Transcriptional regulation of flavonoid biosynthesis in Artemisia annua by AaYABBY5

Artemisia annua is a medicinal plant rich in terpenes and flavonoids with useful biological activities such as antioxidant,anticancer,and antimalarial *** transcriptional regulation of flavonoid biosynthesis in *** has not been *** this study,we identified a YABBY family transcription factor,AaYABBY5,as a positive regulator of anthocyanin and total flavonoid contents in ***5 was selected based on its similar expression pattern to the phenylalanine ammonia lyase(PAL),chalcone synthase(CHS),chalcone isomerase(CHI),and flavonol synthase(FLS)genes.A transient dual-luciferase assay in Nicotiana bethamiana with the AaYABBY5 effector showed a significant increase in the activity of the downstream LUC gene,with reporters AaPAL,AaCHS,AaCHI,and *** yeast one-hybrid system further confirmed the direct activation of these promoters by *** expression analysis of stably transformed AaYABBY5 overexpression,AaYABBY5 antisense,and control plants revealed a significant increase in the expression of AaPAL,AaCHS,AaCHI,AaFLS,AaFSII,AaLDOX,and AaUFGT in AaYABBY5 overexpression ***,their total flavonoid content and anthocyanin content were also found to ***5 antisense plants showed a significant decrease in the expression of flavonoid biosynthetic genes,as well as a decrease in anthocyanin and total flavonoid *** addition,phenotypic analysis revealed deep purple-pigmented stems,an increase in the leaf lamina size,and higher trichome densities in AaYABBY5 overexpression ***,these data proved that AaYABBY5 is a positive regulator of flavonoid biosynthesis in *** study provides candidate transcription factors for the improvement of flavonoid concentrations in *** and can be further extended to elucidate its mechanism of regulating trichome development.

Plantlets Regeneration via Somatic Embryogenesis from the Nucellus Tissues of Kinnow Mandarin (Citrus reticulata L.)

Studies were initiated to explore the role of nucellus tissues and growth regulators in plantlets regeneration via somatic embryogenesis of Kinnow mandarin [Citrus reticulata L. (Blanco)]. Nucellus tissues were cultured on MS medium supplemented with different concentrations and combinations of auxins, cytokinins and malt extract for primary callus induction. The best response for primary callus induction (90%) was obtained when MS medium was supplemented with 5 mg/l 2,4-D and 500 mg/l malt extract. Best results for embryogenic callus induction (80%) were obtained in C8 medium. The induction of somatic embryos was highest when MS medium was supplemented with 1 mg/l BAP and maturation of somatic embryos occurred when MS medium was supplemented with 5 mg/l 2,4-D and 1 mg/l BAP. Maximum plantlets were regenerated (92%) from the somatic embryos on half strength MS medium with no hormones. The plantlets were successfully acclimatized in different potting mixtures and highest survival rate (100%) was achieved in potting mixture containing sand and peat moss (2:1).

Plants from the start are being used for the welfare of human and animals. About 25,000 biological active compounds are reported by different scientists. Plants itself are a complete treatment bioagent. People are still using plants and their decoction for different diseases. Saussurea lappa Clarke is the member of family Compositae. This plant is famous due to its high medical importance. The plant is commonly named as Kuth root or costus and has wide use for anticancer, antiulcer, hepatoprotective, anti-viral, anticonvulsant, antiarthritic, activities. Biologically active substance of in this plant is lactone cynaropicrin, dehydrocostus, germacrene,?lappadilactone. This plant can be used to extract such bioactive compounds which can help the scientist to discover new and potential drugs. Due to such chemical composition and medicinal importance this review has been prepared for the awareness of the people to conserve their medicinal plants which can be used for potential drug discovery.

Objective: Evaluating the bio-reducing activity and various potential, annotates the biological entitties of Fagonia cretica. Method: By fractionating the crude extract with different ethanol-water fraction mixture. This substantiates the reduction potential for the biological synthesis of AgNPs, its antioxidant, anti-urease and anti-Tyrosinase activity. Results: UV-visible spectra confirm the bio-reducing potential for Ag NPs with the optimized 50% ethanol-water fraction mixture. Transmission electron microscopy (TEM) confirmed the round shaped AgNPs with an average size of 16 nm in optimized 50% fraction mixture. Further evaluating Ethanol-water fraction with 90% and 70% of plant extract showed more potentials for Anti-Tyrosinase and Anti-urease activity. Moreover, diluted 30% fraction exhibits two folds higher Anti-oxidant and Anti urease activity. Conclusion: Overall, this work showed that 50% Ethanol-water fraction has potentially active molecules (among all fractions) primarily involved in bio-reducing potential of Ag NPs and enzymatic assay while 90% mixture proved to be least active. This study suggested electing the optimum mixture fraction of 50% ethanol-water in order to screen the bioactive compounds and to inculcate their activity status for clinical trials.

