The Establishment of an Agrobacterium-Mediated Transformation Platform for the Non-Embryogenic Calli of Vitis vinifera L.

Non-embryogenic calli （NEC） was inevitably and heavily produced when grape embryogenic calli （EC） was induced from explants or during the subculture of EC.A stable and highly efficient NEC transformation platform is required to further sort out and verify key genes which determine/switch the identity of NEC and *** this research,a vector pA5 containing a chitinase signal sequence fused to gfp （green fluorescent protein） and an HDEL motive was used to target and immobilize into Agrobacterium strain EHA105 to establish a transformation platform for Vitis vinifera *** *** was determined that NEC 10 d after subculture was the best target tissue;30 min for inoculation followed by 3 d co-cultivation with the addition of 200 μmol L-1 acetosyringone （AS） was optimized as *** use of bacterial densities as 1.0 at OD600 did not result in serious tissue hypersensitive reaction and it had higher *** at 200 mg L-1 was picked for positive expression *** stable transformation of NEC was proved by reverse transcription-polymerase chain reaction techniques （RT-PCR） and fluorescent microscopy after three sub-cultures of the selected cell *** efficient genetic transformation protocol of grape NEC was achieved and some of the optimized parameters were different from that reported for *** transformation platform could facilitate the verification of candidate somatic embryogenesis （SE） decisive genes,and the successfully transformed NEC with certain genes can also be used as bioreactors for the production of functional products,as NEC not only proliferates fast,but also keeps in a rather stable condition.

The effect of setting time(0-36 h)at 4℃ on the quality changes of shrimp(Solenocera crassicornis)gel was evaluated in this *** quality of shrimp gel over setting time was evaluated by physical properties including gel strength,drip loss,moisture distribution and *** physical properties of the shrimp gel were effectively improved after more than 24 h of *** addition,the mechanism of the shrimp gel formation was also studied by combining chemical bonds,total sulfhydryl,disulfide bond,intrinsic fluorescence spectra and protein *** results indicated that the main forces affecting the physical properties of the shrimp gel were hydrogen bond and hydrophobic interaction,and the formation of disulfide bond during the setting process was beneficial to the construction of a uniform gel ***,setting for 24-30 h at 4℃ was an effectual way to improve the shrimp gel qualities.

The Ugi four-component reaction (U-4CR) was utilized to prepare divalent and trivalent cluster mannosides. Thus, two target compounds 6 and 8 were obtained efficiently using carboxymethyl 2, 3, 4, 6-tetra-O-acetyl--D-mannopyranoside 4 as acid component, and 1, 6-hexanediamine or tris(2-aminoethyl)amine as the multivalent scaffolds.

Background:The constructing PERMA model in psychological intervention was applied to patients with chronic cancer pain to provide data reference for reducing pain,fear of cancer recurrence,and improving the level of psychological ***:To explore the clinical effects of constructing PERMA model in psychological intervention for patients with chronic cancer ***:A total of 98 patients with chronic cancer pain admitted to our hospital from March 2021 to March 2023 were randomly divided into two groups,49 cases in each *** control group received routine intervention,while the research group constructed PERMA model based on the routine intervention for psychological *** pain severity,fear of cancer recurrence,psychological capital and quality of life before and after intervention were compared between the two ***:After two months of intervention,the pain in the research group was milder than that in the control group(pructed in psychological interventions for patients with chronic cancer pain can assist analgesic medications to reduce pain and alleviate the fear of cancer recurrence,increase the level of psychological capital,and thus improve quality of life.

Research on Nitrogen Removal and Microorganism in a Subsurface Flow Constructed Wetland System in Sihong County

Experiments in monitoring the removal of organic material and nitrogen and determining the amounts of mi- croorganism at different sites in the subsurface flow constructed wetland in Sihong county were performed. The results show that the removal of CODCr agrees with the kinetic equation of a first order reaction. The removal of pollutants varies with different seasons. The removal rates of CODCr, NH3-N, TN in the spring are 15%–23% higher than those in the autumn. The amount of ammonifier is larger than that of denitrifying bacteria and the amount of denitrifying bacte- ria is larger than that of nitrosomonas. The amount of bacteria around the plant roots is larger than that on the surface of the packing medium. No apparent change is observed for the amount of denitrifying bacteria and nitrosomonas between spring and autumn.

Ge surface passivation by GeO2 fabricated by N2O plasma oxidation

In this paper, Ge surface passivation by Ge O2 grown by N2 O plasma oxidation is presented and experimentally demonstrated. Results show that stoichiometrically Ge O2 can be achieved by N2 O plasma oxidation at 350°C. The transmission electron microscope observation reveals that the Ge O2/Ge interface is automatically smooth and the thickness of Ge O2is～0.9 nm with 120 s N2 O plasma oxidation. The interface state density of Ge surface after N2 O plasma passivation is about～3×1011 cm-2e V-1. With Ge O2 passivation,the hysteresis of MOS capacitor with Al2O3 as gate dielectric is reduced to～55 m V, compared to 130 m V of the untreated one. The Fermi-level at Ge O2/Ge interface is unpinned, and the surface potential is effectively modulated by the gate voltage, which is promising for high performance NMOSFETs fabrication.

