Arsenic Disulfide Induced Apoptosis and Concurrently Promoted Erythroid Differentiation in Cytokine-Dependent Myelodysplastic Syndrome-progressed Leukemia Cell Line F-36p with Complex Karyotype Including Monosomy 7

Objective： Acute myeloid leukemia progressed from myelodysplastic syndrome （MDS/AML） is generally incurable with poor prognosis for complex karyotype including monosomy 7 （-7）. Qinghuang Powder （青黄, QHP）, which includes Qing Dai （ Indigo naturalis） and Xiong Huang （realgar） in the formula, is effective in treating MDS or MDS/AML even with the unfavorable karyotype, and its therapeutic efficacy could be enhanced by increasing the Xiong huang content in the formula, while Xiong huang contains 〉 90% arsenic disulfide （As2S2）. F-36p cell line was established from a MDS/AML patient with complex karyotype including -7, and was in cytokine-dependent. The present study was to investigate the effects of As2S2 on F-36p cells. Methods： Cell proliferation was measured by an 3-（4,5-dimethylthiazol-2-yl）-2, 5-diphenyltetrazolium bromide （MTT） assay. Cell apoptosis was identified by Annexin V-staining. Cell viability was determined by a propidium iodide （PI） exclusion. Erythroid differentiation was evaluated by the expression of cell surface antigen CD235a （GpA）. Results： After treatment with As2S2 at concentrations of 0.5 to 16 p. mol/L for 72 h, As2S2 inhibited the proliferation of F-36p cells. The 50% inhibitory concentrations （IC50） of As2S2 against the proliferation of F-36p cells was 6 I~ mol/L. The apoptotic cells significantly increased in a dose-dependent mannar （P〈0.05）. The cell viabilities were significantly inhibited by As2S2 dose-dependent in a dose-dependent manner （P〈0.05）. Significant increases of CD235a-positive cells were concurrently observed （P〈0.05） also in a dose- dependent manner. Conclusions： As2S2 could inhibit proliferation and viability, induce apoptosis, and concurrently promote erythroid differentiation dose-dependently in F-36p cells. Bacterial ArtA protein specifically binds to the internal region of IS1 <i>in vitro</i>

The internal region of bacterial translocatable IS1 acts as a cis-element to stimulate transcription from the various promoters located upstream. The product of the artA gene is genetically shown to stimulate transcription with the cis-element. Here, a codon-optimized artA gene was synthesized and cloned to express the ArtA protein. ArtA was purified as the Histagged protein. Nitrocellulose filter binding assay showed that ArtA specifically binds to the IS1 internal region. Electrophoretic mobility shift assay also showed specific binding of ArtA to the IS1 internal region. These results imply that ArtA directly binds to the IS1 internal region and stimulates transcription.

Surgically treated diaphragmatic perforation after radiofrequency ablation for hepatocellular carcinoma

We review 6 cases of diaphragmatic perforation, with and without herniation, treated in our institution. All patients with diaphragmatic perforation underwent radiofrequency ablation(RFA) treatments for hepatocellular carcinoma(HCC) performed at Kurume University Hospital and Tobata Kyoritsu Hospital. We investigated the clinical profiles of the 6 patients between January 2003 and December 2013. We further describe the clinical presentation, diagnosis, and treatment of diaphragmatic perforation. The change in the volume of liver and the change in the Child-Pugh score from just after the RFA to the onset of perforation was evaluated using a paired t-test. At the time of perforation, 4 patients had herniation of the viscera, while the other 2 patients had no herniation. The majority of ablated tumors were located adjacent to the diaphragm, in segments 4, 6, and 8. The average interval from RFA to the onset of perforation was 12.8 mo(range, 6-21 mo). The median Child-Pugh score at the onset of perforation(8.2) was significantly higher compared to the median Child-Pugh score just after RFA(6.5)(P = 0.031). All patients underwent laparotomy and direct suture of the diaphragm defect, with uneventful post-surgical recovery. Diaphragmatic perforation after RFA is not a matter that can be ignored. Clinicians should carefully address this complication by performing RFA for HCC adjacent to diaphragm.

