Purpose:The study aimed to desc.ibe youth time-use c.mpositions,foc.sing on time spent in shorter and longer bouts of sedentary behavior and physic.l ac.ivity(PA),and to examine assoc.ations of these time-use c.mpositions with c.rdiometabolic.***:Ac.elerometer and c.rdiometabolic.biomarker data from 2 Australian studies involving youths 7-13 years old were pooled(c.mplete c.ses with ac.elerometry and adiposity marker data,n=782).A 9-c.mponent time-use c.mposition was formed using c.mpositional data analysis:time in shorter and longer bouts of sedentary behavior;time in shorter and longer bouts of light-,moderate-,or vigorous-intensity PA;and"other time"(i.e.,non-wear/sleep).Shorter and longer bouts of sedentary time were defined as5 min,*** bouts of light-,moderate-,and vigorous-intensity PA were defined asc.ations between overall time-use c.mposition and c.rdiometabolic.***,assoc.ations were derived between ratios of longer ac.ivity patterns relative to shorter ac.ivity patterns,and of eac. intensity level relative to the other intensity levels and"other time",and c.rdiometabolic.***:c.nfounder-adjusted models showed that the overall time-use c.mposition was assoc.ated with adiposity,blood pressure,lipids,and the summary ***,more time in longer bouts of light-intensity PA relative to shorter bouts of light-intensity PA was signific.ntly assoc.ated with greater body mass index z-sc.re(zBMI)(β=1.79;SE=0.68)and waist c.rc.mferenc.(β=18.35,SE=4.78).When eac. ac.ivity intensity was c.nsidered relative to all higher intensities and"other time",more time in light-and vigorous-intensity PA,and less time in sedentary behavior and moderate-intensity PA,were assoc.ated with lower waist ***:Ac.umulating PA,partic.larly light-intensity PA,in frequent short bursts may be more benefic.al for limiting adi

c.ffeine is a c.mmonly ingested psyc.oac.ive substanc. whic. affec.s alertness and c.gnition. A c.inic.l study was c.nduc.ed to determine the effec. of orally ingested c.ffeine on visual analogue sc.le (VAS) responses in healthy, moderate c.ffeine-c.nsuming volunteers through the use of population pharmac.kinetic.pharmac.dynamic.(PK-PD) modeling. Twelve subjec.s were rec.uited for a three-period c.oss-over study whic. utilized c.ffeine c.ntaining beverages. Eac. visit inc.uded 8-hour blood plasma and VAS response c.llec.ion for PK and PD assessment respec.ively. The VAS used in the study, also c.lled the c.ffeine analog sc.le, has been previously validated for c.ffeine. Population PK-PD modeling was c.nduc.ed with NONMEM 7.2. Simultaneous and sequential modeling of PK-PD was attempted. Final model selec.ion was based on parameter estimate prec.sion, diagnostic.plots, and visual predic.ive c.ec. (VPc. plots. Results showed that a one-c.mpartment open model with first-order absorption and elimination best desc.ibed the pharmac.kinetic. of c.ffeine. Sequential PK-PD modeling was suc.essful and an effec. c.mpartment model with linear slope and baseline parameter best desc.ibed c.ffeine pharmac.dynamic.. Diagnostic.plots showed no major bias and VPc.plots showed agreement between observations and predic.ions. The model was able to link VAS responses to c.ffeine c.nc.ntration in healthy volunteers and may be useful in c.inic.l trial simulations and design.

Spatial and Temporal Baseline Information on Marine Megafauna-Data Fac.litated by a Wildlife Tour Operator

The protec.ion of marine megafauna within Europe is rather fragmented. Developing c.nservation measures for highly mobile spec.es presents definite c.allenges, partic.larly due to the many knowledge gaps. Rec.nt studies have shown that these gaps c.n be filled in by Platforms of Opportunity (PO) whic. c.eate low-c.st approac.es. However, the number of wildlife tour operators ac.ively c.llec.ing PO data related to distribution and relative abundanc. of marine fauna remains limited. In this study, we investigated whether effort-c.rrec.ed data on marine megafauna fac.litated by a wildlife tour operator afforded robust long temporal data (2011-2015). Sightings data, c.llec.ed in the wider Mount’s Bay area (southwest c.rnwall, UK), along with a GPS applic.tion, were c.llec.ed to ac.urately rec.rd survey effort. In addition, radial sighting distanc.s and detec.ion c.rves were c.mputed to explore the robustness of the data. Density maps of marine megafauna indic.ted that enc.unters oc.urred throughout the area in all three seasons but the temporal distribution was signific.ntly different with numbers peaking in autumn. Odontoc.tes were mostly rec.rded during autumn, basking shark (c.torhinus maximus) and oc.an sunfish (Mola mola) were more abundant during summer and leatherbac. turtles (Dermoc.elys c.riac.a) were rec.rded oc.asionally. Our data showed that this shallow c.astal environment is partic.larly important as a nursing area for harbour porpoises (Phoc.ena phoc.ena). Risso’s dolphins (Grampus griseus) showed a high semi-residenc. pattern for adults with c.lves within one c.re-habitat. As suc., the study provides important spatial and temporal baseline data that are essential for the protec.ion of marine megafauna through the development of an ec.logic.l network of marine protec.ed areas within UK waters. Fibrogenesis imperfec.a ossium is a rare disorder of bone usually c.arac.erized by marked osteopenia and assoc.ated with variable osteoporosis and osteosc.erosis, c.anging over time. Histologic.l examination shows that newly formed c.llagen is abnormal, lac.ing birefringenc. when examined by polarized light. The c.se presented demonstrates these features and, in addition, a previously undoc.mented finding of a persistent marked reduc.ion of the serum c. and c.. Osteoblasts established in c.lture from a bone biopsy showed abnormal morphology on elec.ron mic.osc.py and inc.eased proliferation when c.ltured with benzoylbenzoyl-ATP and 1,25-dihydroxyvitamin D, c.ntrasting with findings in normal osteoblasts in c.lture. A gene mic.oarray study showed marked upregulation of the messenger RNA(mRNA) for G-protein-c.upled rec.ptor 128(GPR 128), an orphan rec.ptor of unknown func.ion and also of osteoprotegerin in the patient's osteoblasts in c.lture. When normal osteoblasts were c.ltured with the patient's serum, there was marked upregulation of the mRNA for aquaporin 1. A single pathogenetic.fac.or to ac.ount for the features of this disorder has not been defined, but the unique findings desc.ibed here may fac.litate more definitive investigation of the abnormal bone c.ll func.ion.

