In this study,we identified the most deleterious nsSNP in rB1 gene through structural and functional properties of its protein (prB) and investigated its binding affinity with *** of 956 SNPs,we investigated 12 nsSNPs in coding region in which three of them (SNPids rs3092895,rs3092903 and rs3092905) are commonly found to be damaged by I-Mutant 2.0,SIFT and PolyPhen *** this effort,we modeled the mutant prB proteins based on these deleterious *** a comparison of total energy,stabilizing residues and rMSD of these three mutant proteins with native prB protein,we identified that the major mutation is from Glutamic acid to Glycine at the residue position of 746 of ***,we compared the binding efficiency of both native and mutant prB (E746G) with *** found that mutant prB has less binding affinity with E2F-2 as compared to native *** is due to sixteen hydrogen bonding and two salt bridges that exist between native type and E2F-2,whereas mutant type makes only thirteen hydrogen bonds and one salt bridge with *** on our investigation,we propose that the SNP with an id rs3092905 could be the most deleterious nsSNP in rB1 gene causing retinoblastoma.

Influence of Quantal and Statistical Fluctuations on Phase Transitions in Finite Nuclei

Investigations on thermal evolution of pairing-phase transition and shape-phase transition in light nuclei are made as a function of pair gap, deformation, temperature and angular momentum using a finite temperature statistical approach with main emphasis to fluctuations. The occurrence of a peak structure in the specific heat predicted as signals of the pairing-phase and shape-phase transitions are reviewed and it is found that they are not actually true phase transitions and it is only an artifact of the mean field models. Since quantal number and spin fluctuations and statistical fluctuations in pair gap, deformation degrees of freedom and energy when incorporated, it wash out the pairing-phase transition and smooth out the shape-phase transition. Phase transitions due to collapse of pair gap and deformation is discussed and a clear picture of pairing-phase transition in light nuclei is presented in which pairing transition is reconciled.

Our laboratory study concerns the competitive interaction between two marine rotifer species Bra- chionus plicatilis and Brachionus rotundiformis at five algal （ Nannochloropsis salina） concentrations （0.4＊106 to 32.4x106 cells/ml） and at four initial inoculation densities （numerically, 100% B. pli- catilis; 75% B. plicatilis and 25% B. rotundiforrnis, 50% each of the two species; 25% B. plicatilis and 75% B. rotundiformis and 100% B. rotundiforrnis. The initial biomass varied as 0.33 and 0.22 #g/ml for B. plicatilis and B. rotundiformis respectively. Experiments were performed at （25±1）C. Population densities were enumerated and the medium was changed daily up to 8 d in the ex- periment. At the lowest food level tested, B. rotundiformis formed superior competitor than B. plicatilis, regardless of starting inoculation density. Generally when the food concentrations in- creased, B. plicatilis showed a greater increase in biomass than B. rotundiforrnis. B. rotundiformis formed the largest in population growth, regardless of increasing food concentrations. When grown alone, B. plicatilis reached peak abundances of （1.311 5±0.028） and （137.5±0.014） μg/ml at low and high food densities respectively. The corresponding values of B. rotundiformis were 0.724 5±0.016 and 18.15±0.021. The adverse effects of B. rotundiforrnis on the peak abundances of B. plicatilis were observed at the lowest food level and higher initial density. The rate of population growth in controls varied from （0.792±0.162） to （1.482±0.132） ttm/d for B. plicatilis and （0.445~0.041） to （0.856±0.012） μm/d for B. rotundiformis, depending on food level. When both species were intro- duced together, low food levels favoured higher abundance of B. rotundiforrnis than B. plicatilis, suggesting that increased population density of the smaller B. rotundiforrnis was more successfull than larger B. plicatilis in brackish waters. Biocatalysts are intrinsically reactive and hence their operational stability is of vital significance for any bioprocess. The setback in biocatalyst stability has been tackled from diverse prospects. Inherently, stable biocatalysts are markedly realized and a regular attempt is being made to seek out new organisms that harbor them. Here, we analyzed the industrial biocatalyst lipase A （Native） of Bacillus subtilis and its six thermostable mutants （2M, 3M, 4M, 6M, 9M and 12M） computationally using conformational sampling technique. Consequently, the various structural events deciphering thermostability like root mean square deviation, root mean square fluctuation, radius of gyration and polar surface area showed mutant 12M to be highly stable with statistical validation. Besides, static model analysis involving intra-molecular interactions, secondary structure, solvent accessibility, hydrogen bond pattern, simulated thermal denaturation and desolvation energy also supported 12M comparatively. Of note, the presence of high secondary structural rigidity and hydrogen bonds increased thermostability and functionality of 12M, thus selecting it as a best template for designing thermostable lipases in future. Also, this study has a significant implication toward a better understanding of conformational sampling in enzyme catalysis and enzyme engineering.

