Subcutaneous Model for the Study of Dengue Virus Infection in Immune Competent Mice

Various mouse models to study dengue have been described by different authors, some of them using immunodeficient or some using humanized mice. Our group reported previously a deadly murine model, which used the intracranial inoculum of highly virulent Dengue virus (DENV) in immune competent mouse. Here we present a model of immune competent mouse (C57BL/6), infected subcutaneously by the same highly virulent DENV (DENV3 genotype I). In this immunocompetent systemic mice model, the cytokine levels and hematological parameters such as total and differential leukocyte and platelets counts, together with weight loss, were considered important monitoring parameters, allowing a better understanding of the systemic human disease. Mice were inoculated subcutaneously and evaluated by the percentage weight variation as well as the clinical signs. Hematological parameters and cytokines levels were measured and viral titration in brain tissue or serum neutralization was performed to confirm mice infection. The subcutaneously DENV inoculated mice showed weight loss after infection, but they did not show any other clinical signs. The leukocytes and platelets decreased after subcutaneous inoculation. The cytokines TNF alpha and IFN gamma increased after infection in mice. The subcutaneous model provided scope for improved understanding of the dengue pathogenesis, as well as possible mechanism for protection to subsequent mouse infected by intracranial route in mice. This model could be used to study the vertebrate immune response and evaluation of drugs or vaccine against dengue virus.

The growing demand for electricity in large Brazilian cities and the distance to the region with the largest energy production have encouraged the construction of transmission lines thousands of kilometers long, crossing several states of the federation. Therefore, the heterogeneity of requirements in federal, state and district environmental laws tend to lead to uncertainty for the investor regarding procedures and investments necessary for the completion of the project, as well as the time that the requirements will take to be fulfilled. In the present study, we identify and compare the criteria used by the various environmental licensing agencies for classifying transmission system projects in the different procedures(simplified and ordinary) of environmental licensing. It was possible to expose the differences and demonstrate some inconsistencies in the legislation of some Brazilian states. In particular, we note an absence of proportionality between the licensing requirements and the expected environmental impacts for projects on the physical, biological and social environments. Additionally, it was identified subjectivity in the requirement for social participation through public hearings, demanded by the legislation for the projects classified in the ordinary procedure.

Role of pentoxifylline in non-alcoholic fatty liver disease in high-fat diet-induced obesity in mice

AIM:To study pentoxifylline effects in liver and adipose tissue inflammation in obese mice induced by high-fat diet(HFD).METHODS: Male swiss mice(6-wk old) were fed a highfat diet(HFD; 60% kcal from fat) or AIN-93(control diet; 15% kcal from fat) for 12 wk and received pentoxifylline intraperitoneally(100 mg/kg per day) for the last 14 d. Glucose homeostasis was evaluated by measurements of basal glucose blood levels and insulin tolerance test two days before the end of the protocol. Final body weight was assessed. Epididymal adipose tissue was collected and weighted for adiposity evaluation. Liver and adipose tissue biopsies were homogenized in solubilization buffer and cytokines were measured in supernatant by enzyme immunoassay or multiplex kit, respectively. Hepatic histopathologic analyses were performed in sections of paraformaldehyde-fixed, paraffin-embedded liver specimens stained with hematoxylin-eosin by an independent pathologist. Steatosis(macrovesicular and microvesicular), ballooning degeneration and inflammation were histopathologically determined. Triglycerides measurements were performed after lipid extraction in liver tissue. RESULTS: Pentoxifylline treatment reduced microsteatosis and tumor necrosis factor(TNF)-α in liver(156.3 ± 17.2 and 62.6 ± 7.6 pg/mL of TNF-α for non-treated and treated obese mice, respectively; P < 0.05). Serum aspartate aminotransferase levels were also reduced(23.2 ± 6.9 and 12.1 ± 1.6 U/L for nontreated and treated obese mice, respectively; P < 0.05) but had no effect on glucose homeostasis. In obese adipose tissue, pentoxifylline reduced TNF-α(106.1 ± 17.6 and 51.1 ± 9.6 pg/mL for non-treated and treated obese mice, respectively; P < 0.05) and interleukin-6(340.8 ± 51.3 and 166.6 ± 22.5 pg/mL for non-treated and treated obese mice, respectively; P < 0.05) levels; however, leptin(8.1 ± 0.7 and 23.1 ± 2.9 ng/mL for non-treated and treated lean mice, respectively; P < 0.05) and plasminogen activator inhibitor-1(600.2 ± 32.3

Ex-Patients with COVID-19: Observations after Healing and Its Consequences*

During the SARS-Cov-2 or COVID-19 clinical course pandemic of the coronavirus family, we observed symptoms of the respiratory tract, fever, cough, sore throat, and fatigue. The most common symptoms were loss of smell and taste and, when aggravated, it could develop into pneumonia and even death. We selected 40 former COVID-19 patients, who answered a questionnaire with questions about the symptoms and complications after they were cured and which symptoms remained or appeared after the total isolation quarantine for a specific time. Among the answers, the most cited was about “forgetting”. Another observation noted was the number of cases in children, which surprised us, considering that, at the beginning of the pandemic, the danger was more related to the elderly population and those with comorbidities.