Treatment of cutaneous lichen planus with ALA-mediated topical photodynamic therapy

Purpose:To evaluate the efectiveness of topical 5-aminolevulinie acid(ALA)-mediated pho-todynamic therapy(PDT)for the treatment of cutaneous lichen planus(LP).Methods:A total of 17 symptomatic LP lesions in 7 Chinese patients were *** cream(10%)was applied topically to LP lesions for *** lesions were iradiated with a 635 nm diode laser at the dose level of 100J/cm^(2).The treatment was repeated at two week *** assessmnent was conducted before each ***-up was performed once a month for up to six ***:Lesions showed significant improvement after one to four courses of *** response was achieved in 13 lesions(five patients)and partial remission in four lesions(two patients).The complete response rate was 71%.There was no significant side ffects except the feeling of pain that most patients could ***-up of five patients who achieved complete response showed no signs of ***:Topical ALA PDT is effective in the treatment of cutaneous LP.

An epididymis-specific carboxyl esterase CES5A is required for sperm capacitation and male fertility in the rat

Despite the fact that the phenomenon of capacitation was discovered over half century ago and much progress has been made in identifying sperm events involved in capacitation, few specific molecules of epididymal origin have been identified as being directly involved in this process in vivo. Previously, our group cloned and characterized a carboxyl esterase gene CesSa in the rat epididymis. The CES5A protein is mainly expressed in the corpus and cauda epididymidis and secreted into the corresponding lumens. Here, we report the function of CESSA in sperm maturation. By local injection of Lentivirus-mediated siRNA in the *** region of the rat epididymis, CesSa-knockdown animal models were created. These animals exhibited an inhibited sperm capacitation and a reduction in male fertility. These results suggest that CESSA plays an important role in sperm maturation and male fertility.

Soil moisture response to land use and topography across a semi-arid watershed: Implications for vegetation restoration on the Chinese Loess Plateau

Soil moisture is a limiting factor of ecosystem development in the semi-arid Loess Plateau. Characterizing the soil moisture response to its dominant controlling factors, such as land use and topography, and quantifying the soil-water carrying capacity for revegetation is of great significance for vegetation restoration in this region. In this study, soil moisture was monitored to a depth of 2 m in three land use types(native grassland, introduced grassland,and forestland), two landforms(hillslope and gully),and two slope aspects(sunny and shady) in the Nanxiaohegou watershed of the Loess Plateau,Northwest China. The MIKE SHE model was then applied to simulate the soil moisture dynamics under different conditions and determine the optimal plant coverage. Results showed that the average soil moisture was higher in native grassland than in introduced grassland and Platycladus orientalis forestland for a given topographic condition;however,a high soil moisture content was found in Robinia pseudoacacia forestland, with a value that was even higher than the native grassland of a sunny slope. The divergent results in the two forestlands were likely attributed to the differences in plant coverage. Gully regions and shady slopes usually had higher soil moisture, while lower soil moisture was usually distributed on the hillslope and sunny ***, the mean absolute relative error and Nash-Sutcliffe efficiency coefficient of the MIKE SHE model ranged between 2.8%–7.8% and 0.550–0.902,respectively, indicating that the model could effectively simulate the soil moisture dynamics. The optimal plant coverage was thus determined for hillslope P. orientalis by the model, corresponding to a leaf area index(LAI) value of 1.92. Simultaneous determination of naphazoline and pyridoxine in eye drops using excitation–emission matrix fluorescence coupled with second-order calibration method based on alternating trilinear decomposition algorithm

A novel method is developed for the direct determination of naphazoline hydrochloride（NAP） and pyridoxine hydrochloride（VB6） in commercial eye drops. By using excitation–emission matrix（EEM）fluorescence coupled with second-order calibration method based on the alternating trilinear decomposition（ATLD） algorithm, the proposed approach can achieve quantitative analysis successfully even in the presence of unknown and uncalibrated interferences. The method shows good linearity for NAP and VB6 with correlation coefficients greater than 0.99. The results were in good agreement with the labeled contents. To further confirm the feasibility and reliability of the proposed method, the same batch samples were analyzed by multiple reaction monitoring（MRM） based on LC–MS/MS method.T-test demonstrated that there are no significant differences between the prediction results of the two methods. The satisfactory results obtained in this work indicate that the use of the second-order calibration method coupled with the EEM is a promising tool for industrial quality control and pharmaceutical analysis due to its advantages of high sensitivity, low-cost and simple implementation.
