Anti-hepatocarcinoma effect of polysaccharides extracted from Lactarius hatsudake Tanaka mushroom: in vitro and in vivo studies

Edible mushroom polysaccharides demonstrate a range of bioactivities and have been investigated as potential adjuvant therapies for cancer treatment. Our prior research found Lactariks hatsudake Tanaka polysaccharide (LHP) effective against hepatocellular carcinoma， but the mechanisms remain unclear. This study aims to clarify the anti-cancer molecular mechanisms by examining the effects on specific signaling pathways through in vivo and in vitro analyses. Animal studies showed that LHP significantly inhibited the growth of transplanted liver cancer without toxicity. Histological and immunohistochemical analyses revealed that LHP increased nuclear cohesion in tumor cells and altered the expression of key regulatory proteins such as Ak strain transformation， nuclear factor kappa B， caspase-3， p21， and p53. These findings suggest that LHP's anticancer effects involve promoting apoptosis and disrupting the cell cycle. At the cellular level， assays such as MTS， flow cytometry， and transwell invasion confirmed that LHP inhibited cancer cell proliferation and induced apoptosis. Polymerase chain reaction and western blot assays revealed that LHP intervention reduced cyclin-dependent kinase 4 activity by upregulating p53 and p21， leading to cell cycle arrest and inhibited cell proliferation. LHP also promoted apoptosis by altering Bcl-2-associated X protein， B-cell lymphoma 2， and cleaved caspase-3 levels. Moreover， LHP could affect the Ak strain transformation， nuclear factor kappa B， and mitogen-activated protein kinase signaling pathways to exert anticancer effects. These findings underscore the potential of LHP as a novel and multifaceted therapeutic agent. Furthermore， the elucidation of LHP-related mechanisms offers a crucial theoretical foundation for developing innovative and effective treatments for hepatocellular carcinoma. Additionally， this supports its potential development as a functional food or adjuvant therapy for cancer， which holds significant implication

Fragility is one of the central concepts in glass and liquid sciences,as it characterizes the extent of deviation of viscosity from Arrhenius behavior and is linked to a range of glass ***,the intervention of crystallization often prevents the assessment of fragility in poor glass-formers,such as supercooled metallic *** experimental data on their compositional dependence are scarce,let alone fundamentally *** this work,we use fast scanning calorimetry to overcome this obstacle and systematically study the fragility in a ternary La–Ni–Al system,over previously inaccessible composition *** observe fragility dropped in a small range with the Al alloying,indicating an alloying-induced fragility *** use x-ray photoelectron spectroscopy,resistance measurements,electronic structure calculations,and DFT-based deep-learning atomic simulations to investigate the cause of this fragility *** results show that the fragility crossover can be fundamentally ascribed to the electronic covalency associated with the unique Al–Al *** findings provide insight into the origin of fragility in metallic liquids from an electronic structure perspective and pave a new way for the design of metallic glasses.

Previous studies have confirmed that acupuncture for irritable bowel syndrome (IBS) provided an additional benefit over usual care alone. Therefore, we performed a multicenter, randomized, sham-controlled trial to assess the efficacy and safety of acupuncture versus sham acupuncture for refractory IBS in patients in the context of conventional treatments. Patients in the acupuncture and sham acupuncture groups received real or sham acupuncture treatment in 3 sessions per week for a total of 12 sessions. The primary outcome was a change in the IBS-Symptom Severity Scale (IBS-SSS) score from baseline to week 4. A total of 521 participants were screened, and 170 patients (85 patients per group) were enrolled and included in the intention-to-treat analysis. Baseline characteristics were comparable across the two groups. From baseline to 4 weeks, the IBS-SSS total score decreased by 140.0 (95% CI: 126.0 to 153.9) in the acupuncture group and 64.4 (95% CI: 50.4 to 78.3) in the sham acupuncture group. The between-group difference was 75.6 (95% CI: 55.8 to 95.4). Acupuncture efficacy was maintained during the 4-week follow-up period. There were no serious adverse events. In conclusion, acupuncture provided benefits when combined with treatment as usual, providing more options for the treatment of refractory IBS.

