Identification of a fatty acid synthase gene(FAS1)from Laodelphax striatellus planthoppers contributing to fecundity

Fatty acid synthase (FAS) is a multifunctional enzyme that plays an important role in the formation of fatty acids. The fatty acids take part in many processes, such as cell signaling and energy metabolism, and in insects they are important in both cuticular hydrocarbon (CHC) formation and reproduction. Here we characterized the sequence structure and function of an FAS from the small brown planthopper (SBPH), Laodelphax striatellus. The full-length open reading frame (ORF) sequence of LsFAS1 was 7122 bp, encoding a predicted protein of 2373 amino acid residues. There were 7 functional domains in the LsFAS1 protein sequence. Gene expression screening by real-time quantitative polymerase chain reaction (RT-qPCR) showed that LsFAS1 was expressed in all developmental stages. Relative expression was highest at the 4th-instar and female adult stages. Among different tissues, the expression level of LsFAS1 in the ovary was the highest. Phylogenetic analysis showed that LsFAS1 clustered in a clade with 2 FASs from Nilaparvata lugens. Furthermore, these 3 FASs are related to cockroach BgFAS and locust LmFAS. After RNA interference-mediated knock-down, most treated insects died at eclosion. In addition, the lifespan of dsFAS1-treated female adults was shorter than that of the dsGFP-injected control, and offspring production decreased. Also, the expression of vitellogenin (Vg) and vitellogenin receptor (VgR) genes decreased. Virgin females dissected at days 2 and 4 post-eclosion showed many matured oocytes in planthoppers treated with dsGFP but not with dsFAS1. These data highlight the importance of LsFAS1 in SBPH, including a role in reproduction.

Energetic Particle Physics on the HL-2A Tokamak: A Review

Interaction between shear Alfvén wave(SAW) and energetic particles(EPs) is one of major concerns in magnetically confined plasmas since it may lead to excitation of toroidal symmetry breaking collective instabilities,thus enhances loss of EPs and degrades plasma confinement. In the last few years, Alfvénic zoology has been constructed on HL-2A tokamak and series of EPs driven instabilities, such as toroidal Alfvén eigenmodes(TAEs),revered shear Alfvén eigenmodes(RSAEs), beta induced Alfvén eigenmodes(BAEs), Alfvénic ion temperature gradient(AITG) modes and fishbone modes, have been observed and investigated. Those Alfvénic fluctuations show frequency chirping behaviors through nonlinear wave-particle route, and contribute to generation of axisymmetric modes by nonlinear wave-wave resonance in the presence of strong tearing modes. It is proved that the plasma confinement is affected by Alfvénic activities from multiple aspects. The RSAEs resonate with thermal ions, and this results in an energy diffusive transport process while the nonlinear mode coupling between core-localized TAEs and tearing modes trigger avalanche electron heat transport events. Effective measures have been taken to control SAW fluctuations and the fishbone activities are suppressed by electron cyclotron resonance heating. Those experimental results will not only contribute to better understandings of energetic particles physics, but also provide technology bases for active control of Alfvénic modes on International Thermonuclear Experimental Reactor(ITER) and Chinese Fusion Engineering Testing Reactor(CFETR).

Investigation of Work Hardening Behavior of Inconel X-750 Alloy

The constant strain rate uniaxial compression tests were conducted in this paper for studying the work hardening behavior and revealing the underlying microstructure evolution involved in the plastic response of the nickel-based Inconel X-750 alloy. The work hardening rate versus true strain plots of Inconel X-750 alloy resembled that of low-stacking-fault energy （SFE） alloys with distinct four stages. The dislocations were found in the planar arrangements at a strain of 0.1 located at the onset of stage II, and the dislocation density was increased and the planar arrangement configuration was partially destroyed at a strain of 0.36 located in stage III. It was unexpected that deformation twins were observed at a strain of 0.69 located in stage IV although the alloy has been classified into materials with a higher SFE value. The result is different with a similar study, in which the deformation twins were absent in Ni-Cr-based alloy Inconel 625 even when the strain was as high as 0.65. It was deemed that the low level of solution strengthening favored the deformation of matrix and the activation of slip system for twining in Inconel X-750 alloy. Unlike the low-SFE alloys that the twins were always formed at the end of stage Ⅰ, the higher SFE delayed the twin formation to stage IV for Inconel X-750 alloy. The well- developed planar dislocation configuration gave rise to the stage Ⅱ with a slightly decreasing rate, the collapse of planar dislocation arrangements caused the occurrence of stage Ⅲ with an accelerated decreasing rate, and the twin formation led to the stage IV with a nearly constant work hardening rate.

Autophagy: a potential target for the treatment of intraocular neovascularization

The formation of neovascularization is a common pathological feature of many ocular vascular diseases, and is an important cause of vision loss in patients. Neovascularization can cause retinal hemorrhage, vitreous hemorrhage, and other serious complications, leading to loss of vision. The treatment of intraocular neovascularization is the focus of ophthalmology research. In recent years, some studies have found that autophagy is closely related to vascular endothelial growth factor and the formation of neovascularization. Autophagy is expected to become a new target for the treatment of intraocular neovascularization. Therefore, this article reviews the research on autophagy and the formation of intraocular neovascularization.

