Caffeic acid phenethyl ester protects against oxidative stress and dampens inflammation via heme oxygenase 1

Periodontal disease is associated with chronic oxidative stress and inflammation. Caffeic acid phenethyl ester(CAPE), which is a potent inducer of heme oxygenase 1(HO1), is a central active component of propolis, and the application of propolis improves periodontal status in diabetic patients. Here, primary murine macrophages were exposed to CAPE. Target gene expression was assessed by whole-genome microarray, RT-PCR and Western blotting. The antioxidative and anti-inflammatory activities of CAPE were examined by exposure of the cells to hydrogen peroxide, saliva and periodontal pathogens. The involvement of HO1 was investigated with the HO1 inhibitor tin protoporphyrin(SnPP) and knockout mice for Nrf2, which is a transcription factor for detoxifying enzymes. CAPE increased HO1 and other heat shock proteins in murine macrophages. A p38 MAPK inhibitor and Nrf2 knockout attenuated CAPE-induced HO1 expression in macrophages. CAPE exerted strong antioxidative activity. Additionally, CAPE reduced the inflammatory response to saliva and periodontal pathogens. Blocking HO1 decreased the antioxidative activity and attenuated the anti-inflammatory activity of CAPE. In conclusion, CAPE exerted its antioxidative effects through the Nrf2-mediated HO1 pathway and its anti-inflammatory effects through NF-κB inhibition. However, preclinical models evaluating the use of CAPE in periodontal inflammation are necessary in future studies.

Root Exudation of Organic Acids of Herbaceous Pioneer Plants and Their Growth in Sterile and Non-Sterile Nutrient-Poor,Sandy Soils from Post-Mining Sites

Nutrient-poor, sandy soils form the prevailing substrate at post-mining sites of the Lusatian region(Brandenburg, Germany) and present a challenge for vegetation development. We studied the organic acid quantity and composition of three commonly occurring pioneer plant species, the legumes Lotus corniculatus L. and Trifolium arvense L. and the grass Calamagrostis epigeios(L.) Roth, to determine if plant growth and exudation differed with(non-sterilized soil) and without(sterilized soil) an indigenous soil microbial community. We investigated whether organic acids were found in the rhizosphere and surrounding soil and whether this influenced nutrient mobilization. This study consists of linked field investigations and a greenhouse experiment. Plants were grown in the greenhouse in either sterilized or non-sterilized sandy soil from a reclamation site in the Lusatian mining landscape(Welzow Su¨d, East Germany). After seven months, the plant biomass, root morphology, organic acids, and water-soluble nutrients and root colonization with arbuscular mycorrhizal fungi(AMF) and dark septate endophytes(DSE) were analyzed. Roots of all three plants in the field and greenhouse experiments were highly colonized with AMF. Calamagrostis epigeios and T. arvense had a significantly higher colonization frequency with DSE than L. corniculatus. The quantity and composition of organic acids strongly differed among plant species, with the highest number of organic acids found for L. corniculatus and lowest for C. epigeios. The quantity of organic acids was greatly reduced in all plants under sterilized soil conditions. However, the composition of organic acids and plant growth in sterilized soil were reduced for both legumes, but not for C. epigeios, which had a higher biomass under sterilized conditions. 目的　通过观察抗高血糖药物二甲双胍对肝糖异生中的限速酶磷酸烯醇丙酮酸羧激酶 (PEPCK)基因表达的调节以及胰岛素信号转导抑制剂的影响 ,以探讨二甲双胍在肝细胞中作用的分子机制。方法　将小鼠肝癌细胞系 (H4IIE)细胞用不同浓度的二甲双胍温育 16小时 ,加入不同的调节因子如胰岛素、环磷酸腺苷 (cAMP)、地塞米松、胰岛素信号转导抑制剂等。PEPCK的mRNA水平通过Northern杂交分析。结果　治疗浓度 0 1mmol/L二甲双胍显著降低了基础PEPCK的mRNA水平 ,并通过与胰岛素的联合作用抑制了cAMP/地塞米松刺激的PEPCK的基因表达。胰岛素信号转导抑制剂如磷酯酰肌醇 3激酶 (PI3K)抑制剂渥蔓青霉素 (wortmannin)和有丝分裂原激活蛋白激酶 (MAPK)抑制剂UO12 6对二甲双胍的作用无显著影响 ,但wortmannin显著阻止了胰岛素对PEPCK基因表达的调节作用。结论　二甲双胍可通过独立的非胰岛素信号转导途径或是与胰岛素的联合作用抑制PEPCK的基因表达 ,表明二甲双胍降低肝糖异生的分子机制可能与PEPCK基因表达的抑制有关。

Sclerostin is a Wnt signalling antagonist that controls bone metabolism. Sclerostin is expressed by osteocytes and cementocytes; however, its role in the formation of dental structures remains unclear. Here, we analysed the mandibles of sclerostin knockout mice to determine the influence of sclerostin on dental structures and dimensions using histomorphometry and micro-computed tomography （μCT） imaging, μCT and histomorphometric analyses were performed on the first lower molar and its surrounding structures in mice lacking a functional sclerostin gene and in wild-type controls, pCT on six animals in each group revealed that the dimension of the basal bone as well as the coronal and apical part of alveolar part increased in the sclerostin knockout mice. No significant differences were observed for the tooth and pulp chamber volume. Descriptive histomorphometric analyses of four wild-type and three sclerostin knockout mice demonstrated an increased width of the cementum and a concomitant moderate decrease in the periodontal space width. Taken together, these results suggest that the lack of sclerostin mainly alters the bone and cementum phenotypes rather than producing abnormalities in tooth structures such as dentin.

