Petiveria alliacea Suppresses Airway Inflammation and Allergen-Specific Th2 Responses in Ovalbumin-Sensitized Murine Model of Asthma

Objective： To examine the effect of metanol extract of Petiveria alliacea （PM） on airway inflammation in a murine model of chronic asthma. Methods： Two-month-old male BALB/c mice （n=6-8/group） were sensitized on days 0 and 14 by intraperitoneal injection of 20μg ovalbumin （OVA）. On day 25, the mice received an airway challenge with OVA （3%, w/v, in phosphate buffered saline）. PM was administered orally by oral gavage to mice at doses of 100, 200 and 400 mg/kg body weight once daily from days 18 to 23. Control mice were orally administered phosphate buffered saline （PBS） to induce a model of asthma. At the end of the test, respiratory reactivity was assayed, the total cell number, interleukin-4 （IL-4）, IL-5, IL-13, tumor necrosis factor-alpha （TNF-α） and reactive oxygen species （ROS） in the bronchoalveolar lavage fluid （BALF） were determined and the levels of serum IgE, intercellular cell adhesion molecule 1 （ICAM-1） and eotoxin were measured. In addition, lung tissue was used to qualify the IL-4, IL-5, IL-13, TNF-α and transforming growth factor beta 1 （TGF-β1）. Histologic examination was performed to observe inflammatory cellular infiltration. Results： The administration of PM in comparison with the OVA-only treated group significantly attenuated the infiltration of eosinophUs and other inflammatory cells （P〈0.01）. Airway resistance （RI） in the OVA-only induced group was significantly higher than that of the PBS control group （P〈0.01） when methacholine was added. TNF-α, IgE, TGF-β1 and cytokine levels IL-4, IL-5, IL-β in the BALF decreased compared to control mice （P〈0.01 or P〈0.05）. PM treatment also inhibited the production of chemokines, eotaxin and ICAM-1 in BALF （P〈0.01）, which improved lung function. Histopathological examination revealed that the sensitized treated PM groups had significant lower in inflammatory scores similar to dexamethasone treatments and the untreated group. Effects of bixin in high-fat diet-fed-induced fatty liver in C57BL/6J mice

Objective:To evaluate the anti-obesity activity of bixin(BIX) on C57BL/6J mice which were fed a high-fat diet(HFD) and to determine the mechanism of this ***:C57BL/6J mice were separately fed a high-calorie diet or a normal diet for 8weeks,then they were treated with BIX for another 13 *** administration for 13 weeks,the animals were *** adiposity,serum lipid level,and insulin resistance were *** addition,a histological assay of pancreas and liver,an evaluation of the inhibitory properties on pancreatic lipase,and a-amylase were ***:Administration of BIX significantly decreased the body weight gain,adipocyte size,fat pad weights,hepatic lipid levels in HFD-induced obese *** addition,reduced liver weight exhibited decreased serum leptin levels,malic enzyme,glucose-6-phosphate dehydrogenase,hepatic fatty acid synthase,aspartate aminotransferase,alanine aminotransferase and hepatic phosphatidate phosphohydrolase ***,superoxide dismutase,catalase,glutathione peroxidase,and glutathione levels were increased in hepatic *** also decreased lipid and carbohydrates absorption due to inhibition of pancreatic lipase and *** term supplementation of BIX significantly decreased hyperlipidemia,insulin resistance and glucose *** levels of hepatic steatosis and the islets of Langerhans appeared less shrunken in HFD-fed ***:The antiobesity effect of BIX appears to be associated at least in part,to its inhibitory effect on lipids and carbohydrate digestion enzymes such as pancreatic lipase,a-glucosidase,and *** results suggested that BIX also act as an antioxidant and may treat visceral obesity normalizing glucose levels,improving insulin resistance and increasing energy ***,achiote which has a main component,the carotenoid BIX,could be a viable food for the treatment of obesity and diabetes.

In vitro germination and shoot proliferation of the threatened species Cinchona officinalis L(Rubiaceae).

Cinchona officinalis （Rubiaceae） is an endemic species of the Loja Valley in southern Ecuador with medicinal uses. Because of over-exploitation in the nine- teenth century and more recent disturbances to its ecosystem, C. officinalis populations are threatened. Currently, natural regeneration of the populations is low, despite its high plant regeneration and seed formation capacity. In the present study, an efficient protocol for germination, shoot proliferation and plantlets regeneration was developed for this species. Phenolic content and germination rate of C. officinalis seeds were compared with a control species, C. pubescens. Nodal segments from seedlings of C. officinalis were cultured on Gamborg medium supplemented with different combinations of plant growth regulators. Because the phenol content is high in C. officinalis, the phenolic should be removed with hydrogen peroxide or water washes to stimulate germination. Shoots and callus developed from nodal segments within 45 days using most of the tested combinations of plant growth regulators. The best rates of shoot proliferation, callus formation and adventitious buds were obtained in medium supplemented with 5.0 mg L^-1 6-benzyl-aminopurine and 3.0 mg L^-1 indole- 3-butyfic acid.