Dispersion of the Mechanical Parts Performance Indicators Based on the Concept of Random Vector

To solve the precision and reliability problem of various machinery equipments and military vehicles, some military organisations, the industrial sector and the academia at home and abroad begin to pay attention to the statistical distribution of machining dimensions, material properties and service loads, and the system reliability optimization design with constraints and reliability optimization design of various mechanical parts is studied in this way. However, the above researches focus on solving the strength and the life problem, and no studies have been done on the discrete degree and discrete pattern of other performance indicators. The concept of using a random vector to describe the mechanical parts performance indicators is presented; characteristics between the value of the vector variance matrix determinant and the sum of the diagonal covariance matrix in describing the performance indicators of vector dispersion are studied and compared. A clutch diaphragm spring is set as an example, the geometric dimension indicator is described with random vector, and the applicability of using variance matrix determinant and variance matrix trace of geometric dimension vector to describe discrete degree of random vector is studied by using Monte-Carlo simulation method and component discrete degree perturbation method. Also, the effects of different components of diaphragm spring geometric dimension vector on the value of covariance matrix determinant and the sum of covariance matrix diagonal of diaphragm spring performance indicators vector are analyzed. The present study shows that the impacts of the dispersion of diaphragm spring cone angle on every performance dispersion are all ranked first, and far exceed that of other dimension dispersion. So it must be strictly controlled in the production process. 为探究金顶侧耳麦角硫因的降脂作用机理,采用酶动力学、紫外吸收、内源荧光、同步荧光、三维荧光等多光谱和分子对接技术研究了其与胰脂肪酶之间的相互作用。结果表明:金顶侧耳麦角硫因对胰脂肪酶具有可逆非竞争型抑制作用,其半抑制浓度为15.29 mg/mL。紫外光谱显示随着加样浓度的升高,胰脂肪酶的肽键C=O基团产生π→π*跃迁现象,疏水性减弱,亲水性变强,极性增大。荧光光谱揭示它能改变胰脂肪酶的空间结构,以静态方式有效猝灭其内源荧光,在298 K下结合常数K_(a)达到1.55×10^(3)L/mol,位点数约为1。两者结合的热力学参数焓变ΔH和熵变ΔS分别为-8.57 kJ/mol与2.55 J/(mol·K),主要以氢键与静电作用互作;同步和三维荧光发现,随着样品浓度增加,最大吸收峰分别红移6和3 nm,荧光强度分别下降32.2%和25.6%,进一步证实两者结合使胰脂肪酶分子微环境疏水性减弱,极性增强。分子对接表明,麦角硫因分子可在酶催化活性中心外以氢键、电荷吸引及范德华力结合在残基位点上,从而抑制该酶活性。本研究有助于从分子水平上深入了解金顶侧耳麦角硫因的胰脂肪酶抑制作用,为后续该类食品资源加工提供基础。

New formula for predicting standard liver volume in Chinese adults

AIM To obtain a reference range of morphological indices and establish a formula to accurately predict standard liver volume(SLV) in Chinese *** Computed tomography(CT)-estimated total liver volume(CTLV) was determined in 369 Chinese adults. Age,sex,body weight,body height,body mass index,and body surface area(BSA) were recorded using CT. Total splenic volume,portal venous diameter(PVD),splenic venous diameter(SVD),and portal venous cross-sectional area(PVCSA) were also measured by CT. Stepwise multiple linear regression analysis was performed to evaluate the impact of each parameter on CTLV and to develop a new SLV formula. The accuracy of the new formula was compared with the existing formulas in a validation *** The average CTLV was 1205.41 ± 257.53 cm3(range,593.80-2250.10 cm3). The average of PVD,SVD and PVCSA was 9.34 ± 1.51 mm,7.40 ± 1.31 mm and 173.22 ± 48.11 mm2,respectively. The CT-estimated splenic volume of healthy adults varied markedly(range,46.60-2892.30 cm3). Sex,age,body height,body weight,body mass index,and BSA were significantly correlated with CTLV. BSA showed the strongest correlation(r = 0.546,P 70 years of age,and underestimated liver volume when CTLV was > 1800 *** Our new BSA-based formula is more accurate than other formulas in estimating SLV in Chinese adults.

A more efficient model of the quantum Stackelberg duopoly is proposed by using the asymmetric quantization scheme of Qin et *** this model,two entanglement factorsα(α=√γ1/γ2)andγ(γ=√γ1γ2)are introduced,which greatly expands the functions of the previously reported symmetric *** choosing proper values ofαandγ,one can better manage the market,such as suppressing the first-mover advantage and enhancing the second-mover profit to avoid abuse of oligopolistic competition,and optimizing the total quantity of the products,so to overcome the deficiencies of"first mover always wins"and"positive quantum entanglement always reduces the total quantity"in the symmetric *** proposed model here is believed to be a good tool for the government and the department to improve the economic efficiency and develop the market.

New Physics Effects on Rare Decays B_u^+→π^+■^+■^-, ρ^+■^+■-in a Top Quark Two-Higgs-Doublet Model

Using the form factors from light-cone sum rules, we study the branching ratios and forward-backward asymmetries （FBAs） of the exclusive decays Bu^＋→π^＋e^＋e^- and Bu^＋ →ρ^＋e^＋e^- （e= e,μ） in the standard model （SM） and the top quark two-Higgs-doublet model （T2HDM）. From the numerical results, we find that the new physics contributions cannot provide very large enhancement to the branching ratios and the theoretical predictions are in good agreement with the SM ones. The T2HDM effects on FBAs of these decays are small. Precision measurements of the dilepton invariant mass distributions, especially in the lower dilepton mass region, and the FBAs in the decays Bu^＋ → π^＋（ρ^＋）e^＋ e^- will greatly help in discriminating among the SM and the new physics models.
