Development of a radiolabeled site-specific single-domain antibody positron emission tomography probe for monitoring PD-L1 expression in cancer

Despite advances in immunotherapy for the treatment of cancers,not all patients can benefit from programmed cell death ligand 1(PD-L1)immune checkpoint blockade ***-PD-L1 therapeutic effects reportedly correlate with the PD-L1 expression level;hence,accurate detection of PD-L1 expression can guide immunotherapy to achieve better therapeutic ***,based on the high affinity antibody Nb109,a new site-specifically radiolabeled tracer,^(68)Ga-NODA-cysteine,aspartic acid,and valine(CDV)-Nb109,was designed and synthesized to accurately monitor PD-L1 *** tracer ^(68)Ga-NODA-CDV-Nb109 was obtained using a site-specific conjugation strategy with a radiochemical yield of about 95%and radiochemical purity of 97%.It showed high affinity for PD-L1 with a dissociation constant of 12.34±1.65 *** the cell uptake assay and positron emission tomography(PET)imaging revealed higher tracer uptake in PD-L1-positive A375-hPD-L1 and U87 tumor cells than in PD-L1-negative A375 tumor ***,dynamic PET imaging of a NCI-H1299 xenograft indicated that doxorubicin could upregulate PD-L1 expression,allowing timely interventional *** conclusion,this tracer could sensitively and dynamically monitor changes in PD-L1 expression levels in different cancers and help screen patients who can benefit from anti-PD-L1 immunotherapy.

Many detailed data on past geological hazard events are buried in geological hazard reports and have not been fully utilized. The growing developments in geographic information retrieval and temporal information retrieval offer opportunities to analyse this wealth of data to mine the spatiotemporal evolution of geological disaster occurrence and enhance risk decision making. This study presents a combined NLP and ontology matching information extraction framework for automatically recognizing semantic and spatiotemporal information from geological hazard reports. This framework mainly extracts unstructured information from geological disaster reports through named entity recognition, ontology matching and gazetteer matching to identify and annotate elements, thus enabling users to quickly obtain key information and understand the general content of disaster reports. In addition, we present the final results obtained from the experiments through a reasonable visualization and analyse the visual results. The extraction and retrieval of semantic information related to the dynamics of geohazard events are performed from both natural and human perspectives to provide information on the progress of events.

In-situ construction of hybrid artificial SEI with fluorinated siloxane to enable dendrite-free Li metal anodes

Lithium(Li)metal anode holds great promise for high-energy-density rechargeable ***,it suffers from the Li dendrites growth and uncontrollable side reactions with electrolyte due to the unstable solid electrolyte interphase(SEI)***,we propose a facile strategy for the in-situ fabricate of organic-inorganic composite artificial SEI layers on Li surfaces,which consist of organic fluorinated siloxane and inorganic LiF-rich *** hybrid artificial SEI endows high mechanical strength(13.1 GPa)and Liþtransfer number(0.62).Such robust SEI protective layers can not only guide uniform nucleation and deposition of Li metal by facilitating uniform Li-ion distribution,but also prevent unfavourable side ***,the protected metallic lithium anode(PMTFPS-Li)anode enables stable Li plating/stripping performance in symmetric cells for more than 300 h at 4 mA$h/cm^(2)under a high areal capacity of 4 mA/cm^(2).Moreover,the PMTFPS-Li/S cells could maintain more than 300 stable cycles at 0.5C and the PMTFPS-Li/LFP cells present excellent cycling performance(400 cycles at 1C)and enhanced rate capability(110.4 mA$h/g at 3 C).This work will inspire the design of artificial SEI on Li anodes for advanced Li metal batteries.

Construction and expression of inverted configuration of retroviral vector containing intron 1 of hFIX

Intron was found to play an important role in improving gene expression. To improve the human factor IX(hFIX) expression level in hemophilia B gene therapy study, the retroviral vector containing intron 1 of hFIX gene was constructed in forwarded configuration, but the intron 1 was found spliced in virus particles by rT_PCr detection. So the inverted configuration vector G1NaPAi′IX was suggested and constructed on the basis of SNMBAIXm and transfected into PA317. Then C2C12 cells were transfected using the above virus supernatant and the G418_resistant clones were selected. PCr and rT_PCr detection found that intron 1 structure existed in C2C12 clones and retroviral particles. And the expression level of inverted vector was 3 times higher than that of forwarded vector. These results showed that the inverted configuration vector was in deed able to avoid splicing of intron 1 during the process of retroviral packaging and improved the expression level of hFIX protein.

Humic-Like Substances from Different Compost Extracts Could Significantly Promote Cucumber Growth

The effects of direct extracts of compost (DEC), aerated fermentation extracts of compost (AFEC) and non-aerated fermentation extracts of compost (NAFEC) on cucumber growth and the action mechanisms were evaluated based on the structure and activity analysis of humic-like substances. AFEC increased cucumber growth most significantly, followed by DEC and NAFEC, which was insignificant compared to the control treatment. Humic-like substances from compost extracts played an important role in promoting cucumber growth. Application of humic-like substances stimulated auxin-like activity and increased chlorophyll content and nitrogen accumulation in plants. The positive auxin-like activity of humic-like substances could be attributed to the relative distribution of special carbon groups, such as those with a large amount of peptidic and carbohydratic groups or with a low content of phenolic groups. In conclusion, the best growth promotion by application of AFEC was mainly attributed to the humic-like substances in the AFEC.

