Evolutionary history and phylogeography of Scots pine (Pinus sylvestris L.) in Europe based on molecular markers

In this review we summarized recent historical records and molecular studies on evolutionary history and phylogeography of Scots pine with focus on the European highly fragmented distribution area of the species. Fossilized pollen, plant micro- and macrofossil records provided evidences on the large-scale species’ range shifts and demographic changes during the Quaternary. Populations of Scots pine were documented both in the glacial (incl. full glaciation) and interglacial periods. Recolonization of Europe after the glaciation originated from the (Sub) Mediterranean areas like the Balkan Peninsula but also from around the Eastern Alps and the surroundings of the Danube plain. Fennoscandia and northern European Baltic regions were most probably colonized from two main directions, from Western Europe and from the Russian Plain. Modern history of Scots pine was hardly affected by anthropogenic activities that started to strengthen in the Bronze and Iron Age. Along with the fossil records, molecular genetic tools were used to infer the origin and putative history including migration, differentiation and demography of the species. In this paper we compiled the major publications (30) of molecular genetic studies of the past 20 years derived from distinctly inherited organelle genomes (mitochondrial, chloroplast, nuclear) revealed by different marker systems (mtDNA-cox1, -nad1, -nad3, -nad7, ISSR, cpSSR, nSSR, B-SAP, SNP). It is important to consider that different phylogeographic patterns can be drawn by the analysis of different DNA marker types. Accordingly the use of more than one marker simultaneously outlines the most sophisticated phylogeographical pattern on the genetic lineages and can reveal high differentiation of the European distribution. Combined marker systems and markers derived from coding sequences have also been used to detect species’ phylogeographic patterns, but these were rarely applied to Scots pine. Region-dependent effects of diabetes and insulin-replacement on neuronal nitric oxide synthase-and heme oxygenase-immunoreactive submucous neurons

AIM To investigate the intestinal segment-specific effects of diabetes and insulin replacement on the density of different subpopulations of submucous neurons. METHODS Ten weeks after the onset of type 1 diabetes samples were taken from the duodenum, ileum and colon of streptozotocin-induce diabetic, insulin-treated diabetic and sex-and age-matched control rats. Whole-mount preparations of submucous plexus were prepared from the different gut segments for quantitative fluorescent immunohistochemistry. The following double-immunostainings were performed: neuronal nitric oxide synthase(n NOS) and Hu C/D, heme oxygenase(HO) 1 and peripherin, as well as HO2 and peripherin. The density of n NOS-, HO1-and HO2-immunoreactive(IR) neurons was determined as a percentage of the total number of submucous neurons. RESULTS The total number of submucous neurons and the proportion of n NOS-, HO1-and HO2-IR subpopulations were not affected in the duodenal ganglia of control, diabetic and insulin-treated rats. While the total neuronal number did not change in either the ileum or the colon, the density of nitrergic neurons exhibited a 2-and 3-fold increase in the diabetic ileum and colon, respectively, which was further enhanced after insulin replacement. The presence of HO1-and HO2-IR submucous neurons was robust in the colon of controls(38.4%-50.8%), whereas it was significantly lower in the small intestinal segments(0.0%-4.2%, P < 0.0001). Under pathophysiological conditions the only alteration detected was an increase in the ileum and a decrease in the colon of the proportion of HO-IR neurons in insulin-treated diabetic animals. CONCLUSION Diabetes and immediate insulin replacement induce the most pronounced region-specific alterations of n NOS-, HO1-and HO2-IR submucous neuronal density in the distal parts of the gut.

Tensile and fracture properties of an Mg-RE-Zn alloy at elevated temperatures

Magnesium alloy EZ10 （Mg-RE-Zn） was deformed in tension at temperatures from 20 up to 520℃. A rapid decrease of the yield and tensile strength with temperature was observed at temperatures higher than 300 ℃. On the other hand, ductility of sam-ples rapidly increased in the same temperature range. Light microscopy and scanning electron microscopy was used to reveal the rea-son for these behaviours. Intermetallic particles in grain boundaries are responsible for excellent mechanical properties at lower tem-peratures. Diffusional processes occurring at temperatures higher than 300 ℃ significantly influenced the deformation mechanism as well as the fracture character.

Changes of plasma fasting carnitine ester profile in patients with ulcerative colitis

AIM： To determine the plasma carnitine ester profile in adult patients with ulcerative culitis （UC） and compared with healthy control subjects. METHOD： Using ESI triple quadrupole tandem mass spectrometry, the carnitine ester profile was measured in 44 patients with UC and 44 age- and sex-matched healthy controls. RESULTS： There was no significant difference in the fasting free carnitine level between the patients with UC and the healthy controls. The fasting propionyl- （0.331 ± 0.019 vs 0.392 ± 0.017 μmol/L）, butyryl- （0.219 ± 0.014 vs 0.265 ± 0.012）, and isovalerylcarniUne （0.111 ± 0.008 vs 0.134 ± 0.008） levels were decreased in the UC patients. By contrast, the level of octanoyl- （0.147 ± 0.009 vs 0.114 ± 0.008）, decanoyl- （0.180 ± 0.012 vs 0.137 ± 0.008）, myristoyl- （0.048 ± 0.003 vs 0.039 ± 0.003）, palmitoyl- （0.128 ± 0.006 vs 0.109 ± 0.004）, palmitoleyl- （0.042±0.003 vs 0.031 ± 0.002） and oleylcarnitine （0.183 ± 0.007 vs 0.163 ± 0.007; P 〈 0.05 in all comparisons） were increased in the patients with UC. CONCLUSION： Our data suggest selective involvement of the carnitine esters in UC patients, probably due to their altered metabolism.

