AIM： To characte.ize.the.mitochondrial dysfunction in e.pe.ime.tal cirrhosis and to study whe.he. insulin-like.growth factor-Ⅰ （IGF-Ⅰ ） the.apy （4 wk） is able.to induce.be.e.icial e.fe.ts on damage. mitochondria le.ding to ce.lular prote.tion. Me.HODS： Wistar rats we.e.divide. into thre. groups： Control group, untre.te. cirrhotic rats and cirrhotic rats tre.te. with IGF-Ⅰ tre.tme.t （2 μg/1O0 g bw/d）. Mitochondrial function was analyze. by flow cytome.ry in isolate. he.atic mitochondria, caspase.3 activation was asse.se. by We.te.n blot and apoptosis by TUNe. in the.thre. e.pe.me.tal groups. Re.ULTS： Untre.te. cirrhotic rats showe. a mitochondrial dysfunction characte.ize. by a significant re.uction of mitochondrial me.brane.pote.tial （in status 4 and 3）; an incre.se.of intramitochondrial re.ctive.oxige. spe.ie. （ROS） ge.e.ation and a significant re.uction of ATPase.activity. IGF-Ⅰ the.apy normalize. mitochondrial function by incre.sing the.me.brane.pote.tial and ATPase.activity and re.ucing the.intramitochondrial fre. radical production. Activity of the.e.e.tron transport comple.e. Ⅰ and Ⅲ was incre.se. in both cirrhotic groups. In addition, untre.te. cirrhotic rats showe. an incre.se.of caspase.3 activation and apoptosis. IGF- Ⅰ the.apy re.uce. the.e.pre.sion of the.active.pe.tide.of caspase.3 and re.ulte. in re.uce. apoptosis. CONCLUSION： The.e.re.ults show that IGF- Ⅰ e.e.ts a mitochondrial prote.tion in e.pe.ime.tal cirrhosis le.ding to re.uce. apoptosis and incre.se. ATP production.

De.IGN: Obse.vational study inve.tigating mRNA transcript e.pre.sion of le.tin and the.le.tin re.e.tor from human subcutane.us fat spe.ime.s, as we.l as le.tin se.um le.e.s, both in re.ation to the.te.toste.one.and se. hormone. during the.MC in obe.e.and non-obe.e.wome.. Me.HODS: Nine.e.n wome., thirte.n with a body mass inde. (BMI) > 25 Kg/m2 and 6 with BMI 2 we.e.include.. RT-PCR was use. to de.e.t L-mRNA transcripts of le.tin and its re.e.tor in sample. from 28 fat biopsie. take. from se.e. volunte.rs (20 from OW and 8 from NOW).