Comparison of four methods to evaluate sperm DNA integrity between mouse caput and cauda epididymidis

It is well known that transit through the epididymis involves an increase in the compaction of sperm chromatin, which acquires fully condensed status at the caput epididymidis. The purpose of this study was to compare the terminal deoxyribonucleotidyl transferase-mediated dUTP nick end-labelling （TUNEL） assay, the comet assay, the sperm chromatin structure assay （SCSA） and the sperm chromatin dispersion （SCD） test by analysing spermatozoa from the caput and cauda epididymidis in order to demonstrate the ability of each technique to discriminate between different degrees of sperm maturity related to chromatin compaction and DNA fragmentation. Our results suggest that some populations of DNA-fragmented spermatozoa associated with immature sperm can only be identified using the comet assay and the SCSA but not with the SCD test or the TUNEL assay.

Treatment of benign paroxysmal positional vertigo. A clinical review

Benign paroxysmal positional vertigo(BPPV) is the most frequent episodic vestibular disorder. It is due to otolith rests that are free into the canals or attached to the cupulas. Well over 90% of patients can be successfully treated with manoeuvres that move the particles back to the utriculus. Among the great variety of procedures that have been described, the manoeuvres that are supported by evidenced-based studies or extensive series are commented in this review. Some topics regarding BPPV treatment, such as controlling the accuracy of the procedures or the utility of post-manoeuvre restrictions are also discussed.

Simultaneous pancreas-kidney transplantation is the treatment of choice forinsulin-dependent diabetes that associates end-stage diabetic nephropathy, sinceit achieves not only a clear improvement in the quality of life, but also provides along-term survival advantage over isolated kidney transplant. However, pancreastransplantation still has the highest rate of surgical complications among organtransplants. More than 70% of early graft losses are attributed to technical failures,that is, to a non-immunological cause. The so-called technical failures includegraft thrombosis, bleeding, infection, pancreatitis, anastomotic leak and pancreaticfistula. Pancreatic graft thrombosis leads these technical complications as the mostfrequent cause of early graft loss. Currently most recipients receive postoperativeanticoagulation with the aim of reducing the rate of thrombosis. Hemoperitoneumin the early postoperative period is a frequent cause of relaparotomy, but it is notusually associated with graft loss. The incidence of hemoperitoneum is clearlyrelated to the use of anticoagulation in the postoperative period. Post-transplantpancreatitis is another cause of early postoperative complications, less frequentthan the previous. In this review, we analyze the most common surgicalcomplications that determine pancreatic graft losses.

The impact of biophilic design in Maggie’s Centres:A meta-synthesis analysis

This paper presents a meta-synthesis conducted to identify,compare and synthesize published qualitative data related to the Maggie’s Centres,as a paradigmatic architectural example in the provision of non-institutional cancer *** centres are internationally renowned for their unique architecture,where the careful design of indoors and outdoors spaces plays a critical role in their agenda for supporting *** research has thoroughly analysed this approach to therapeutic environments,not only confirming the importance of designed space as originally conceived in Maggie’s philosophy but also identifying precise supporting effects associated with specific elements within their *** paper aims to provide a new reading of Maggie’s contribution to cancer support by offering an examination of all these data through the lens of biophilia,which clarifies the impact of design decisions connected to nature on cancer patients’*** analytic process included a systematic search strategy,extraction and classification of salient concepts using an opencoding approach,and lastly an interpretive *** systematically selected data helped to identify and rank the biophilic design parameters that appear the most critical for promoting and supporting human health and wellbeing in non-clinical therapeutic environments,from the user’s *** also provided a compilation of distinctive design interventions related to biophilic parameters,which provides benchmark information for future research and design guidance in these environments.

The Agave genus has played a key role in the food security of communities at the local and regional level for millennia. The use and knowledge of these plants led to the development of the oldest agricultural centres in America. Therefore, a review of the literature on the ethnic and cultural particularities of the use of agave species in food and current knowledge on their nutritional contribution to the human diet is presented. 56 species of agaves with food uses were registered by 55 ethnic groups in the American continent. Of this total, 23 species also have a medicinal purpose. The number of agave species used by these ethnic groups shows an important agromorphological diversity. People collect and cultivate agaves to use their stem and foliar bases, the floral scape, the flowers and flower buds, the leaves, and the sap to prepare different foods. In many cases these foods help them survive in adverse conditions. The contribution of agaves to human nutrition has been studied from nutraceutical components such as saponins, inulin, fructans, phenolic compounds and antioxidants, dietary fiber, amino acids, and minerals. In the face of global climate change, agaves represent a strategic biological resource for the world, making their use, conservation and study a priority. Agave products will continue to show their health benefits. Human use of agaves aids diversification of the genus while contributing to the well-being and permanence of a variety of cultures.
