Reorganization of 3D genome architecture provides insights into pathogenesis of early fatty liver disease in laying hens

Background Fatty liver disease causes huge economic losses in the poultry industry due to its high occurrence and lethality ***-dimensional(3D)chromatin architecture takes part in disease processing by regulating tran-scriptional *** study is carried out to investigate the alterations of hepatic 3D genome and H3K27ac profiling in early fatty liver(FLS)and reveal their effect on hepatic transcriptional reprogramming in laying *** Results show that FLS model is constructed with obvious phenotypes including hepatic visible lipid deposi-tion as well as higher total triglyceride and cholesterol in serum.A/B compartment switching,topologically associat-ing domain(TAD)and chromatin loop changes are identified by high-throughput/resolution chromosome conforma-tion capture(HiC)*** genes of these alternations in hepatic 3D genome organization significantly enrich pathways related to lipid metabolism and hepatic damage.H3K27ac differential peaks and differential expres-sion genes(DEGs)identified through RNA-seq analysis are also enriched in these ***,certain DEGs are found to correspond with changes in 3D chromatin structure and H3K27ac binding in their *** motif analysis reveals that candidate transcription factors are implicated in regulating transcriptional ***,disturbed folate metabolism is observed,as evidenced by lower folate levels and altered enzyme *** Our findings establish a link between transcriptional reprogramming changes and 3D chromatin struc-ture variations during early FLS formation,which provides candidate transcription factors and folate as targets for FLS prevention or treatment.

Risk assessment of oil and gas investment environment in countries along the Belt and Road Initiative

With the implementation of the Belt and Road Initiative, China is deepening its cooperation in oil and gas resources with countries along the Initiative. In order to better mitigate risks and enhance the safety of investments, it is of significant importance to research the oil and gas investment environment in these countries for China's overseas investment macro-layout. This paper proposes an indicator system including 27 indicators from 6 dimensions. On this basis, game theory models combined with global entropy method and analytic hierarchy process are applied to determine the combined weights, and the TOPSIS-GRA model is utilized to assess the risks of oil and gas investment in 76 countries along the Initiative from 2014 to 2021. Finally, the GM(1,1) model is employed to predict risk values for 2022-2025. In conclusion, oil and gas resources and political factors have the greatest impact on investment environment risk, and 12 countries with greater investment potential are selected through cluster analysis in conjunction with the predicted results. The research findings may provide scientific decisionmaking recommendations for the Chinese government and oil enterprises to strengthen oil and gas investment cooperation with countries along the Belt and Road Initiative.

Nitrous oxide(N_(2)O)is a long-lived greenhouse gas that mainly originates from agricultural *** and more studies have explored the sources,influencing factors and effective mitigation measures of N_(2)O in recent ***,the hierarchy of factors influencing N_(2)O emissions from agricultural soils at the global scale remains *** this study,we carry out correlation and structural equation modeling analysis on a global N_(2)O emission dataset to explore the hierarchy of influencing factors affecting N_(2)O emissions from the nitrogen(N)and non-N fertilized upland farming systems,in terms of climatic factors,soil properties,and agricultural *** results show that the average N_(2)O emission intensity in the N fertilized soils(17.83 g N ha^(-1)d^(-1))was significantly greater than that in the non-N fertilized soils(5.34 g N ha^(−1) d^(−1))(prent climatic zones,without fertilizer,the primary influence factors on soil N_(2)O emissions are soil physical properties in subtropical monsoon zone,whereas climatic factors are key in the temperate *** fertilizer,the primary influence factors for subtropical monsoon and temperate continental zones are soil physical properties,while agricultural measures are the main factors in the temperate monsoon *** enhanced agricultural practices,such as reduced N fertilizer rate combined with the addition of nitrification and urease inhibitors can potentially mitigate N_(2)O emissions by more than 60%in upland farming systems.

