Developing a fluorescence substrate for HRP-based diagnostic assays with superiorities over the commercial ADHP

The combination of horseradish peroxidase(HRP)and a fluorescence substrate has been attracting great interests in developing sensitive biochemical analysis and immunoassays.10-Acetyl-3,7-dihydroxyphenoxazine(ADHP or Amplex red)is the most sensitive fluorogenic substrate known for HRP in current market,however,it suffers from some drawbacks,such as non-specific reactivity to carboxylesterase and limited fluorescence *** the present study,a novel HRP substrate10-cyclopropylcarbonyl-dichloro-dihydroxyphenoxazine(AR-2),has been prepared,which exhibited improved sensitivity than ADHP in sensing ***,the fluorescence of AR-2/HRP demonstrated improved tolerance to physiological relevant p H fluctuation as compared to ADHP/*** detection of uric acid/urate oxidase reaction indicated excellent application prospect of AR-2/HRP for monitoring H_(2)O_(2)-generating biochemical *** interestingly,an enzyme-linked immunosorbent assay(ELISA)using AR-2 as the fluorescence reporter has been successfully used in detecting IgG against severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)from human serum ***,AR-2 exhibits improved performances over the commercial ADHP,which will be an ideal alternative to ADHP in HRP-based fluorescence biochemical analysis and immunoassays.

Induced Absolute Configuration of Achiral Tetradentate Ligands in Metal–Organic Frameworks for Circularly Polarized Luminescence

The crystallization of chiral molecules is of great significance to understand the origin and evolution of hierarchical chirality and reveal the relationships between structural chirality and circularly polarized luminescence(CPL)***,we report two pairs of chiral metal–organic frameworks(MOFs)(DCF-17/LCF-17,DCF-18/LCF-18)by utilizing tetradentate ligands tetra(3-imidazoylphenyl)ethylene(TIPE)and 4,4'-[4',5'-bis[4-(4-pyridinyl)phenyl][1,1':2',1”-terphenyl]-4,4”-diyl]bis[pyridine](TPPP)as *** can be observed that the spontaneous resolution of the achiral ligands is converted into the induced resolution,and the ligands form the absolute configuration by using enantiopure camphoric acid(cam)as chiral induced reagent(CIR).As a result,the racemate MOFs can be driven to generate absolute homochiral *** two achiral MOFs[Cd(D-cam)(TPPP)_(0.5)](AF-1,AF=achiral framework)and[Cd(L-cam)(TPPP)_(0.5)](AF-2)were *** position disorder of D/L-cam skeleton causes the generation of nonchiralization,further leading to disappearance of symmetry breaking of *** the perspective of structure,this is the first report which reveals the chiral transfer and nonchiralization between chiral induced agents and tetradentate ***,DCF-17 and LCF-17 show CPL with luminescence dissymmetry factor(glum)of–1.0×10^(-2)and+9.2×10^(–3),*** work provides the useful evidences to reveal the induced chiral crystallization and the construction of CPL-active crystalline materials.

Heat-resistant alloys with excellent mechanical properties are widely used in various fields,and further improvement in their properties is essential to meet the requirements in new-generation advanced supercritical boilers,nuclear reactors,superheaters,and other new materials *** effectively enhance the comprehensive performance of heat-resistant alloys,second-phase particle strengthening has been widely studied,and in the face of different service envi-ronments of advanced heat-resistant steels,the selection of suitable second-phase particles is essential to maximize the performance of these *** this end,three major types of reinforcing phases in heat-resistant alloys such as carbides,rare earth oxides,and intermetallic compounds are summarized.A comparative analysis of the precipitation behavior of the reinforcing phases with different types as well as the risks and means of controlling their use in service,is *** parameters for the application of various types of second-phase particles in heat-resistant alloys are provided to support the design and preparation of new ultrahigh-performance heat-resistant alloys.

Generalized Newton-Busemann Law for Two-dimensional Steady Hypersonic-limit Euler Flows Passing Ramps with Skin-frictions

By considering Radon measure solutions for boundary value problems of stationary non-isentropic compressible Euler equations on hypersonic-limit flows passing ramps with frictions on their boundaries, we construct solutions with density containing Dirac measures supported on the boundaries of the ramps, which represent the infinite-thin shock layers under different assumptions on the skin-frictions. We thus derive corresponding generalizations of the celebrated Newton-Busemann law in hypersonic aerodynamics for distributions of drags/lifts on ramps.

