Oridonin induces apoptosis of HeLa cells via altering expression of Bcl-2/Bax and activating caspase-3/ICAD pathway

AIM: To study the mechanisms by which oridonin inhibited HeLa cell growth in vitro. METHODS: Viability oforidonin-induced HeLa cells was measured by MTT assay. Apoptotic cells with condensed nuclei were visualizedby phase contrast microscopy. Nucleosomal DNA fragmentation was assayed by agarose gel *** activity was assayed using fluorometric protease assay. ICAD, Bcl-2, and Bax proteins expression weredetected by Western blot analysis. RESULTS: Oridonin induced oligonucleosomal fragmentation of DNA andincreased caspase-3 activity, on the other hand, reduced the expression of inhibitor of caspase-3-activated DNase(ICAD), a caspase-3 substrate, at 12 h in HeLa cells. Oridonin-induced DNA fragmentation, caspase-3 activationand down-regulation of ICAD expression were effectively inhibited by a caspase-3 inhibitor, z-DEVD-fmk (z-Asp-Glu-Val-Asp-fmk). However, pretreatment with an inhibitor of poly (ADP-ribose) polymerase (PARP), 3, 4-dihydro-5-[4-(1-piperidinyl)butoxy]-1(2H)-isoquinolinone (DPQ), did not suppress oridonin-induced HeLa cell death. Inaddition, oridonin-induced apoptosis was associated with an increase in the expression of the apoptosis inducerBax, and a significant reduction in expression of the apoptosis suppressor Bcl-2 in mitochondria. CONCLUSION:Oridonin induces HeLa cells apoptosis by altering balance of Bcl-2 and Bax protein expression and activation ofcaspase-3/ICAD pathway.

Modelling of a shape memory alloy actuator for feedforward hysteresis compensator considering load fluctuation

This paper presents a hysteresis mathematical model of a shape memory alloy(SMA)actuator for feedforward hysteresis *** hysteresis model represents the relation between temperature,stress,and electric ***,based on the laws of thermodynamics,the hysteresis model of the SMA actuator is ***,the inverse of the hysteresis model is obtained to produce a compensator for non‐linear characteristics such as hysteresis and ***,the parameters of the hysteresis model are obtained from each experiment under constant load and constant temperature,and the model validity is confirmed by comparing experimentally obtained ***,the inverse model is applied to a part of feedforward hysteresis *** order to verify the effectiveness of the proposed model as hysteresis compensator,an electrical resistance control using feedback-feedforward control was conducted by numerical *** simulation results indicate advantages of the proposed mathematical model in hysteresis compensation of SMA actuator,and demonstrate that the model is capable of handling load fluctuation.

Role of autophagy in the pathogenesis of inflammatory bowel disease

Inflammatory bowel disease(IBD) results from a complex series of interactions between susceptibility genes, the environment, and the immune ***, some studies provided strong evidence that the process of autophagy affects several aspects of mucosal immune responses. Autophagy is a cellular stress response that plays key roles in physiological processes, such as innate and adaptive immunity, adaptation to starvation, degradation of aberrant proteins or organelles, antimicrobial defense, and protein secretion. Dysfunctional autophagy is recognized as a contributing factor in many chronic inflammatory diseases, including IBD. Autophagy plays multiple roles in IBD pathogenesis by altering processes that include intracellular bacterial killing, antimicrobial peptide secretion by Paneth cells, goblet cell function, proinflammatory cytokine production by macrophages, antigen presentation by dendritic cells, and the endoplasmic reticulum stress response in enterocytes. Recent studies have identified susceptibility genes involved in autophagy, such as NOD2, ATG16L1, and IRGM, and active research is ongoing all over the world. The aim of this review is a systematic appraisal of the current literature to provide a better understanding of the role of autophagy in the pathogenesis of IBD. Understanding these mechanisms will bring about new strategies for the treatment and prevention of IBD.