Gastrointestinal symptoms in a Japanese population:A health diary study

AIM： To investigate the incidence of gastrointestinal symptoms and the nature of consequent utilization of health care services in a Japanese population. METHODS： Using self-report, we conducted a prospective cohort study of a nationally representative sample of the Japanese population over a one-month period to determine the incidence of gastrointestinal symptoms of all kinds and resultant health care utilization. Both information on visits to physicians and use of complementary and alternative medicine therapies were collected. RESULTS： From a total of 3568 in the recruitment sample, 3477 participants completed a health diary （response rate 97%）. The data of 112 participants with baseline active gastrointestinal diseases were excluded from the analysis, leaving 3365 participants in the study. The incidence of gastrointestinal symptoms was 250 and the mean number of symptomatic episodes was 0.66 in a month. Abdominal pain, diarrhea, nausea, constipation and dyspepsia were the most frequent symptoms. Female gender, younger age, and low baseline quality of life were risk factors for developing these symptoms. The participants were more likely to treat themselves, using dietary, complementary or alternative medicines, than to visit physicians, except in the case of vomiting.CONCLUSION： Gastrointestinal symptoms are common in the Japanese population, with an inddence of 25%. Abdominal pain, diarrhea, nausea, constipation and dyspepsia are the most frequent symptoms. Risk factors for developing these symptoms include female gender, younger age, and low baseline quality of life.

Carbon-carbon bond formation reactions are highly important in organic chemistry, because these reactions may be used to construct complicated molecules. Samarium diiodide is known to be a one-electron reducing agent and recently used in many organic reactions. A radicals is postulated as an intermediate, which may be formed by direct reduction of halides, carbonyls and other functional groups.

This study was conducted to assess the existing concentration of 17β-estradiol(E2)in the surface water samples collected from rivers and lakes around Klang Valley,Malaysia.E2,which is a natural feminizing chemical produced in female organisms,regularly used to compare with other environmental estrogens because they behave similarly and react effectively as a hormone at a very low *** was found that the average concentration of E2 in the aquatic environment of Klang Valley was(14.08 ±3.67)pg/mL,which was 14 times higher than those in the Japanese aquatic environment in this *** river system had the average concentration of(20.02±5.26)pg/mL while the lake had an average concentration of(5.91±3.39)pg/*** E2 concentration was presumed high if the sources occurred nearby the *** levels of E2 in the aquatic environment may possess threats to existing aquatic *** high level of E2 has been discovered in the aquatic environment around Klang Valley,further studies and monitoring of E2 and other EDCs concentrations are needed to determine their levels in Malaysian aquatic environment and help to control these chemicals pollution in the aquatic environment.

Effect of Dehydrated Digested Effluent of Manure on Yield and Quality of Annual Forages and Soil Chemical Properties in Southern Kyushu, Japan

Digested effluent of manure (DEM) produced by biogas-plants contains many macro- and micro-nutrients. In an experiment conducted in southern Kyushu (Japan), forage crops cultivated after receiving DEM and chemical fertilizers exhibited similar dry matter yields. From a logistical standpoint, however, DEM in liquid form is difficult to handle and apply due to the low concentration of nutrients. To overcome this shortcoming, we prepared dehydrated DEM (DDEM) by adding DEM to cattle manure without disturbing the manure fermentation process. The objective of this research was to evaluate the effect of DDEM on dry matter yield and nitrogen recovery rate in annual dual-cropping systems (summer crop of maize or sorghum and winter crop of Italian ryegrass) that are typical of the region and to compare these results to commercial cattle manure alone, in combination with chemical fertilizer (CM or CM + CF), or no fertilizer application (NF). In both cropping systems, the DDEM treatment produced similar dry matter yields (2.6 to 3.02 kg·m-2) and apparent nitrogen recovery rates (43% to 53%) as the CM + CF and CM treatments. This suggests that DDEM can potentially replace chemical fertilizers and commercial cattle manure in the region.

Elastography for the pancreas: Current status and future perspective

Elastography for the pancreas can be performed by either ultrasound or endoscopic ultrasound(EUS). There are two types of pancreatic elastographies based on different principles, which are strain elastography and shear wave elastography. The stiffness of tissue is estimated by measuring the grade of strain generated by external pressure in the former, whereas it is estimated by measuring propagation speed of shear wave, the transverse wave, generated by acoustic radiation impulse(ARFI) in the latter. Strain elastography is difficult to perform when the probe, the pancreas and the aorta are not located in line. Accordingly, a fine elastogram can be easily obtained in the pancreatic body but not in the pancreatic head and tail. In contrast, shear wave elastography can be easily performed in the entire pancreas because ARFI can be emitted to wherever desired. However, shear wave elastography cannot be performed by EUS to date. Recently, clinical guidelines for elastography specialized in the pancreas were published from Japanese Society of Medical Ultrasonics. The guidelines show us technical knacks of performing elastography for the pancreas.