Magnetized magma intrusions being sources of a weak and a strong lunar magnetic anomaly revealed by 3D distribution of magnetization

In this work, we aim to investigate the origin of the magnetic carriers in the lunar crust and the intensity of the ancient dynamo field. The magnetization and depth range of magnetic carriers are studied under a weak and a strong magnetic anomaly in Mare Tranquillitatis and in Oceanus Procellarum, respectively, where the surface ages are 3.6 and 3.3 billion years. A sophisticated threedimensional amplitude inversion software program from a geophysical survey is used to reconstruct the distributions of magnetization in the lunar crust. Because no globally measured surface magnetic field exists for the Moon, a crustal magnetic anomaly model with a grid resolution of 0.2° is used. The depth range of the magnetic source is fixed by the boundary identified by a relative criterion, which is 20%of the recovered maximum magnetization. The central burial depths of the magnetic carriers are approximately 15 km and 25 km under Reiner Gamma and Mare Tranquillitatis, respectively. The volumes of the two magnetic sources are at scales of 104 and 105 km3,respectively. The aforementioned differences may imply a hotter crust under Reiner Gamma than Mare Tranquillitatis by 3.3 billion *** results support the view that the magma intrusions magnetized by an ancient magnetic field could be the origin of magnetic anomalies under Reiner Gamma and Mare Tranquillitatis. Compared with previous works, the maximum magnetization of 3 A/m under Reiner Gamma supports the intensity of the field being several microteslas.

From common biomass materials to high-performance tissue engineering scaffold: Biomimetic preparation, properties characterization, in vitro and in vivo evaluations

Converting common biomass materials to high-performance biomedical products could not only reduce the environmental pressure associated with the large-scale use of synthetic materials,but also increase the economic *** as a very promising candidate has drawn considerable attention owing to its abundant sources and remarkable ***,pure chitosan materials usually exhibit insufficient mechanical properties and excessive swelling ratio,which seriously affected their in vivo stability and integrity when applied as tissue engineering ***,simultaneously improving the mechanical strength and biological compatibility of pure chitosan(CS)scaffolds becomes very ***,inspired by the fiber-reinforced con-struction of natural extracellular matrix and the porous structure of cancellous bone,we built silk microfibers/chitosan composite scaffolds via ice-templating *** biomimetic strategy achieved 500%of mechanical improvement to pure chitosan,and meanwhile still maintaining high porosity(>87%).In addition,the increased roughness of chitosan pore walls by embedded silk microfibers significantly promoted cell adhesion and *** importantly,after subcutaneous implantation in mice for four weeks,the composite scaffold showed greater struc-tural integrity,as well as better collagenation,angiogenesis,and osteogenesis abilities,suggesting its great potential in biomedicine.

Comparison of ocular biometric parameters between two swept-source optical coherence tomography devices and Scheimpflug tomography in patients with cataract

AIM:To assess and compare the variations and agreements across different ocular biometric parameters using swept-source optical coherence tomography(SS-OCT)and Scheimpflug tomography in patients diagnosed with ***:This prospective case series was conducted at Tianjin Medical University Eye *** total,212 eyes from 212 patients scheduled for phacoemulsification were *** were evaluated preoperatively using two SSOCT devices(IOLMaster700 and CASIA2)and Scheimpflug tomography(Pentacam).Central corneal thickness(CCT),anterior chamber depth(ACD),aqueous depth(AQD),white-to-white distance(WTW),flat simulated keratometry(Kf),steep simulated keratometry(Ks),mean keratometry(Km),and total corneal keratometry(TKm)were *** correlation coefficient(ICC),95%confidence intervals(CI)and limits of agreement(LoA)widths were conducted to assess differences and correlations between ***:All parameters,except for Ks,were significantly *** comparison revealed no significant differences between keratometry obtained by IOLMaster 700 and *** widths of all paired comparisons for Ks were>0.80 *** for WTW between IOLMaster 700 and CASIA2 and between CASIA2 and Pentacam,other Pearson’s coefficients between devices showed a strong correlation(all r>0.95).The ICC of WTW(ICC=0.438,95%CI 0.167-0.625)showed poor *** reliability of CCT,ACD,and AQD was excellent(all ICC>0.95),whereas that of TKm was good(ICC=0.827,95%CI 0.221-0.939).A significant linear correlation was also observed among ***:The ocular parameters derived from the use of IOLMaster700,CASIA2,and Pentacam exhibit significant discrepancies;as such,measurements from these devices should not be deemed as interchangeable.
