Surat city is continuously expanding with industrial development. Despite several flyovers, traffic congestion continues in the heart of city. Therefore, fresh transportation planning is at anvil. Simultaneously, further expansion plan is under consideration to create yet another, the 10th industrial zone. Purpose of this paper is to explore lean management based sustainable transport system for meeting the increasing needs of people and supply-chain. The research examines different mode of transportation like; vehicles on road, rail on the land, and steamers through the sea route to provide connectivity. This study may support the planners of the smart city. The study intends to find how lean management can be integrated to sustainable transport system meeting objectives of lean and sustainability both. Lean management system claims to be capable of eliminating all type of wastes including those associated with transport system. On the other hand, sustainable transport claims to meet the needs of people in personal economic lives, importantly respecting the need of future generations. Therefore, this applied research aims to find a lean and sustainable transport solution. This research work can be seen as descriptive type as it includes survey of location and fact finding enquiries.

5-Fluorouracil(5-FU)is an anticancer drug extensively used for different *** metabolic activation leads to several nucleoside and nucleotide metabolites essential to exert its cytotoxic activity on multiple cellular targets such as enzymes,DNA and *** this paper,we describe the development of a method based on liquid chromatography coupled with high resolution mass spectrometry suitable for the simultaneous determination of the ten anabolic metabolites(nucleoside,nucleotide and sugar nucleotide)of *** chromatographic separationwas optimized on a porous graphitic carbon column allowing the analysis of the metabolites of 5-FU as well as endogenous *** detection was performed on an Orbitrap■tandem mass *** of the method was verified in intracellular content and in RNA *** limit of detection was equal to 12 pg injected on column for nucleoside metabolites of 5-FU and 150 pg injected on column for mono-and tri-phosphate nucleotide *** effect was evaluated in cellular contents,DNA and RNA extracts for nucleoside and nucleotides *** method was successfully applied to i)measure the proportion of each anabolic metabolite of 5-FU in cellular contents,ii)follow the consequence of inhibition of enzymes on the endogenous nucleotide pools,iii)study the incorporation of metabolites of 5-FU into RNA and DNA,and iv)to determine the incorporation rate of 5-FUrd into 18 S and 28 S sub-units of rRNA.

Droughts occur in all climatic regions around the world costing a large expense to global economies. Reasonably accurate prediction of drought events helps water managers proper planning for utilization of limited water resources and distribution of available waters to different sectors and avoid catastrophic consequences. Therefore, a means to create a simplistic approach for forecasting drought conditions with easily accessible parameters is highly desirable. This study proposes and evaluates newly developed accurate prediction models utilizing various hydrologic, meteorological, and geohydrology parameters along with the use of Artificial Neural Network (ANN) models with various forecast lead times. The present study develops a multitude of forecasting models to predict drought indices such as the Standard Precipitation Index with a lead-time of up to 6 months, and the Soil Moisture Index with a lead-time of 3 months. Furthermore, prediction models with the capability of approximating surface and groundwater storage levels including the Ross River Dam level have been developed with relatively high accuracy with a lead-time of 3 months. The results obtained from these models were compared to current values, revealing that ANN based approach can be used as a simple and effective predictive model that can be utilized for prediction of different aspects of drought scenarios in a typical study area like Townsville, North Queensland, Australia which had suffered severe recent drought conditions for almost six recent years (2014 to early 2019).

＜正＞This paper describes a global CFD model of heat,water vapour and Botrytis *** transfer in a commercial size plastic *** focus in this article on the description of a new specific module to compute the interactions between the airborne spores and their deposition on soil,insect- proof nets and vegetation rows. The equations of flow are solved with the Fluent（?）CFD code and the standard k-ε turbulence model and we present specific models for the determination of source/sink terms in the equation of transport of scalars corresponding to i)the air water vapour and ii)the spore concentration in the *** the particle transport,three kinds of sink terms have been considered for respectively the soil surface,the insect-proof nets surface and for the vegetation *** parameters of the model together with the experimental measurements and climate boundary conditions have been determined in a parallel experimental study for a rose greenhouse with roof and lateral vents equipped with insect-proof nets. Comparisons with experimental results of greenhouse air exchange rates and inside climate （temperature and humidity）are presented and discussed together with measured and computed values of inside air spores concentration and deposition on the crop cover. First results on distributed values of air flow, temperature,humidity and spores concentration and deposition fields within the greenhouse are presented and commented.