Electronic tattoos(e-tattoos),also known as epidermal electronics,are ultra-thin and ultra-soft noninvasive but skin-conformable devices with capabilities including physiological sensing and transdermal stimulation and *** fabrication of e-tattoos out of conventional inorganic electronic materials used to be tedious and *** developed cut-and-paste method has significantly simplified the process and lowered the ***,existing cut-and-paste method entails a medical tape on which the electronic tattoo sensors should be pasted,which increases tattoo thickness and degrades its *** address this problem,here we report a slightly modified cut-and-paste method to fabricate low-cost,open-mesh e-tattoos with a total thickness of just 1.5μm.E-tattoos of such thinness can be directly pasted on human skin and conforms to natural skin *** demonstrate that this ultra-thin,tape-free e-tattoo can confidently measure electrocardiogram(ECG),skin temperature,and skin *** rate and even respiratory rate can be extracted from the ECG signals.A special advantage of such ultra-thin e-tattoo is that it is capable of high-fidelity sensing with minimized motion artifacts under various body *** of perspiration are found to be insignificant due to the breathability of such e-tattoos.

rapid recovery of Classical Swine Fever Virus Directly from Cloned cDNA

The reverse genetics for classical swine fever virus （CSFV） is currently based on the transfection of in vitro transcribed rNA from a viral genomic cDNA clone, which is inefficient and time-consuming. This study was aimed to develop an improved method for rapid recovery of CSFV directly from cloned cDNA. Full-length genomic cDNA from the CSFV Shimen strain, which was flanked by a T7 promoter, the hepatitis delta virus ribozyme and T7 terminator sequences, was cloned into the low- copy vector pOK12, producing pOKShimen-rzTФ. Direct transfection of pOKShimen-rzTqb into PK/T7 cells, a PK-15- derived cell line stably expressing bacteriophage T7 rNA polymerase, allowed CSFV to be rescued rapidly and efficiently, i.e., at least 12 h faster and 31.6-fold greater viral titer when compared with the in vitro transcription-based rescue system. Furthermore, the progeny virus rescued from PK/T7 cells was indistinguishable, both in vitro and in vivo, from its parent virus and the virus rescued from classical reverse genetics. The reverse genetics based on intracellular transcription is efficient, convenient and cost-effective. The PK/T7 cell line can be used to rescue CSFV directly from cloned cDNA and it can also be used as an intracellular transcription and expression system for studying the structure and function of viral genes.

ANALYSIS OF SUrFACE SINK OF EXPLOrATION VACANCY IN GOLD MINING ArEA OF ZHAOYUAN CTIY BASED ON rS AND GIS

Surface sink is a main geological calamity of gold mining areas and a main factor to restrict economic sustainable development of mining zone. Based on former investigations, this article draws the environment information of surface sink of exploration vacancy in gold mining area of Zhaoyuan City, Shangdong Province by rS technology. Through spatial simulation analysis and expert diagnoses on the basis of GIS technology, the article affirms the inducement factors of the surface sink. Then using these factors as distinguishing ones the authors prognosticate the criticality of other exploration vacancies. The results indicate that the surface sink area of study area in Zhaoyuan City, has already come to 0.78km2 and it is forecasted that 0.97km2 of the exploration vacancy belongs to high danger area. Decisive measures need taking in order to prevent this crucial problem. Another 1.57km2 of the exploration vacancies belongs to middle danger area, which will sink when meeting some inducing factors, such as earthquake. Still another 1.53km2 of the exploration vacancies belongs to low danger area that can not lead to surface sink when meeting common inducing factors.

OBJECTIVE: To systematically assess the effects of individualized Chinese medicines on recurrent urinary tract infections(rUTIs).METHODS: This study recruited 230 adult female patients in the remission phase of rUTIs from five hospitals in China. The patients were randomly allocated to two groups: an individualized Chinese medicine group(n = 114) and a control group(n = 116). Patients in the Chinese medicine group received individualized Chinese herbs, which were evaluated for syndrome *** in the control group received antibiotic treatment combined with a Chinese medicine placebo. The duration of treatment was three courses of four weeks each, with a three-month subsequent follow-up. UTI recurrence rate,Traditional Chinese Medicine(TCM) syndrome scores,36-item Short Form Survey(SF-36) score, and urine secretory immunoglobulin A(SIgA) were measured and analyzed before and after treatment in each group. rESULTS: Patients from the Chinese medicine group exhibited significant decreases in both short-and longterm UTI recurrence rates compared with the control group(P rome scores between the Chinese medicine and control groups were significant(P rage SF-36 quality-of-life scores in the Chinese medicine group were also significantly higher than those in the control group after treatment(P roup also demonstrated a significant increase in urine SIg A expression. CONCLUSION: Taken together, compared to the oftenused long-term antimicrobial prophylaxis during the remission stage of r UTIs, treating patients with an individualized Chinese medicine decoction by syndrome differentiation could effectively reduce the recurrence rate, improve the patients' TCM syndrome scores and quality of life, and enhance immunity, which in turn helps to prevent antibiotic resistance.