Thyroglossal Duct Anomalies in Pediatric and Adult Population: A 10-Year Experience in a Tertiary Care Hospital

Introduction: Thyroglossal duct cyst is a well-recognized congenital midline neck swelling observed in early childhood and rarely in adults. It may reveal itself as a painless cyst, abscess, or as fistula. Several studies have been conducted across the world, but the literature is scarce on its presentation and complications in the Middle East. This indexing study aims to report a 10-year experience with thyroglossal duct cyst (TGDC) presentation, excision, and recurrence at a teaching hospital in Saudi Arabia (KSA). Methods: A retrospective chart review was conducted at the Armed Forces Hospital Southern Region, KSA from December 2008 to December 2018. Data were retrieved from the electronic medical record system of the hospital and validated with the histopathology records. A total of 48 patients diagnosed as TGDC, sinus or fistula were identified. Stata ver. 16.1 was used to analyze the data and results formulated using regression model and Pearson’s chi-square test. Results: The majority of our patients were female (60.4%), and the most common presentation was a midline neck swelling which moved with tongue protrusion. Surgical excision via Complete Sistrunk procedure was the operation of choice in 60.5% followed by excision of the cyst with tract and sinus tract. Previous infection and surgical history contributed to complications in our patients, but no association of age or gender was observed. Patients presenting with infected and discharging cysts were found to be much more likely to develop a recurrence (9 patients). Cysts deep to the hyoid showed more recurrence [n = 7 (78%)] compared to superficial cysts [n = 2 (22%)] and posterior and multi-tract lesions also showed significant recurrence. Conclusion: Despite being a common cause of neck swelling, thyroglossal duct cyst is often missed on physical exams. To reduce complications, timely diagnosis and treatment are necessary.

荧光检测是医学诊断和生物技术中广为应用的分析测试技术,但是对于痕量分析或弱荧光分子体系,荧光检测技术仍然存在一定的局限。近年来,表面增强荧光技术逐渐兴起。当荧光分子分布于金或银等金属纳米结构或其溶胶附近时,其荧光强度较之自由态荧光强度显著增加。这为进一步提高荧光检测的检测限和灵敏度提供了新的可能,从而使表面增强荧光技术有潜力成为继表面增强拉曼技术之后,应用于表面科学研究、超灵敏生物检测的又一个有力工具。本文提出了一种使用Ag@SiO核壳纳米粒子对固体基底上自组装的染料单层的荧光进行增强的新方法。制备了壳厚分别为3纳米和15纳米的两种Ag@SiO核壳纳米粒子作为增强剂。把薄壳增强剂滴加到自组装了孟加拉玫瑰红染料单层的基底上,自然风干后,得到了最高约27倍的荧光增强。而当使用厚壳增强剂时,得到了约11.7倍的荧光增强。系统研究了增强剂的壳厚以及表面密度对增强效果的影响。实验结果表明本方法对基于荧光的表面分析具有潜在的应用价值。

Should dopamine be the first line inotrope in the treatment of neonatal hypotension? Review of the evidence

AIM: To determine if dopamine is effective in treating neonatal hypotension and safe to use comparing to other inotropes. METHODS: This is a review of evidence on inotropic treatment of neonatal hypotension. Databases searched were MEDLINE and the Cochrane Library, a total of 134 studies were identified. Only studies with high quality evidence(level 1a and b and 2a) were included. After review, only eight studies were included in the final analysis. Pooled risk ratios derived for each outcome [Mantel-Haenzel(M-H) fixed effect] with CI, as reported in the Cochrane reviews were plotted in forest plot form. RESULTS: Eight articles met inclusion criteria, which all included treatment in preterm infants. Dopamine increased mean arterial blood pressure(BP)(n = 163; r = 0.88, 95%CI: 0.76 to 0.94) and systolic BP(n = 142; r = 0.81, 95%CI: 0.42 to 0.94) comparing to placebo. Dopamine has been shown overall to be statistically more effective in increasing BP than dobutamine(n = 251, r = 0.26, 95%CI: 0.20-0.32). However there were no differences in short term outcomes(periventricular leucomalacia, periventricular haemorrhage) and mortality between both drugs. There is no statistical evidence of dopamine being more effective than adrenaline or corticosteroids. There was no difference in morbidity and mortality outcomes when dopamine was compared to hydrocortisone(RR 1.81, 95%CI: 0.18 to 18.39) or adrenaline. CONCLUSION: In preterms, dopamine is the most studied drug, and we suggest it could be used as first line treatment in hypotension.

为响应绿色、环保发展的需求,无氟连铸保护渣成为了国内外研究热点,其中,以硼硅酸钙(CaSiBO)为主要析晶相的含BO无氟保护渣具有较好的应用前景。本文结合分子动力学模拟、旋转黏度计测试、红外光谱实验以及Fact Sage热力学计算,研究BO对无氟保护渣的微观结构和黏度特性的影响机制。结果表明,CaO-SiO-BO三元渣系形成了稳定的[SiO]–四面体、[BO]–三面体和[BO]–四面体结构单元,BO使Si-O网络结构一定程度上聚合,而对B-O网络结构聚合度的影响因其含量而异,红外光谱所得实验结果与分子动力学模拟结果很吻合;w(BO)=4%～12%范围内增加,无氟保护渣1300℃黏度和液相线温度降低,而熔渣网络结构聚合度基本不变,表明BO使无氟保护渣1300℃黏度降低的主要原因是熔渣过热度的增加;由于析晶矿相和固溶体比例的变化,w(BO)=4%～6%范围内无氟保护渣黏度-温度曲线呈现碱性渣特征,w(BO)=8%～12%范围内无氟保护渣黏度-温度曲线呈现出酸性渣特征。
