In this study, we explore the potentia. of spa.ia. meta.olomics for eva.ua.ing meta.olic phenotypes a.d thera.y outcomes during the loca. delivery of the a.tica.cer drug, oxa.ipla.in, to the lung. 12 ma.e Yorkshire pigs underwent a.3 h left lung in vivo lung perfusion (IVLP) with va.ious doses of oxa.ipla.in (7.5, 10, 20, 40, a.d 80 mg/L), which were a.ministered to the perfusion circuit reservoir a. a.bolus. Biocompa.ible solid-pha.e microextra.tion (SPME) microprobes were combined with globa. meta.olite profiling to obta.n spa.iotempora. informa.ion a.out the a.tivity of the drug, determine toxic doses tha. exceed thera.eutic effica.y, a.d conduct a.mecha.istic explora.ion of a.socia.ed lung injury. Mild a.d subclinica. lung injury wa. observed a. 40 mg/L of oxa.ipla.in, a.d significa.t compromise of the hemodyna.ic lung function wa. found a. 80 mg/L. This result wa. a.socia.ed with ma.sive a.tera.ions in meta.olic pa.terns of lung tissue a.d perfusa.e, resulting in a.tota. of 139 discrimina.t compounds. Uncontrolled infla.ma.ory response, a.norma.ities in energy meta.olism, a.d mitochondria. dysfunction next to a.celera.ed kynurenine a.d a.dosterone production were recognized a. distinct fea.ures of dysregula.ed meta.olipidome.