Characterization of the High-Strength Mg–3Nd–0.5Zn Alloy Prepared by Thermomechanical Processing

Magnesium alloys based on Nd and Zn are promising materials for both aviation industry and medical *** mechanical properties of these materials can be achieved by thermomechanical processing such as extrusion or rolling and by aging treatment, which can significantly strengthen the alloy. The question remains especially about the connection of texture strength created in the alloys based on the specific conditions of preparation. This work focuses on the Mg–3 Nd–0.5 Zn magnesium alloy prepared by hot extrusion of the as-cast state at two different temperatures combined with heat pre-treatment. Extrusion ratio of 16 and rate of 0.2 mm/s at 350 and 400 °C were selected for material preparation. The structures of prepared materials were studied by scanning electron microscopy and transmission electron microscopy. The effect of microstructure on mechanical properties was evaluated. Obtained results revealed the strong effect of thermal pre-treatment on final microstructure and mechanical properties of extruded materials. The Hall–Petch relation between grain size and tensile yield strength has been suggested in this paper based on the literature review and presented data. The observed behavior strongly supports the fact that the Hall–Petch of extruded Mg–3Nd–0.5 Zn alloys with different texture intensities cannot be clearly estimated and predicted. In addition, Hall–Petch relations presented in literature can be sufficiently obtained only for fraction of the Mg–3Nd–0.5 Zn alloys.

Chalcone analogue as potent anti-malarial compounds against Plasmodium falciparum:Synthesis,biological evaluation,and docking simulation study

To investigate in vitro antimalarial activity of chalcone derivative compounds against Plasmodium falciparum 3D7 (Pf3D7) strain and in silico antimalarial *** of the chalcone derivatives was conducted via Claisen-Schmidt method using NaOH 60% base as catalyst. An in vitro antimalarial activity assay was carried out according to the Rieckmann method against the chloroquine-sensitive Pf3D7 strain. Molecular docking studies of the prepared compounds were performed using Discovery Studio 3.1 (Accelrys, Inc., San Diego, USA) software to dihydrofolate reductases-thymidylate synthase (PfDHFR-TS) protein with Protein Data Bank ID of *** (sensitive-protein) and ID: *** (resistance-protein).ResultsThis work has successfully synthesized seven chalcone derivatives with a great antimalarial activity. It has been revealed that allyloxy, hydroxy and alkoxy functional groups could increase the antimalarial activity of the chalcone derivatives. The best antimalarial activity of the prepared compounds was possessed by 3b with an IC50 value of 0.59 μM and categorized as an excellent antiplasmodial. Molecular docking studies of 3b showed binding interaction with the amino acid residues such as Ala16, Ile164, Phe58, Tyr170 of the *** protein and also Ala16, Phe58, Ile112, Met55 of the *** *** in vitro antimalarial assay of the prepared chalcone derivative (3a-g) showed an excellent and good antiplasmodial activity against the chloroquine-sensitive Pf3D7 strain. In silico antimalarial studies revealed that 3a-g made binding interaction with both sensitive-protein (***) and resistance-protein (***), which means that they were both active against chloroquine-sensitive and resistant plasmodium strain.

Subcellular distribution of nitric oxide synthase isoforms in the rat duodenum

Inflammation and its effects in the bone marrow microenvironment represent a paradigmatic condition in which the hematopoietic niche and the immune systems,thought to properly sustain blood cell production and distinguish between friend and foe,can actively sustain a corrupted neighborhood within a chronic aberrant inflamed *** bone marrow niche hijacks the physiologic *** interactions between the hematopoietic stem cells and the niche in the bone marrow are critical determinants of *** examined several approaches to confront the available evidence;three key points emerged,pointing to the chronic inflammation process,especially the chronic infection and systemic inflammatory states,as leading causes of hematopoietic stem cell *** hematopoiesis,defined as a relative expansion of individual clones,is caused by somatic alterations in essential hematopoietic genes,which increase stem cell ***,terminal differentiation plays a significant role in progenitor loss and inflammatory signaling,promoting clonal selection and clonal hematopoiesis *** myeloid malignancies as paradigmatic examples are discussed as a condition associated with inflammation,including the 5q-syndrome,Philadelphia negative myeloproliferative neoplasms,and chronic myeloid *** with increased fitness and hematopoietic stem cell attrition,extrinsic stress,enhanced stressor-specific fitness,and intrinsic defect across the hematopoietic process represent the route for novel insights in defective *** discussion in this review also points out that the hematopoietic niches’inflammatory stimulation may affect differentiation patterns and the function of downstream cells.

Identify multiple myeloma stem cells: Utopia?

Multiple myeloma(MM) is a hematologic malignancy of monoclonal plasma cells which remains incurable despite recent advances in therapies. The presence of cancer stem cells(CSCs) has been demonstrated in many solid and hematologic tumors, so the idea of CSCs has been proposed for MM, even if MM CSCs have not been define yet. The existence of myeloma CSCs with clonotypic B and clonotypic non B cells was postulated by many groups. This review aims to focus on these distinct clonotypic subpopulations and on their ability to develop and sustain MM. The bone marrow microenvironment provides to MM CSCs self-renewal, survival and drug resistance thanks to the presence of normal and cancer stem cell niches. The niches and CSCs interact each other through adhesion molecules and the interplay between ligands and receptors activate stemness signaling(Hedgehog, Wnt and Notch pathways). MM CSCs are also supposed to be responsible for drug resistance that happens in three steps from the initial cancer cell homing microenvironment-mediated to development of microenvironment-independent drug resistance. In this review, we will underline all these aspects of MM CSCs.
