Genomic analysis uncovers functional variation in the C-terminus of anthocyanin-activating MYB transcription factors

MYB transcription factors regulate diverse aspects of plant development and secondary metabolism,often by partnering in transcriptional regulatory ***,we harness genomic resources to identify novel MYBs,thereby producing an updated eudicot MYB phylogeny with revised relationships among subgroups as well as new information on sequence variation in the disordered C-terminus of anthocyanin-activating ***®and hidden Markov model scans of gene annotations identified a total of 714 MYB transcription factors across the genomes of four crops that span the eudicots:apple,grape,kiwifruit and *** model-based phylogenetic inference identified novel members of previously defined subgroups,and the function of specific anthocyanin-activating subgroup 6 members was assayed transiently in tobacco *** conservation within subgroup 6 highlighted one previously described and two novel short linear motifs in the disordered C-terminal *** novel motifs have a mix of hydrophobic and acidic residues and are predicted to be relatively ordered compared with flanking protein *** of motifs with the Eukaryotic Linear Motif database suggests roles in protein–protein *** of motifs and their flanking regions from strong anthocyanin activators into weak activators,and vice versa,affected *** conclude that,although the MYB C-terminal sequence diverges greatly even within MYB clades,variation within the C-terminus at and near relatively ordered regions offers opportunities for exploring MYB function and developing superior alleles for plant breeding.

Dear Editor,Since the late 1980s, trauma in China has been identified as a major public health challenge, with trafic-related fatalities accounting for 80% of accidental deaths[1]. In 2019, Shenzhen had a total population of approximately 20 million, and from 2010 to 2017, both emergency medical services and the total number of trauma patients increased, with trauma accounting for 47.0% and 38.4% of all patients in 2010 and 2017, respectively[2,3]. This report describes the efforts to implement programs and establish a trauma system in Shenzhen, China.

Perceived Injustices in Forest Policy Interventions are Causes of Forest Resources Degradation and Loss in Ghana: A Review

Forests provide diverse co-benefits, including livelihoods enhancement, poverty alleviation, and biodiversity conservation and ecosystem services such as climate mitigation. Consequently, Ghana has embraced several global, regional, national, and local forest policy interventions to safeguard its forest resources’ sustainable management. However, the forest policy interventions have not adequately addressed the co-benefits issues of forest resources leading to several forest illegalities that drive forest resources degradation and loss. These forest policies and interventions on the ground primarily favour carbon stocks enhancement and sequestration activities for carbon mitigation purposes compared to the benefits, access and rights that forest-dependent people and communities are supposed to derive from forest resources. These perceived injustices in the access and distribution of forest resources have culminated into all forms of forest illegalities driving Ghana’s forest resources into massive degradation and loss. Currently, Ghana is experiencing a 2% rate of annual deforestation and forest degradation, which translates into approximately 135,000verdana;">verdana;">verdana;"> ha/year of forest cover loss due to anthropogenic causes. Our review is based on relevant and critical forest documents, and scientific peer-reviewed papers on Ghana’s forest policy interventions and dynamics recently published. Speciation, sources, and risk assessment of heavy metals in suburban vegetable garden soil in Xianyang City, Northwest China

