Upper Lillooet River Hydroelectric Project： The Challenges of Constructing a Power Tunnel for Run-of-River Hydro Projects in Mountainous British Columbia

The Upper Lillooet River Hydroelectric Project （ULHP） is a run-of-river power generation scheme located near Pemberton, British Columbia, Canada, consisting of two separate hydroelectric facilities （HEFs） with a combined capacity of 106.7 MW. These HEFs are owned by the Upper Lillooet River Power Limited Partnership and the Boulder Creek Power Limited Partnership, and civil and tunnel construction was completed by CRT-ebc. The Upper Lillooet River HEF includes the excavation ofa 6 m wide by 5.5 m high and approximately 2500 m long tunnel along the Upper Lillooet River Valley. The project is in a moun- tainous area; severe restrictions imposed by weather conditions and the presence of sensitive wildlife species constrained the site operations in order to limit environmental impacts. The site is adjacent to the Mount Meager Volcanic Complex, the most recently active volcano in Western Canada. Tunneling conditions were very challenging, including a section through deposits associated with the most recent eruption from Mount Meager Volcanic Complex （-2360 years before the present）. This tunnel section included welded breccia and unconsolidated deposits composed of loose pumice, organics （that represent an old forest floor）, and till, before entering the underlying tonalite bedrock. The construction of this section of the tunnel required cover grouting, umbrella support, and excavation with a combination of road header, hydraulic hammer, and drilling-and-blasting method. This paper provides an overview of the project, a summary of the key design and construction schedule challenges, and a description of the successful excavation of the tunnel through deposits associated with the recent volcanic activity.

Do microsporidia function as ＂biological weapon＂ for Harmonia axyridis under natural conditions？

Invasive alien species, such as the multicoloured Asian ladybird Harmonia axyridis, are often regarded as major drivers ofbiodiversity loss. Therefore understanding which characteristics or mechanisms contribute to their invasive success is important. Here the role of symbiotic microsporidia in the hemolymph of H. axyridis was investigated in the context of intraguild predation between wild-caught H. axyridis and the native ladybird species Coccinella septempunctata. The microsporidia were recently discussed to contribute to the unpalatability of Harmonia for other coccinellids during intraguild predation and to function as ＂biological weapons＂. In the present study, visual detection of microsporidia in hemolymph samples revealed that 73.5 % of H. axyridis were infected. Intraguild predation experiments between larvae of the two species showed a significant competitive advantage for H. axyridis, even against larger larvae of C. septempunctata. Adult C. septempunctata always killed and fed on H. axyridis larvae. However only 11.4 % （4 of 47） of C. septempunctata that fed on infected H. axyridis died within 4 months. In contrast to previous studies this suggests that microsporidia or harmonine, the chemical defense compound of H. axyridis, do not lead to death of C. septempunctata preying on larvae ofH. axyridis. Instead our results support the idea that competitive advantage during intraguild predation greatly facilitates the success of H. axyridis and that this may help this highly invasive species to outcompete native species. The impact of microsporidia on Harmonia itself as well as on interspecific interactions require further studies.

Immune-mediated anti-neoplastic effect of intratumoral RSV envelope glycoprotein expression is related to apoptotic death of tumor cells but not to the size of syncytia

AIM： To promote the development of improved tumor vaccination strategies relying on the intratumoral expression of viral fusogenic membrane proteins, we elucidated whether the size of syncytia or the way tumor cells die has an effect on the therapeutic outcome. METHODS： In two syngeneic subcutaneous murine colon cancer models we assessed the anti-neoplastic effect on vector-treated and contralateral untreated tumors. RESULTS： Intratumoral injection of a replication-defective adenovirus encoding respiratory syncytial virus fusion protein （RSV-F） alone （***-F） or together with the attachment glycoprotein RSV-G （***-F/G） led to a significant growth reduction of the vector-treated and contralateral untreated tumors. The treatment response was associated with a strong tumor-specific CTL response and significantly improved survival with medians of 46 d and 44 d, respectively. Intratumoral injection of Ad,RSV-G or a soluble RSV-F encoding adenovirus （***-Fsol） had no significant anti-neoplastic effect. The median survival of these treabnent groups and of ***-treated control animals was about 30 d. CONCLUSION： Although in vitro transduction of colon cancer cell lines with ***-F/G resulted in about 8-fold larger syncytia than with ***-F, the in vivo outcome was not sig nificantly different, Transduction of murinecolon cancer cell lines with *** or ***-F/G caused apoptotic cell death, in contrast to transduction with ***-G or ***-Fsol, suggesting an importance of the mode of cell death. Overall, these findings provide insight into improved tumor vaccination strategies relying on the intratumoral expression of viral fusogenic membrane proteins.

Assessing the sensitivity of Melanoplus frigidus （Orthoptera： Acrididae） to different weather conditions＂ A modeling approach focussing on development times

The temperature and soil moisture conditions as well as vegetation patterns were studied to describe the habitat and to model the life cycle of Melanoplusfrigidus, a true alpine grasshopper of the Scandes. In the low alpine belt of the Norwegian Scandes the species colonizes only the warmest microhabitats with maximum soil surface temperatures of 31℃. Vegetation of these habitats consists of shrub-rich heath dominated by Vaccinium myrtillus and Calluna vulgaris. Using continuously measured temperature data, the development times for four different seasons were modeled and related to field observations. The maximum delay of adult molt was estimated to amount to 3 weeks, the delay being determined by the variation in spring temperature conditions between different years. The possibilities of using M. frigidus as an indicator organism of climate change effects on alpine zoo-coenoses of the Scandes are discussed.

Bofedal response to climate variability, local management, and water extraction: A case study of Chucuyo, Northern Chile

Andean high-altitude wetlands are important ecosystems that serve a range of socialecological functions. In the Andes, bofedales, a specific type of peat-producing wetland, are essential for the sustainability of mountain ecosystems and indigenous pastoralist communities. The Chucuyo bofedal in northern Chile is affected by climatic variability and water extraction for agricultural uses via the Lauca canal. Herders in the local community also actively manage this wetland according to their traditional ecological knowledge to ensure permanent fodder for their animals. In this article, we analyze the annual behavior of the Chucuyo bofedal after the wet season. Based on precipitation data, extracted water flows, vegetation vigor from satellite images, and an ethnographic approach, we determined that the bofedal’s productivity was negatively correlated with the amount of precipitation and positively correlated with the surface area of the wetland. However, water extraction via the Lauca channel had no significant relationship with either surface area or vegetation vigor. We identify community practices and traditional ecological knowledge as key elements in the maintenance of these ecosystems. This situation is critical in the context of an aging population and the current pattern of migration out of the region to urban areas. The results provide substantial empirical evidence for future decision-making regarding the conservation of these ecosystems.

In a recent publication in Cell,Hasegawa and colleagues reveal that cytotoxic CD4-positive cytotoxic T lymphocytes(CD4+CTLs)eliminate senescent fibroblasts with a surface expression of the immunologically important human leukocyte antigen class Il(HLA-)and the human cytomegalovirus glycoprotein B(HCMV-gB).1 This implies that a virome-immune cell interaction controls the fate of human skin cells that are linked to the aging process and associated diseases.

In the early 2000s, the approach to perioperative fluid management in major abdominal surgery underwent a paradigm change in favor of restrictive, rather than standard or liberal, fluid regimens. The basis for this shift in philosophy was a result of randomized controlled trials such as one by Brandstrup et al., which demonstrated improved morbidity and outcomes with perioperative fluid restriction (1). With similar results reported across a variety of other prospective trials, the practice of perioperative fluid restriction garnered more support and acceptance (2,3).