Background: Despite humans being exposed to complex chemical mixtures, much of the available research continues to focus on a single compound or metabolite or a select subgroup of compounds inconsistent with the nature of human exposure. Uncertainty regarding how best to model chemical mixtures coupled with few analytic approaches remains a formidable challenge and served as the impetus for the study. Objectives: To identify the polychlorinated biphenyl (PCB) congener(s) within a chemical mixture that was most associated with an endometriosis diagnosis using novel graphical modeling techniques. Methods: Bayesian Belief Network (BBN) models were developed and empirically assessed in a cohort comprising 84 women aged 18 - 40 years who underwent a laparoscopy or laparotomy between 1999 and 2000;79 (94%) women had serum concentrations for 68 PCB congeners quantified. Adjusted odds ratios (AOR) for endometriosis were estimated for individual PCB congeners using BBN models. Results: PCB congeners #114 (AOR = 3.01;95% CI = 2.25, 3.77) and #136 (AOR = 1.79;95% CI = 1.03, 2.55) were associated with an endometriosis diagnosis. Combinations of mixtures inclusive of PCB #114 were all associated with higher odds of endometriosis, underscoring its potential relation with endometriosis. Conclusions: BBN models identified PCB congener 114 as the most influential congener for the odds of an endometriosis diagnosis in the context of a 68 congener chemical mixture. BBN models offer investigators the opportunity to assess which compounds within a mixture may drive a human health effect.

Quantitation of Genetox Impurities Using a Surrogate Standard Approach

With the ever increasing complexity of active pharmaceutical ingredient (API) preparations, more potential genotoxic impurities (PGI’s) are being observed. It is thus necessary to determine if these PGI’s are present in the final API’s, and if they are present, to ensure the levels are acceptable for any clinical uses. For PGI’s that have authentic standards available, quantitation can be accomplished in a straightforward manner. However, for PGI’s that are expected to form through rearrangements or side reactions, authentic standards may not be readily available, significantly complicating the analysis. In this study we describe a surrogate standard approach for quantifying PGI’s that allows for relative response factor calculations of PGI species utilizing both gas chromatography-mass spectrometry (GC-MS) and liquid chromatography-mass spectrometry (LC-MS).

Otoferlin,an integral membrane protein implicated in a late stage of exocytosis,has been reported to play a critical role in hearing although the underlying mechanisms remain ***,its widespread tissue distribution infers a more ubiquitous role in synaptic vesicle ***,an excitatory neurotransmitter,is converted to its inhibitory counterpart,g-aminobutyric acid(GABA),by L-glutamic acid decarboxylase(GAD),which exists in soluble(GAD67)and membrane-bound(GAD65)*** the first time,we have revealed a close association between otoferlin and GAD65 in both HEK293 and neuronal cells,including SHSY5Y neuroblastoma and primary rat hippocampus cells,showing a direct interaction between GAD65 and otoferlin’s C2 *** primary rat hippocampus cells,otoferlin and GAD65 co-localized in a punctate pattern within the cell body,as well as in the axon along the path of vesicular ***,GABA is virtually abolished in otoferlin-knockdown neuronal cells whereas otoferlin overexpression markedly increases endogenous *** attenuation in otoferlin-knockdown primary cells is correlated with diminished L-type calcium *** previously unknown and close correlation demonstrates that otoferlin,through GAD65,modulates GABAergic *** discovery of otoferlin–GAD65 functional coupling provides a newavenue for understanding the molecular mechanism by which otoferlin functions in neurological pathways.

In stressful or anxiety-provoking situations,most people with Parkinson’s disease(PD)experience a general worsening of motor symptoms,including their gait ***,a proportion of patients actually report benefits from experiencing-or even purposely inducing-stressful or high-arousal *** data from a large-scale international survey study among 4324 people with PD and gait impairments within the online Fox Insight(USA)and ParkinsonNEXT(NL)cohorts,we demonstrate that individuals with PD deploy an array of mental state alteration strategies to cope with their gait ***,these strategies differ along an axis of arousal-some act to heighten,whereas others diminish,overall sympathetic ***,our observations suggest that arousal may act as a double-edged sword for gait control in *** propose a theoretical,neurobiological framework to explain why heightened arousal can have detrimental effects on the occurrence and severity of gait impairments in some individuals,while alleviating them in ***,we postulate that this seemingly contradictory phenomenon is explained by the inherent features of the ascending arousal system:namely,that arousal is related to task performance by an inverted u-shaped curve(the so-called Yerkes and Dodson relationship).We propose that the noradrenergic locus coeruleus plays an important role in modulating PD symptom severity and expression,by regulating arousal and by mediating network-level functional integration across the *** ability of the locus coeruleus to facilitate dynamic‘cross-talk’between distinct,otherwise largely segregated brain regions may facilitate the necessary cerebral compensation for gait impairments in *** the presence of suboptimal arousal,compensatory networks may be too segregated to allow for adequate ***,with supraoptimal arousal,increased cross-talk between competing inputs of these complementary networks may emerge and become dysf