目的：评价自酸蚀粘结系统中不同的使用方法对牙本质粘结强度的影响。材料与方法：将255颗拔出的牛牙颊面进行研磨以暴露出平坦的牙本质表面，再将牙齿分成4个实验组，使用4种自酸蚀粘结系统．分别为One Up Bond F Plus, Clearfil SE Bond Xeno Ⅲ．以及FuturaBondNR．对照组为传统的酸蚀；中洗粘结系统Adper Single Bond 2。所有实验组均主动或被动地使用一或两层的自酸蚀粘结剂．再将复合树脂粘结至牙本质．24h后在一个万能试验机上以1mm／min的速度对试样进行剪切测试。对得到的数据进行双因素方差分析．Dunnett以及Tukey检验（5％）。结果：不同的粘结剂类型、使用方法以及相互作用等因素间均有显著性的差异。所有粘结系统均表现出显著差异．主动使用两层自酸蚀粘结剂产生的平均粘结强度要明显高于被动使用的。结论：主动使用自酸蚀粘结剂能有效增加牙本质的剪切粘结强度，而且不同的使用方法对粘结的影响取决于所测试的粘结剂类型。

Understanding Ehrlichia canis Infections in Dogs of Costa rica: Hematological Findings and Indicative Clinical Signs

Background: A cross-sectional study combining different serological and molecular techniques for the detection of Ehrlichia canis in dogs was carried out to determine hemopathological findings and suggestive clinical signs associated with acute, subclinical and chronic infections in the dog population of Costa rica. Objectives: The present study describes and analyzes, in a more representative sampling frame, the clinical and hematological presentation of E. canis infection in dogs of Costa rica in all its clinical stages. Methods: A descriptive analysis of the clinical signs was performed from a 441-dog sample. Serological and molecular techniques for the detection of Ehrlichia canis in dogs were applied. One and two-way ANOVA were carried out to determine the effect of the infection status on the hematological parameters. Results: A total of 0.7% (3/407) dogs were found with acute (seronegative but PCR positive), 29.7% (121/407) with subclinical (seropositive and PCR negative), and 2.5% (10/407) with chronic (seropositive and PCR positive) E. canis infections. Significant hemopathological findings were determined in dogs with acute (thrombocytosis), subclinical and chronic (anemia, thrombocytopenia, leukopenia) E. canis infections. Conclusions: Future studies must determine if dogs with subclinical E. canis infections eliminated the agent without any medication, or if they continue to be persistently infected, and will develop the chronic disease at some point in their lives.

Hematopoietic tumors are associated with diagnostic and therapeutic challenges and a poor prognosis. Furthermore, their pathophysiology in dogs is not well understood. Chronic lymphocytic leukemia has been rarely reported in golden retrievers, perhaps due to difficulties in diagnosis and care. An 8-year-old spayed female golden retriever was referred to our hospital with a 2-month history of fatigue, weakness, and selective appetite. The dog was diagnosed with chronic lymphocytic leukemia based on blood smear evaluation. Despite treatment, the dog deteriorated and the owners opted for euthanasia. Histopathological examination revealed atypical lymphocytes in the spleen, extramedullary hematopoiesis in the spleen and liver, bone marrow depletion, moderate neutrophilic myocarditis, and acute renal tubular necrosis. In an immunohistochemical panel, cells in the spleen and bone marrow were positive for anti-CD20, anti-CD45, anti-CD3, anti-IBA-1, anti-Ki67, anti-MUM-1, and anti-PAX-5. In conclusion, the leukemia diagnosis was confirmed through reactive histiocytosis, marked extramedullary hematopoiesis, and other clinical and laboratory findings. Herein, we report a case of chronic lymphocytic leukemia in a female dog with clinical, laboratory, and necroscopy findings.

We report the preparation of poly(3,4-ethylene dioxythiophene)(PEDOT)-modified polyvinylidene fluoride electrospun fibers and their use as a novel adsorbent material for the removal of the anionic dye Methyl Orange(MO)from aqueous *** novel adsorbent material can be used to selectively remove MO on a wide p H range(3.0-10.0),with a maximum capacity of 143.8 mg/g at p H *** used in a recirculating filtration system,the maximum absorption capacity was reached in a shorter time(20 min)than that observed for batch mode experiments(360 min).Based on the analyses of the kinetics and adsorption isotherm data,one can conclude that the predominant mechanism of interaction between the membrane and the dissolved dye molecules is ***,considering the estimated values for the Gibbs energy,and entropy and enthalpy changes,it was established that the adsorption process is spontaneous and occurs in an endothermic *** good mechanical and environmental stability of these membranes allowed their use in at least 20 consecutive adsorption/desorption cycles,without significant loss of their *** suggest that the physical-chemical characteristics of PEDOT make these hybrid mats a promising adsorbent material for use in water remediation protocols and effluent treatment systems.