Light grazing alleviates aeolian erosion–deposition effects on microbial communities in a semi-arid grassland

Background Soil erosion affects the stability of terrestrial ecosystems and ecosystem services by directly or indirectly impacting the cycling of soil materials and energy and reducing the fertility of grassland soils. However, research on microbial adaptation to grazing and soil erosion is limited, particularly in relation to grassland ecosystem restora-tion. Here, we assess microbial communities subjected to simulated soil erosion and grazing in a semi-arid grassland of Inner Mongolia, *** No significant change was observed in soil variables. However, the structure of the soil microbial commu-nity underwent significant changes as a result of soil erosion and soil erosion plus grazing, leading to a significant increase in the relative abundance of Cyanobacteria(116.80% vs 116.38%). Wind erosion and deposition contributed to an increase in the network complexity of soil bacterial and fungal communities. However, much of this effect was alleviated by grazing. Simultaneously, aeolian processes and grazing regulate soil microbial community assembly, leading to inconsistent patterns of change in bacterial and fungal communities. Under wind erosion and deposi-tion, the relative contribution of deterministic processes(4.44% vs 31.11%) in bacterial communities increased, while the relative contribution of stochastic processes(2.23% vs 20%) in fungal communities reduced. Grazing resulted in a decrease in the relative contribution of deterministic processes(8.89%) in the bacterial community and an increase in the relative contribution of stochastic processes(8.89%) in the fungal *** This study presents a comprehensive investigation of the response of soil microbial communities to aeolian erosion–deposition and grazing in a semi-arid grassland. Ultra-high-temperature application of MXene: Stabilization of 2D Ti_(3)C_(2)T_(x) for cross-scale strengthening and toughening of 3D TiC

Transition metal carbide/nitride cores within MXenes make them considerably useful for ultra-high-temperature ***,extensive research on Ti_(3)C_(2)T_(x) MXene has revealed its tendency to undergo a phase transition to TiCy at temperatures above 800℃due to high activity of a superficial Ti atomic ***,spark plasma sintering of Ti_(3)C_(2)T_(x) and TiC is performed to prevent the Ti_(3)C_(2)T_(x) phase transition at temperatures up to 1900℃through the fabrication of composites at a pressure of 50 *** a focused ion beam scanning electron microscope to separate layered substances in the composites and examining selected area diffraction spots in a transmission electron microscope enabled identification of non-phase-transitioned ***-principles calculations based on density functional theory indicated the formation of strong chemical bonding interfaces between Ti_(3)C_(2)T_(x) and TiC,which imposed a stability constraint on the Ti atomic layer at the Ti_(3)C_(2)T_(x) *** performance tests,such as three-point bending and fracture toughness analysis,demonstrated that the addition of Ti_(3)C_(2)T_(x) can effectively improve the cross-scale strengthening and toughening of the TiC matrix,providing a new path for designing and developing two-dimensional(2D)carbides cross-scale-enhanced three-dimensional(3D)carbides with the same elements relying on a wide variety of MXenes.

We present an innovative approach for the simultaneous agile manipulation of high-refractive-index(HRI) and low-refractive-index(LRI) particles. Our method involves introducing a dual-curvilinear optical vortex beam(DC-OVB) generated by superimposing a pair of curved beams: HRI and LRI particles are controlled by the bright curve and the dark channel between the two curves, respectively. The proposed DC-OVB provides customizable motion paths and velocities for both LRI and HRI particles. Each curve of the DC-OVB can support a distinct orbital flow density(OFD), enabling the application of torques to HRI and LRI particles, guiding them to orbit along specified trajectories and prompting them to execute various curvilinear motions simultaneously,including curvilinear movement, revolution, and rotation.

The Effect and Mechanism of Fructus lycii on Improvement of Exercise Fatigue Using a Network Pharmacological Approach with in vitro Experimental Verification

Objective This study aimed to investigate the effect and underlying mechanism of Fructus lycii in improving exercise *** A network pharmacological approach was used to explore potential mechanisms of action of Fructus *** muscle C2C12 cells and immunofluorescence were employed to verify the effect and mechanism of the representative components in Fructus lycii predicted by network pharmacological *** Six potential active components,namely quercetin,β-sitosterol,stigmasterol,7-Omethylluteolin-6-C-beta-glucoside_qt,atropine,and glycitein,were identified to have potency in improving exercise fatigue via multiple pathways,such as the PI3K-Akt,neuroactive ligand-receptor interaction,IL-17,TNF,and MAPK signaling *** immunofluorescence results indicated that quercetin,a significant active component in Fructus lycii,increased the mean staining area of 2-NBDG,TMRM,and MitoTracker,and decreased the area of CellRox compared to the ***,the protein expression levels of p-38 MAPK,p-MAPK,p-JNK,p-PI3K,and p-AKT markedly increased after quercetin *** Fructus lycii might alleviate exercise fatigue through multiple components and *** these,quercetin appears to improve exercise fatigue by enhancing energy metabolism and reducing oxidative *** PI3K-AKT and MAPK signaling pathways also appear to play a role in this process.
