electrocoagulation at different time intervals. the depth of tissue damage was his-topathologically analyzed by single senior pathologist unaware of the coagulation method and power setting applied. to analyze the results, the mucosa was divided in superficial mucosa (epithelial layer of the esophagus and superficial portion of the glandular layer of the stomach and colon) intermediate mucosa (until thelamina propria of the esophagus and until the bottom of the glandular layer of the stomach and colon) and muscularis mucosa. Necrosis involvement of the layers was compared in several combinations of power and time interval. RESULtS: Involvement of the intermediate mucosa of the stomach and of the muscularis mucosa of the three organs was more frequent when higher amounts of en-ergy were used with argon plasma. In the esophagus and in the colon, injury of the intermediate mucosa was frequent, even when small amounts of energy were used. the use of bipolar electrocoagulation resulted in more frequent involvement of the intermediate mucosa and of the muscularis mucosa of the esophagus and of the colon when higher amounts of energy were used. In the stomach, these involvements were rare. the risk of injury of the muscularis propria was significant only in the colon when argon plasma coagulation was ***: tissue damage after argon plasma coagulation is deeper than bipolar electrocoagulation. Both of them depend on the amount of energy used.

Kidney GAtA3^(+) regulatory t cells play roles in the convalescence stage after antibody-mediated renal injury

In this paper, Radio Frequency IDentification （RFID） tag set cardinality estimation problem is considered under the model of multiple independent reader sessions with unreliable radio communication links in which transmission errors might occur. After the R-th reader session, the number of tags detected in j （j = 1, 2, ..., R） reader sessions is updated, which we call observed evidence. then, in order to maximize the likelihood function of the number of tags and the transmission error probability given the observed evidences, we propose three different estimation methods depending on how to treat the discrete nature of tag set cardinality. Computer simulation results demonstrate the effectiveness of the proposed method.

FoxP3^(+)regulatory t cells(tregs)play crucial roles in peripheral immune *** addition,tregs that reside or accumulate in nonlymphoid tissues,called tissue tregs,exhibit tissue-specific functions and contribute to the maintenance of tissue homeostasis and *** an experimental mouse model of crescentic glomerulonephritis induced by an anti-glomerular basement membrane antibody,tregs started to accumulate in the kidney on day 10 of disease onset and remained at high levels(~30–35%of CD4^(+)t cells)during the late stage(days 21–90),which correlated with stable disease *** depletion on day 21 resulted in the relapse of renal dysfunction and an increase in th1 cells,suggesting that tregs are essential for disease control during the convalescence *** tregs that accumulated in the kidney showed tissue treg phenotypes,including high expression of GAtA3,St2(the IL33 receptor subunit),amphiregulin(Areg),and PPARγ.Although t-bet^(+) tregs and RORγt^(+) tregs were observed in the kidney,GAtA3^(+) tregs were predominant during the convalescence stage,and a PPARγagonist enhanced the accumulation of GAtA3^(+) tregs in the *** understand the function of specific genes in kidney tregs,we developed a novel t cell transfer system to t cell-deficient *** experiment demonstrates that St2,Areg,and CCR4 in tregs play important roles in the accumulation of GAtA3^(+) tregs in the kidney and in the amelioration of renal *** data suggest that GAtA3 is important for the recruitment of tregs into the kidney,which is necessary for convalescence after renal tissue destruction.