Contraindications for video capsule endoscopy

Video capsule endoscopy(VCE) has been applied in the last 15 years in an increasing field of applications. Although many contraindications have been put into perspective, some precautions still have to be considered. Known stenosis of the gastrointestinal tract is a clear contraindication for VCE unless surgery is already scheduled or at least has been considered as an optional treatment modality. In patients with a higher incidence of stenosis, as in an established diagnosis of Crohn's disease, clinical signs of obstruction, prior radiation or surgical small bowel resection, a preceding test with the self-dissolving patency capsule can override this contraindication. Endoscopic placement of the capsule should be considered in patients with swallowing disorders to avoid aspiration. Esophageal or gastric motility disorders may require endoscopic capsule transport or application of prokinetics if the real-time viewer proofs delayed transit. In pregnant women, VCE should be restricted to urgent cases where diagnosis cannot be postponed after delivery, as data on safety are missing. There is theoretical and clinical evidence that patients with implanted cardiac devices such as a pacemaker, cardioverters or left heart assist devices, can safely undergo VCE in spite of still existing contraindication by manufacturers. Children from the age of 2 years have safely undergone VCE. Although video capsules are not proven safe with magnetic resonance imaging(MRI), first single cases of patients incidentally undergoing MRI with an incorporated capsule have been reported, showing susceptibility artifacts but no signs of clinical harm.

Comparison of a Manual and an Automated Tracking Method for Tibetan Plateau Vortices

Tibetan Plateau vortices （TPVs） are mesoscale cyclones originating over the Tibetan Plateau （TP） dnring the extended summer season （April-September）. Most TPVs stay on the TP, but a small number can move off the TP to the east. TPVs are known to be one of the main precipitation-bearing systems on the TP and moving-off TPVs have been associated with heavy precipitation and flooding downstream of the TP （e.g., in Sichuan province or over the Yangtze River Valley）. Identifying and tracking TPVs is difficult because of their comparatively small horizontal extent （400-800 kin） and the limited availability of soundings over the TP, which in turn constitutes a challenge for short-term predictions of TPV-related impacts and for the climatological study of TPVs. In this study, （i） manual tracking （MT） results using radiosonde data from a network over and downstream of the TP are compared with （ii） results obtained by an automated tracking （AT） algorithm applied to ERA-Interim data. Ten MT-TPV cases are selected based on method （i） and matched to and compared with the corresponding AT-TPVs identified with method （ii）. Conversely, ten AT-TPVs are selected and compared with the corresponding MT-TPVs. In general, the comparison shows good results in cases where the underlying data are in good agreement, but considerable differences are also seen in some cases and explained in terms of differences in the tracking methods, data availability/coverage and disagreement between sounding and ERA-Interim data. Recommendations are given for future efforts in TPV detection and tracking, including in an operational weather forecasting context.

Hydrochemistry,Origin and Evolution of Sedimentary Sub- surface Fluids:*** diagenetic pore- water evolution in low-sedimentation rate offshore basins

Pore water studies enable (1) the detection of diagenetic reactions actively occurring in the sediment atthe time of sampling,(2) the distinction between principal modes of solute transport, i.e., between advection(convection) and diffusion, and (3) the assessment of mineral-solution equilibria, Pore waters are, therefore,preferred diagnostic objects in the study of diagenesis, particularly early diagenesis. The single most important factor for pore-fluid evolution in modern offshore basins is sedimentationrate which is closely correlated with organtic matter content of the sediment. Organic matter represents themost reactive sediment constituent which, through bacterial decomposition, provides some of the mainsolutes involved in early diagenetic mineralization reactions. On the basis of sedimentation rate and organicmatter content, it is convenient to distinguish two end-members of basins (environments) with respect toearly diagenesis: Ⅰ. Low to in termediate-sedimentation rate basins with convection or diffusion-controlledpore-water profiles and Ⅱ. high-sedimentation rate basins with reaction-controlled pore-water profiles. The first group of basins or environments, which is the subject of this paper, is typically represented bypelagic sediments. Three principally different trends of pore-water evolution occur: 1. convection-controlledprofiles with no pronounced vertical gradients for the dissolved species; 2. diffusion-controlled profiles withvertical gradients but linear correlations between major ions, especially Ca and Mg, and 3. profiles withgradients but no linear correlation between the major ions. The later are transitional to the trends seen inhigh-sedimentation rate basins. With respect to redox-potentials, the diagenetic environments of low-sedimentation rate basins are generally oxic to suboxic.