In this work, the most detrimental missense mutations of aspartoacylase that cause Canavan's disease were identified computationally and the substrate binding efficiencies of those missense mutations were analyzed. Out of 30 missense mutations, I-Mutant 2.0, SIFT and PolyPhen programs identified 22 variants that were less stable, deleterious and damaging respectively. Subsequently, modeling of these 22 variants was performed to understand the change in their conformations with respect to the native aspartoacylase by computing their root mean squared deviation (rMSD). Furthermore, the native protein and the 22 mutants were docked with the substrate NAA (N-Acetyl-Aspartic acid) to explain the substrate binding efficiencies of those detrimental missense mutations. Among the 22 mutants, the docking studies identified that 15 mutants caused lower binding affinity for NAA than the native protein. Finally, normal mode analysis determined that the loss of binding affinity of these 15 mutants was caused by altered flexibility in the amino acids that bind to NAA compared with the native protein. Thus, the pre- sent study showed that the majority of the substrate-binding amino acids in those 15 mutants displayed loss of flexibility, which could be the theoretical explanation of decreased binding affinity between the mutant aspartoacylases and NAA.

Aluminium alloy AA2219 was reinforced with TiB2 particles introduced in-situ by the saltmetal reaction technique. The microstructural examinations of the composites clearly reveal the formation of TiB2 particles with a hexagonal morphology. The addition of TiB2 particles results in increased mechanical properties, such as 0.2%YS, UTS and hardness. The improvement in mechanical properties is correlated to the microstructure.

In this work, the most detrimental missense mutations of Madl protein that cause various types of cancer were identified computationally and the substrate binding efficiencies of those missense mutations were analyzed. Out of 13 missense mutations, I Mutant 2.0, SIFT and PolyPhen programs identified 3 variants that were less stable, deleterious and damaging respectively. Subsequently, modeling of these 3 variants was performed to understand the change in their conformations with respect to the native Madl by computing their root mean squared deviation （rMSD）. Furthermore, the native protein and the 3 mutants were docked with the binding partner Mad2 to explain the substrate binding efficiencies of those detrimental missense mutations. The docking studies identified that all the 3 mutants caused lower binding affinity for Mad2 than the native protein. Finally, normal mode analysis determined that the loss of binding affinity of these 3 mutants was caused by altered flexibility in the amino acids that bind to Mad2 compared with the native protein. Thus, the present study showed that majority of the substrate binding amino acids in those 3 mutants displayed loss of flexibility, which could be the theoretical explanation of decreased binding affinity between the mutant Madl and Mad2.

With the growing global urban population and the emergence of megacities, there is a huge demand for arable land to meet the food demand and reduce malnutrition. Conventional agricultural practices lead to deforestation of the land for crop production and agricultural intensification to produce higher yield per unit area. These activities have been established to have negative impact on the environment thereby causing soil and water pollution. It is important to consider the use of vertical farming technology, which utilizes both horizontal and vertical space, and efficiently uses nutrients, water, and time (off season production with artificial lighting) more effectively to produce higher yield per unit volume of space than the conventional outdoor farming. Microgreens are taken into consideration to be grown under innovative vertical farming technology since they are rich in phytonutrients and they can be harvested in a short period of time. This paper reviews the current growing conditions of microgreens in vertical farming such as crop selection, media, light, nutrient solution, and containers while identifying knowledge gaps. Further, study in this area may lead to improved growing conditions to help solve the global issues and challenges surrounding food security, safety, and resource optimization.