Purpose:Under-foot impact loadings can cause serious lower limb injuries in many activities,such asautomobile collisions and underbody explosions to military *** present study aims to comparethe biomechanical responses of the mainstream vehicle occupant dummies with the human body lowerlimb model and analyze their robustness and applicability for assessing lower limb injury risk in underfoot impact loading ***:The Hybrid III model,the test device for human occupant restraint(THOR)model,and a hybridhuman body model with the human active lower limb model were adopted for under-foot impactanalysis regarding different impact velocities and initial lower limb ***:The results show that the 2 dummy models have larger peak tibial axial force and highersensitivity to the impact velocities and initial postures than the human lower limb *** particular,the Hybrid III dummy model presented extremely larger peak tibial axial forces than the human lowerlimb *** the case of minimal difference in tibial axial force,Hybrid III's tibial axial force(7.5 KN)isstill 312.5%that of human active lower limb's(2.4 KN).Even with closer peak tibial axial force values,thebiomechanical response curve shapes of the THOR model show significant differences from the humanlower limb ***:Based on the present results,the Hybrid III dummy cannot be used to evaluate the lowerlimb injury risk in under-foot loading *** contrast,potential improvement in ankle biofidelity and related soft tissues of the THOR dummy can be implemented in the future for betterapplicability.

基于机器学习的交通碳排放预测模型构建与分析

针对交通运输碳排放问题,基于2005~2019年30个省份的面板数据,采用多种机器学习算法构建不同预测模型对30个省份的交通碳排放量与影响因素进行分析.首先,基于固定效应模型思想将省份差异转化为影响因素,进一步采用Pearson相关系数法与Spearman秩相关系数相结合的方法对18个交通碳排放影响因素进行筛选;其次,采用K-折交叉验证方法,并绘制学习曲线对各预测模型性能进行测试,选用MSE、MAE、R2和MAPE作为模型的评价指标进行分析,来选定最佳预测模型,并选择SHAP值来计算最佳预测模型中各解释变量的重要度.结果表明,省份差异、社会商品消费总额、城市绿地面积、货运周转量、私家车数量、交通运输业产值和常住人口这7个因素之间多重共线性弱且均通过显著性检验,可作为交通运输碳排放预测模型的解释变量;随机森林算法和XGBoost算法预测结果均表现优异,R2均高于0.97,误差均低于10%,且不存在过拟合与欠拟合现象,其中XGBoost算法表现最优,而KNN算法表现欠佳;各解释变量的重要度排名为:省份差异>社会商品消费总额>私家车数量>常住人口>货运周转量>城市绿地面积>交通运输业产值,综合相关性与重要性分析来看,在交通运输碳排放预测中,省份差异是一个不可忽视的变量.研究结果可为政策制定者和决策者提供参考,促进交通运输行业的可持续发展.

Crocus sativus (saffron) is a globally autumn-flowering plant, and its stigmas are the most expensive spice and valuable herb medicine. Crocus specialized metabolites, crocins, are biosynthesized in distant species, Gardenia (eudicot) and Crocus (monocot), and the evolution of crocin biosynthesis remains poorly understood. With the chromosome-level Crocus genome assembly, we revealed that two rounds of lineage-specific whole genome triplication occurred, contributing important roles in the production of carotenoids and apocarotenoids. According to the kingdom-wide identification, phylogenetic analysis, and functional assays of carotenoid cleavage dioxygenases (CCDs), we deduced that the duplication, site positive selection, and neofunctionalization of Crocus-specific CCD2 from CCD1 members are responsible for the crocin biosynthesis. In addition, site mutation of CsCCD2 revealed the key amino acids, including I143, L146, R161, E181, T259, and S292 related to the catalytic activity of zeaxanthin cleavage.