Intensive anthropogenic activities can lead to soil heavy metal contamination resulting in potential risks to the environment and to human health. To reveal the concentrations, speciation, sources, pollution level, and ecological risk of heavy metals in vegetable garden soil, a total of 136 soil samples were collected from three vegetable production fields in the suburbs of Xianyang City, Northwest China. These samples were analyzed by inductively coupled plasma- atomic emission spectrometry and atomic fluorescence spectrometry. The results showed that the mean concentrations of Cd, Co, Cu, Mn, Pb, Zn, and Hg in vegetable garden soil were higher than the corresponding soil element background values of Shaanxi Province. The heavy metals studied in vegetable garden soil were primarily found in the residual fraction, averaging from 31.26% (Pb) to 90.23% (Cr). Considering the non-residual fractions, the mobility or potential risk was in the order of Pb (68.74%)>Co (60.54%)>Mn (59.28%) >Cd (53.54%) ≫Ni (23.36%) >Zn (22.73%)>Cu (14.93%)>v (11.81%)>Cr (9.78%). Cr, Mn, Ni, v, and As in the studied soil were related to soilforming parent materials, while Cu, Hg, Zn, Cd, Co, and Pb were associated with the application of plastic films, fertilizers, and pesticides, as well as traffic emissions and industrial fumes. Cr, Ni, v, and As presented low contamination levels, whereas Co, Cu, Mn, Pb, and Zn levels were moderate, and Cd and Hg were high. Ecological risk was low for Co, Cr, Cu, Mn, Pb, Zn, and As, with high risk observed for Cd and Hg. The overall pollution level and ecological risk of these heavy metals were high.

vC2002, isolated from postweaning multisystemic wasting syndrome(PMWS)-affected pig, is a mixture of two porcine circovirus genotype 2b(PCv2b) viruses, K2 and K39. Preliminary experiments disclosed short-term adverse effects of K39, but not K2, on porcine foetuses. These findings led to the hypothesis that infection of immuno-incompetent foetuses with K2 confers a status of immunotolerance, and postnatal super-infection with K39 triggers PMWS. To explore this hypothesis, nine 55-day-old foetuses were inoculated in utero(three with K2-104.3TCID50, three with K39-104.3TCID50 and three with medium), and foeto-pathogenicity examined. At 21 days post-inoculation(dpi), K2 did not induce pathology, whereas pathological effects of K39 were evident. Twenty-four 45-day-old foetuses were subsequently inoculated to examine the long-term effect of K2, including six with K2-high dose-104.3TCID50, six with K2-low dose-102.3TCID50 and 12 mock-inoculated controls. Both doses resulted in five mummified foetuses and one live-born piglet each(69dpi). K2 was recovered from all mummies. K2 and K2-specific antibodies were not detected in serum of the two live-born piglets at birth, indicating full control of K2 infection. The K2-low dose-infected piglet was immunostimulated at day 2, but not the K2-high dose-infected piglet. Both non-stimulated and stimulated K2-infected piglets were super-inoculated with K39 at day 6 or 8(taken as 0 days post super-inoculation). Low viral replication was observed in the non-stimulated K2-K39 piglet(up to 103.3 TCID50/g; identified as K39). In contrast, viral replication was extremely high in the stimulated K2-K39 piglet(up to 105.6TCID50/g) and identified as K2, indicating that K2 infection is controlled during foetal life, but emerges after birth upon immunostimulation. However, none of the piglets showed any signs of PMWS.

Correlation between plasmid DNA damage induced by PM_(10) and trace metals in inhalable particulate matters in Beijing air

Based on the study of Beijing PM10 bioreactivity with the newly developed plasmid DNA assay method, and analysis for trace elements of PM10, the cause of plasmid DNA damage by PM10 was investigated. The study showed that plasmid DNA oxidative damages by PM10 are of difference in different seasons at various areas. The concentrations of TM50 of PM10 in whole samples respectively collected at urban and comparison sites during winter were 900 μg mL?1 and 74 μg mL?1, while those in their corresponding soluble fractions were 540 μg mL?1 and 86 μg mL?1. In contrast, TM50 contents of PM10 from summer whole samples at urban areas and comparison sites were 116 μg mL?1 and 210 μg mL?1, whereas those in their soluble fractions were 180 μg mL?1 and 306 μg mL?1. The difference of bioreactivity of Beijing PM10 resulted from the variation of trace elements. The oxidative damage of plasmid DNA caused by Pb, Zn, As in PM10 (whole sample) was relatively strong. TM50 and Mn, v, Zn display stronger correlation in the soluble fraction. It implies that Zn could be the major trace element in Beijing PM10 which contributes to oxidative damage to plasmid DNA.
