Characterization of chickpea germplasm conserved in the Indian National Genebank and development of a core set using qualitative and quantitative trait data

Chickpea is the third most important pulse crop as a source of dietary protein. Ever-increasing demand in Asian countries calls for breeding superior desi-type varieties, in turn necessitating the availability of characterized germplasm to breeders. The Indian National Genebank,located at the National Bureau of plant Genetic Resources, New Delhi, conserves 14,651 accessions of chickpea. The entire set was characterized in a single large-scale *** variation was observed for eight quantitative and 12 qualitative agro-morphological traits. Allelic richness procedure was employed to assemble a core set comprising 1103 accessions, 70.0% of which were of Indian origin. Comparable values of total variation explained by the first three principal components in the entire collection(51.1%) and the core(52.4%)together with conservation of nine pairwise r values among quantitative traits in the core collection and a coincidence rate around 99.7% indicated that the chickpea core was indeed an excellent representation of the entire chickpea collection in the National Genebank. The chickpea core exhibited greater diversity than the entire collection in agro-morphological traits, as assessed by higher variance and Shannon–Weaver diversity indices, indicating that the chickpea core maximized the phenotypic diversity available in the Indian chickpea germplasm. The chickpea core, comprising mainly indigenous desi genotypes, is expected to be an excellent resource for chickpea breeders. AIM: To examine eplerenone(Inspra, pfizer), a mineralocorticoid receptor antagonist, as a treatment option for chronic central serous chorioretinopathy(CSCR).METHODS: A retrospective consecutive case series was conducted for patients receiving oral eplerenone for chronic CSCR. At baseline and each follow-up visit,spectral domain optical coherence tomography(SD-OCT)imaging was performed, including manual measurements of the height and diameter size of subretinal fluid. The primary outcome measure was the reduction in subretinal fluid following initiation of ***: A total of 17 eyes of 13 patients treated with25 and 50 mg of oral eplerenone per day were *** fluid(SRF) decreased over time following eplerenone therapy(p = 0.007 and p =0.002, diameter and height respectively). Maximum SRF height decreased from a mean of 131.5 μm at baseline to 15.3 μm at day181+. SRF diameter decreased from an average of 2174.4μm at baseline to 46.9 μm at day 181 +. Log MAR visual acuity improved from 0.42(Snellen equivalent: 20/53) at baseline to 0.29(Snellen equivalent: 20/39) at day 181 +(p = 0.024). Central subfield thickness(CST) decreased from 339.5 μm at baseline to 270.3 μm at day 181+(p = 0.029).CONCLUSION: Eplerenone therapy resulted in significant anatomic and visual improvements in eyes with chronic CSCR.

Leucaena leucocephala is fast growing leguminous tree species, acclimatized to variety of soil and climatic conditions. It is widely used for pulp production in India. pulp mainly consists of cellulose, which is a simple polymer of unbranched β-1, 4-linked glucan chains. The polymerization of glucose residues into a β-1, 4-linked backbone is catalysed by the enzyme cellulose synthase (CesA). Here, cDNAs encoding CesA genes from Leucaena were isolated and characterized. The two complete cDNAs of 3.228 kb and 3.222 kb encoding CesA gene from L. leucocephala were designated as Ll-7CesA (FJ871987) and Ll-8CesA (GQ267555) respectively. In-silico studies showed that Ll-7CesA has 95.2% identities and Ll- 8CesA has 95.8% identities with Acacia mangium CesA2. phylogenetic analysis revealed significant similarity with known dicot CesA genes. The deduced amino acid sequence of both CesA genes contained the conserved D, D, D, QxxRW motif, eight membrane spanning regions and a putative zinc binding domain, which are characteristic of glycosyltransferases. DNA blot analysis suggested, CesA gene to be in multiple copies in Leucaena genome. Semi quantitative and quantitative real-time pCR expression analysis of Ll-7CesA gene showed more expression in stem than leaf and not detected in root where as Ll-8CesA gene was expressed more in stem than leaf and root. Overall Ll-8CesA was expressed in all tested tissues and could be involved in active cellulose biosynthesis.
