Plants detect and respond to the proximity of competitors using light signals perceived by photoreceptor proteins.A low ratio of red to far-red radiation(R:FR ratio)is a key signal of competition that is sensed by the photoreceptor phytochrome B(phyB).Low R:FR ratios increase the synthesis of growth-related hormones,including auxin and gibberellins,promoting stem elongation and other shade-avoidance *** photoreceptors that help plants to optimize their developmental configuration and resource allocation patterns in the canopy include blue light photoreceptors,such as cryptochromes and phototropins,and UV receptors,such as *** photoreceptors act by directly or indirectly controlling the activity of two major regulatory nodes for growth and development:the COP1/SPA ubiquitin E3 ligase complex and the PIF transcription *** is also an important modulator of hormonal pathways that regulate plant defense against herbivores and pathogens,including the jasmonic acid signaling pathway,In this Perspective,we discuss recent advances on the studies of the mechanisms that link photoreceptors with growth and *** these mechanisms is important to provide a functional platform for breeding programs aimed at improving plant productivity,stress tolerance,and crop health in species of agronomic interest,and to manipulate the light environments in protected agriculture.

Dear Editor, Forward genetic screens are commonly used as unbiased tools to isolate genes responsible for a phenotype of interest. In Arabidopsis thaliana, especially T-DNA activation tagging pop- ulations are frequently employed. Pharmacogenetics is the study of the association between variability in drug response and （or） drug toxicity and polymorphisms in genes. The goal of this field of science is to adapt drugs to a patient＇s specific genetic background and therefore make them more efficacious and safe. In this article we describe the variants in genes that influence either the efficacy or toxicity of common drugs used in the treatment of inflammatory bowel diseases （IBD）, ulcerative colitis （UC）, and Crohn＇s disease （CD） including sulfasalazine and mesalazine, azathioprine （AZA） and 6-mercaptopurine （6-MP）, methotrexate （MTX）, glucocorticosteroids （CSs） and infliximab. Furthermore, difficulties with pharmacogenetic studies in general and more specifically in IBD are described. Although pharmacogenetics is a promising field that already contributed to a better understanding of some of the underlying mechanisms of action of drugs used in IBD, the only discovery translated until now into daily practice is the relation between thiopurine S-methyltransferase （TPMT） gene polymorphisms and hematological toxicity of thiopurine treatment. In the future it is necessary to organize studies in well characterized patient cohorts who have been uniformly treated and systematically evaluated in order to quantitate drug response more objectively. An effort should be made to collect genomic DNA from all patients enrolled in clinical drug trials after appropriate informed consent for pharmacogenetic studies.

AIM To determine the prevalence of work disability in inflammatory bowel disease(IBD), and to assess risk factors associated with work *** For this retrospective cohort study, we retrieved clinical data from the Dutch IBD Biobank on July 2014, containing electronic patient records of 3388 IBD patients treated in the eight University Medical Centers in the Netherlands. Prevalence of work disability was assessed in 2794 IBD patients and compared with the general Dutch population. Multivariate analyses were performed for work disability(sick leave, partial and full disability) and long-term full work disability(> 80% work disability for > 2 years).RESULTS Prevalence of work disability was higher in Crohn's disease(CD)(29%) and ulcerative colitis(UC)(19%) patients compared to the general Dutch population(7%). In all IBD patients, female sex, a lower education level, and extra-intestinal manifestations, were associated with work disability. In CD patients, an age > 40 years at diagnosis, disease duration > 15 years,smoking, surgical interventions, and anti-TNFα use were associated with work disability. In UC patients, an age > 55 years, and immunomodulator use were associated with work disability. In CD patients, a lower education level(OR = 1.62, 95%CI: 1.02-2.58), and in UC patients, disease complications(OR = 3.39, 95%CI: 1.09-10.58) were associated with long-term full work *** The prevalence of work disability in IBD patients is higher than in the general Dutch population. Early assessment of risk factors for work disability is necessary, as work disability is substantial among IBD patients.

Introduction: Crohn’ s disease is a chronic inflammatory disorder of the gut . It is assumed that a defective interaction between the bacterial flora of the gut and the innate immune system plays a key role in the pathogenesis of the dis ease. This may lead to specific histological lesions. The epithelioid granuloma is particularly interesting in this regard as it is also observed in several bac terial infections of the gut. Aims and methods: We hypothesised that genetic or environmental factors with a known influence on inflammation or immunity would l ead to an increased prevalence of granulomas. Therefore, surgical specimens from 161 patients were evaluated for the presence of granulomas. Patients were genot yped for the three single nucleotide polymorphisms in caspase recruitment domain 15 (CARD15)/NOD2 associated with CD and for Asp299Gly in Toll- like receptor 4 (TLR4). Results: The overall prevalence of granulomas was 68.9% . We did not f ind a significant correlation between granulomas and TLR4 or CARD15 variants. Th e frequency of granulomas increased with more distal disease (63% small bowel, 72% right colon, 88% left colon, 90% rectum; p = 0.01). Granulomas were m ore frequent in younger patients (odds ratio 0.95 (95% confidence interval 0.9 2- 0.98) p = 0.007). Conclusion: In this study of 161 well documented CD patien ts, we found no significant association between CARD15 and TLR4 variants and gra nulomas. This finding seems to refute our initial hypothesis. However, it may be that additional factors are needed for granuloma development. Granulomas may de velop only when specific bacterial components are present. Therefore, future res earch on granuloma pathogenesis should be orientated towards detection and ident ification of bacterial components in these lesions.