Feasibility of drug delivery to the eye’s posterior segment by topical instillation of PLGA nanoparticles

We investigated the delivery of drugs to the posterior segment of the eye by non-invasive topical instillation using submicron-sized poly(D,L-lactide-co-glycolide)(PLGA) nanoparticles(NPs). Surface-modified PLGA NPs were developed to improve the drug delivery efficiency to the retina and were administered as topical eye drops to mice. Chitosan(CS) and glycol chitosan(GCS), which are mucoadhesive polymers, and polysorbate 80(P80) were used as surface modifiers, and have been reported to increase the association of NPs with ***-6 was used as a model drug and fluorescent marker, and after ocular administration of PLGA NP eye drops, the fluorescence intensity of coumarin-6 was observed in the retina. The fluorescence image analysis indicated that there are several possible routes to the retina and fates of PLGA NPs in ocular tissue, and that these pathways involved the corneal,non-corneal, or uveal routes. Delivery to the mouse retina segments after topical administration was increased by surface modification with CS, GCS, or P80. Surface-modified PLGA NPs are a promising method for retinal drug delivery via topical instillation.

Wavelength conversion based on the cross-gain modulation (XGM) in a traveling-wave type semiconductor optical amplifier (TW-SOA) using assist light is theoretically studied. Taking into account the spatial and temporal variations of carrier density along the SOA length, the dependency of the conversion response on the assist light conditions is simulated both for co- and counter-propagating assist light schemes.

AIM: To study the different death pathways in human cervical cancer HeLa and melanoma A375-S2 cells Initiated by evodiamine. METHODS: Viability of evodiamine-induced HeLa and A375-S2 cells was measured by MTT assay. Apoptotic cells with condensed or fragmented nuclei were visualized by Hoechst 33258 staining. Nucleoso mal DNA fragmentation was assayed by agarose gel electrophoresis. Proportion of cell death through apoptotic and necrotic pathways was determined by LDH activity-based cytotoxicity assays. Cell cycle distribution was observed by flow cytometry. RESULTS: Evodiamine induced HeLa and A375-S2 cell death dose- and time-dependently. Norcantharidin induces apoptosis in HeLa cells through caspase, MAPK, and mitochondrial pathways

AIM: To investigate the mechanism of norcantharidin (NCTD)-induced HeLa cell apoptosis. METHODS: HeLa cell growth inhibition was measured by MTT method. Apoptosis was detected by Hoechst 33258 staining and agarose gel electrophoresis. Caspase activities were assayed using caspase apoptosis detection kit. Western blot analysis was used to evaluate the level of ICAD, ERK/p-ERK, JNK/p-JNK, and Bcl-XL/Bax expression. RESULTS: Norcantharidin inhibited HeLa cell growth in a time- and dose-dependent manner. HeLa cells treated with norcantharidin showed typical characteristics of apoptosis including the morphological changes and DNA fragmentation. Caspase family inhibitor (z-VAD-fmk), caspase-8, -9 inhibitor (z-IETD-fmk, Ac-LEHD-CHO, respectively) and caspase-3 inhibitor (z-DEVD-fmk) partially prevent norcantharidin-induced apoptosis, but initiator caspase-1 inhibitor (Ac-YVAD-fmk) did not. Apoptosis was detected by electron microscopy and agarose gel electrophoresis. Ratio of apoptotic cells was measured by APO-BRDU kit. Cell cycle distribution and changes of mitochondrial membrane potential were monitored by flow cytometry. Caspase activities were assayed by caspase apoptosis detection kit. Western blot analysis was used to evaluate the level of mitochondrial Bcl-2 expression. RESULTS: Diosgenin inhibited HeLa cell growth. HeLa cells treated with diosgenin showed typical characteristics of apoptosis including the morphological changes and DNA fragmentation. UNSTEADY INTERACTION OF SHOCK WAVE DIFFRACTING AROUND A CIRCULAR CYLINDER IN AIR

The reflection and diffraction of a planar shock wave around a circular cylinder are a typical problem of the complex nonlinear shock wave phenomena in *** has long been studied experimentally,analytically as well as *** in 1987 obtained clear experimental pictures ofisopycnics in shock tube under the condi- tion that the impinging shock wave propagates as far as 3 diameters away from the *** know more complete- ly the whole unsteady process,it is desirable to get experimental results in a region which is more than 10 diameters away from the *** is what has been done in this paper by using the pulsed laser holographic interferometry for several shock Mach numbers of the impinging shock. Results for several moments are shown,giving more know- ledge about the whole unsteady flow *** is useful for a reliable and complete understanding of the changing force acting on the cylinder,and provides interesting data to check the performance of many recently developed high resolution numerical methods for unsteady shock wave calculation.
