Nephroprotective activity of Solanum xanthocarpum fruit extract against gentamicin-induced nephrotoxicity and renal dysfunction in experimental rodents

Objective:To evaluate nephroprotective potential of Solarium xanthocarpum(***) fruit extract(SXE) against gentamicin(GM) induced nephrotoxicity) and renal dysfunction. Methods:Twenty-four Wistar rats were divided into four groups(n=6).Control rats that received normal saline(i.p.) and 0.5%carboxymethyl cellulose(p.o.) per day lor 8 *** was induced in rats by intraperitoneal administration of GM(100 mg/kg/d for 8 d) and were treated with SXE(200 and 400 mg/kg/d(p.o.) for 8 d).Plasma and urine urea and creatinine,kidney weight,urine output,blood urea nitrogen,renal enzymatic and non-enzymatic antioxidants and lipid peroxidation was evaluated along with histopathological investigation in various experimental groupsof ***:It was observed that the GM treatment induced significant elevation(P<0.001) in plasma and urine urea,creatinine,kidney weight,blood urea nitrogen, renal lipid peroxidation along with significant decrement(P<0.001) in urine output,renal enzymatic and non-enzymatic *** 200 and 400 mg/kg treatment to GM treated rats recorded significant decrement(up to P<0.001) in plasma and mine urea and creatinine, renal lipid peroxidation along with significanl increment(up to P<0.001) in renal enzymatic and non-enzvmatic *** obsenatioiis of kidney tissues too correlated with the biochemical ***:These finding powerfully supports that S,xanthocarpum fruit extract acts in the kidney as a potent scavenger of free radicals to prevent the toxic effects of GM both in the biochemical and histopathological parameters and thus validates its elhnomedicinal use.

Heat load of a GaAs photocathode in an SRF electron gun

A great deal of effort has been made over the last decades to develop a better polarized electron source for high energy physics. Several laboratories operate DC guns with a gallium arsenide photocathode, which yield a highly polarized electron beam. However, the beam’s emittance might well be improved by using a superconducting radio frequency （SRF） electron gun, which delivers beams of a higher brightness than that from DC guns because the field gradient at the cathode is higher. SRF guns with metal and CsTe cathodes have been tested successfully. To produce polarized electrons, a Gallium-Arsenide photo-cathode must be used： an experiment to do so in a superconducting RF gun is under way at BNL. Since a bulk gallium arsenide （GaAs） photocathode is normal conducting, a problem arises from the heat load stemming from the cathode. We present our measurements of the electrical resistance of GaAs at cryogenic temperatures, a prediction of the heat load and verification by measuring the quality factor of the gun with and without the cathode at 2 K. We simulate heat generation and flow from the GaAs cathode using the ANSYS program. By following the findings with the heat load model, we designed and fabricated a new cathode holder （plug） to decrease the heat load from GaAs.

Conclusions:The results of this study strongly indicate the protective effect of SXE against acute liver injun which may he attributed to its hepatoprotective activity,and there by scientifically support its traditional use.

Toxic Potential of Palytoxin

This review briefly describes the origin,chemistry,molecular mechanism of action,pharmacology,toxicology,and ecotoxicology of palytoxin and its analogues. Palytoxin and its analogues are produced by marine dinoflagellates. Palytoxin is also produced by Zoanthids（i.e. Palythoa）,and Cyanobacteria（Trichodesmium）. Palytoxin is a very large,non-proteinaceous molecule with a complex chemical structure having both lipophilic and hydrophilic moieties. Palytoxin is one of the most potent marine toxins with an LD50 of 150 ng/kg body weight in mice exposed intravenously. Pharmacological and electrophysiological studies have demonstrated that palytoxin acts as a hemolysin and alters the function of excitable cells through multiple mechanisms of action. Palytoxin selectively binds to Na＋/K＋-ATPase with a Kd of 20 p M and transforms the pump into a channel permeable to monovalent cations with a single-channel conductance of 10 p S. This mechanism of action could have multiple effects on cells. Evaluation of palytoxin toxicity using various animal models revealed that palytoxin is an extremely potent neurotoxin following an intravenous,intraperitoneal,intramuscular,subcutaneous or intratracheal route of exposure. Palytoxin also causes non-lethal,yet serious toxic effects following dermal or ocular exposure. Most incidents of palytoxin poisoning have manifested after oral intake of contaminated seafood. Poisonings in humans have also been noted after inhalation,cutaneous/systemic exposures with direct contact of aerosolized seawater during Ostreopsis blooms and/or through maintaining aquaria containing Cnidarian zoanthids. Palytoxin has a strong potential for toxicity in humans and animals,and currently this toxin is of great concern worldwide.