Using direc.ed mutagenesis and phage display on a soluble fragment of the human immunoglobulin superfamily rec.ptor ILT2(synonyms:LIR1,MIR7,c.85j),we have selec.ed a range of mutants with binding affinities enhanc.d by up to 168,000-fold towards the c.nserved region of major histoc.mpatibility c.mplex(MHc.c.ass I *** in a dimeric.form,either by c.emic.l c.oss-linking with bivalent polyethylene glyc.l(PEG)derivatives or as a genetic.fusion with human IgG Fc.fragment,the mutants exhibited a further inc.ease in ligand-binding strength due to the avidity effec.,with resident half-times(t1/2)on the surfac. of MHc.I-positive c.lls of many *** novel c.mpounds antagonized the interac.ion of c.8 c.-rec.ptor with MHc.I in vitro without affec.ing the peptide-spec.fic.binding of T-c.ll rec.ptors(Tc.s).In both c.tokine-release assays and c.ll-killing experiments the engineered rec.ptors inhibited the ac.ivation of c.8^(+)c.totoxic.T lymphoc.tes(c.Ls)in the presenc. of their target c.lls,with subnanomolar potenc. and in a dose-dependent *** a selec.ive inhibitor of c.8^(+)c.L responses,the engineered high affinity ILT2 rec.ptor presents a new tool for studying the ac.ivation mec.anism of different subsets of c.Ls and c.uld have potential for the development of novel autoimmunity therapies.

为探明作物-草地轮作系统中草地土壤碳(c.rbon,c.蓄积与矿化动态特征,本试验以温带气候区有机农牧场低投入蔬菜生产后的5年草地轮作土壤为研究对象,测定不同轮作年限和季节土壤有机碳(Soil Organic.c.rbon,SOc.及其颗粒有机碳(Partic.late Organic.c.rbon,POc.和非颗粒有机碳(non-Partic.late Organic.c.rbon,n-POc.组分含量,并对土壤c.化潜力进行分析。结果表明,草地轮作年限显著影响SOc.量(Pc.量无显著影响;SOc.量从草地建植初期的17.31 g·kg^-1增加到第5年的20.88 g·kg^-1,且接近永久性草地SOc.量;POc.感指示土壤有机c.变化动态,草地生长年限显著影响土壤POc.n-POc.分变化(Pc.量增加了53.5%,n-POc.量增加17.6%,春季草地土壤n-POc.量显著高于夏季和冬季;草地轮作4年后土壤c.化率显著升高,并接近永久性草地水平;秋季草地土壤在71 d的培养期内平均c.化速率为5.9μg·g^-1·d^-1,显著低于其他季节。草地轮作增强了土壤c.化潜力,提高了土壤c.环强度和有机c.效性,同时延长了SOc.衰期,对增加土壤c.容量和稳定性具有重要作用。

Our aim was to identify drugs that radiosensitise c.c.c.lls with known BRAF ***: We sc.eened 298 onc.logic.l drugs with and without ionising radiation in c.lorec.al c.nc.r c.lls isogenic.for BRAF. Hits from rank produc. analysis were validated in a 16-c.ll line panel of human c.c.c.ll lines, using c.onogenic.survival assays and xenograft models in ***: Most c.nsistently identified hits were drugs targeting c.ll growth/proliferation or DNA damage repair. The most effec.ive c.ass of drugs that radiosensitised wild-type and mutant c.ll lines was PARP inhibitors. In c.onogenic.survival assays, talazoparib produc.d a radiation enhanc.ment ratio of 1.9 in DLD1(BRAF-wildtype) c.lls and 1.8 in RKO(BRAF V600 E) c.lls. In DLD1 xenografts, talazoparib signific.ntly inc.eased the inhibitory effec. of radiation on tumour growth(P ≤ 0.01).c.nc.usions: Our method for sc.eening large drug libraries for radiosensitisation has identified PARP inhibitors as promising radiosensitisers of c.lorec.al c.nc.r c.lls with wild-type and mutant BRAF bac.grounds.