Objective:To assess the hepatoprotective effect of Solanum xanthocarpum(S. xanthocarpum) fruit extract against antitubercular drug-induced liver toxicity in experimental ***:Ethanolic(50%) fruit extract ofS. xanthocarpum(100, 200 and 400 mg/kg bw) was administered daily for 35 days in experimental animals. Liver toxicity was induced by combination of three antitubercular drugs [isoniazid(I) 7.5 mg/kg, rifampicin(R) 10 mg/kg and pyrazinamide(P) 35 mg/kg] given orally as suspension for 35 days in rats. The hepatoprotective activity was assessed using various biochemical parameters like aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphatise(ALP), total bilirubin(TBL), albumin(ALB), total protein(TP), lactate dehydroginase(LDH), and serum cholesterol(CHL). Meanwhile,in vivoantioxidant activities as lipid peroxidation(LPO), reduced glutathione(GSH), superoxide dismutase(SOD) and catalase(CAT) were measured in rat liver homogenate. The biochemical observations were supplemented by histopathological ***:The results demonstrated that treatment ***(P<0.05-P<0.001) and dose-dependently prevented drug induced increase in serum levels of hepatic enzymes. Furthermore,S. xanthocarpumsignificantly(up toP<0.001) reduced the LPO in the liver tissue and restored activities of defence antioxidant enzymes GSH, SOD and CAT towards normal levels. Histopathology of the liver tissue showed that S. xanthocarpumattenuated the hepatocellular necrosis and led to reduction in inflammatory cells ***:The results of this study strongly indicate the protective effect of S. xanthocarpumagainst liver injury which may be attributed to its hepatoprotective activity, and thereby scientifically support its traditional use.

Objective:To evaluate the anti-inflammatory effect of Nardostachys jatamansi(***)rhizome against acute,subacute and chronic models of inflammation in experimental ***:*** rhizome extract(150 and 300 mg/kg,p.o.)and the reference drugs phenylbutazone(100 mg/kg,p.o.)and acetylsalicylic acid(300 mg/kg,p.o.)were evaluated using models for inflammation(autacoids induced hind paw oedema,formaldehyde induced hind paw oedema,carrageenin-induced paw oedema,cotton pellet granuloma and subcutaneous air pouch model).Results:In acute inflammation as produced by carrageenin 29.06%and 55.81%,by histamine 25.0%and 39.28%,by 5-hydroxytryptamine 21.37%and 36.95%and by prostaglandin E2-induced hind paw oedema 31.03%and 44.82%protection was *** in subacute anti-inflammatory models using formaldehyde-induced hind paw oedema(after 1.5 h)13.88%and 33.33%and in chronic anti-inflammatory model using cotton pellet granuloma 7.4%and 17.58%protection from inflammation was *** rhizome extract also inhibited the inflammatory mediators(nitric oxide by 12.81%and 38.41%,by prostaglandin E212.58%and 47.82%while by TNF-α13.51%and 41.89%)produced in the ***:The results of this study strongly indicate the protective effect of *** rhizome extract against acute,subacute and chronic models of inflammation,which may be attributed to its anti-inflammatory potential.

Hematological, antioxidant and protective performance of Usnea longissima on chemical induced hepatotoxicity in experimental animals

Objective: To investigated the hematological, antioxidant and protective performance of Usnea longissima (U. longissima) on CCl4 induced hepatotoxicity in experimental animals. Methods: Hepatotoxicity was induced by CCl4 (1 mL/kg body weigt 1:1 CCl4 i.p.), ethanolic U. longissima extracts at a doses (200 and 400 mg/kg body weigt) were administered to and compared with Silymarin (25 mg/kg body weigt) and hematological, antioxidant and enzymatic, non-enzymatic parameters were assessed through the liver functions test. All the observation was also supplemented with histopathological examination of liver sections. Results: Phytochemical investigation showed that ethanolic extract contains poly phenolic compounds tannins, flavonoids, alkaloids and saponins and acute toxicity study shows that ethanolic extract was safe up to 2000 mg/kg body weight. The toxicant induced a rise in the plasma enzyme levels of ALT, AST, ALP and total bilirubin level. This increased level was significantly decreased by the extract at 400 mg/kg body weight than 200 mg/kg body weight. The animals were prevented (partly or fully) which was showed in the histopathological changes using ethonolic U. longissima extract. Conclusions: The outcome of this study reveals that, there is a powerful antioxidant and hepatoprotective activity of U. longissima. It is believed that the present constituents are responsible for courting the hepatic disease and alternative components have the power to act as free radical scavenging properties.
