Extrusion-based 3D bioprinting is a direct deposition approach used to create three-dimensional(3D)tissue scaffolds typically comprising *** are hydrated polymer networks that are chemically or physically ***,3D bioprinting is performed in air,despite the hydrated nature of hydrogels and the potential advantage of using a liquid phase to provide cross-linking and otherwise functionalize the *** this work,we print gelatin alginate hydrogels directly into a cross-linking solution of calcium chloride and investigate the influence of nozzle diameter,distance between nozzle and surface,calcium chloride concentration,and extrusion rate on the dimensions of the printed *** hydrogel layer height was generally found to increase with increasing extrusion rate and nozzle distance,according to the increased volume extruded and the available space,*** addition,the hydrogel width was generally found to increase with decreasing nozzle distance and cross-linking concentration corresponding to confinement-induced spreading and low crosslinking regimes,***/height ratios of^1 were generally achieved when the nozzle diameter and distance were comparable above a certain cross-linking *** these relationships,biocompatible 3D multilayer structures were successfully printed directly into calcium chloride cross-linking solution.

Secondary forests and human-made forest gaps are conspicuous features of tropical ***,behavioral responses to these aspects of anthropogenically modified forests remain poorly ***,we analyze the effects of small human-made clearings and secondary forests on tropical bats by examining the guild-and species-level activity patterns of phyllostomids sampled in the Central Amazon,***,we contrast the temporal activity patterns and degree of temporal overlap of 6 frugivorous and 4 gleaning animalivorous species in old-growth forest and second-growth forest and of 4 frugivores in old-growth forest and forest *** activity patterns of frugivores and gleaning animalivores did not change between old-growth forest and second-growth,nor did the activity patterns of frugivores between old-growth forest and ***,at the species level,we detected significant differences for Artibeus obscurus(old-growth forest ***-growth)and ***(old-growth forest ***).The degree of temporal overlap was greater than random in all sampled ***,for frugivorous species,the degree of temporal overlap was similar between old-growth forest and second-growth;whereas for gleaning animalivores,it was lower in second-growth than in old-growth *** the contrary,forest clearings were characterized by increased temporal overlap between *** in activity patterns and temporal overlap may result from differential foraging opportunities and dissimilar predation ***,our analyses suggest that activity patterns of bats in second-growth and small forest clearings,2 of the most prominent habitats in humanized tropical landscapes,varies little from the activity patterns in old-growth forest.

AIMTo evaluate the intra-operator repeatability in healthy subjects using the WAM-5500 auto-kerato/refractometer and the iTrace aberrometer, to compare the refractive values and the subjective refraction obtained with both devices and to determine which of these three spherocylindrical corrections allows the subject to achieve the best visual ***-two non-presbyopic healthy eyes of 42 subjects were enrolled in this prospective study. refractive values were compared, evaluating the repeatability, the relationship between the methods and the best visual comfort ***, cylinder and axis results showed good intraclass correlation coefficients (ICC); the highest ICC was obtained using the spherical refraction with the autorefractometer and the aberrometer, achieving levels of 0.999 and 0.998, respectively. The power vector (PV) was calculated for each refraction method, and the results indicated that there were no statistically significant differences between them (P>0.05). Direct comparison of PV measurements using the three methods showed that aberrometer refraction gave the highest values, followed by the subjective values; the autorefractometer gave the lowest values. The subjective method correction was most frequently chosen as the first selection. Equal values were found for the autorefractometer and the aberrometer as the second *** iTrace aberrometer and the WAM-5500 auto-kerato/refractometer showed high levels of repeatability in healthy eyes. refractive corrections with the aberrometer, the autorefractometer and subjective methods presented similar results, but spherocylindrical subjective correction was the most frequently selected option. These technologies can be used as complements in refractive evaluation, but they should not replace subjective refraction.

To date,skin wounds are still an issue for healthcare *** numerous approaches have been developed over the years for skin regeneration,recent advances in regenerative medicine offer very promising strategies for the fabrication of artificial skin substitutes,including 3D bioprinting,electrospinning or spraying,among *** particular,skin sprays are an innovative technique still under clinical evaluation that show great potential for the delivery of cells and hydrogels to treat acute and chronic *** sprays present significant advantages compared to conventional treatments for wound healing,such as the facility of application,the possibility to treat large wound areas,or the homogeneous distribution of the sprayed *** this article,we review the latest advances in this technology,giving a detailed description of investigational and currently commercially available acellular and cellular skin spray products,used for a variety of diseases and applying different experimental ***,as skin sprays products are subjected to different classifications,we also explain the regulatory pathways for their commercialization and include the main clinical trials for different skin diseases and their treatment ***,we argue and suggest possible future trends for the biotechnology of skin sprays for a better use in clinical dermatology.

Poly-histidine peptides such as H6(HHHHHH)are used in protein biotechnologies as purification tags,protein-assembling agents and endosomal-escape *** pleiotropic properties of such peptides make them appealing to design protein-based smart materials or nanoparticles for imaging or drug delivery to be produced in form of recombinant ***,the clinical applicability of H6-tagged proteins is restricted by the potential immunogenicity of these *** this study,we have explored several humanized histidine-rich peptides in tumor-targeted modular proteins,which can specifically bind and be internalized by the target cells through the tumoral marker *** were particularly interested in exploring how protein purification,self-assembling and endosomal escape perform in proteins containing the variant histidine-rich *** the tested candidates,the peptide H5 E(HEHEHEHEH)is promising as a good promoter of endosomal escape of the associated fulllength protein upon endosomal *** numerical modelling of cell penetration and endosomal escape of the tested proteins has revealed a negative relationship between the amount of protein internalized into target cells and the efficiency of cytoplasmic *** fact demonstrates that the His-mediated,proton sponge-based endosomal escape saturates at moderate amounts of internalized protein,a fact that might be critical for the design of protein materials for cytosolic molecular delivery.

Transcription factors(TFs)regulate gene expression by binding to cis-regulatory sequences in the promoters of target *** research is helping to decipher in part the cis-regulatory code in eukaryotes,including plants,but it is not yet fully understood how paralogous TFs select their *** we addressed this question by studying several proteins of the basic helix-loop-helix(bHLH)family of plant TFs,all of which recognize the same DNA *** focused on the MYC-related group of bHLHs,that redundantly regulate the jasmonate(JA)signaling pathway,and we observed a high correspondence between DNA-binding profiles in vitro and MYC function in *** demonstrated that A/T-rich modules flanking the MYC-binding motif,conserved from bryophytes to higher plants,are essential for TF recogni*** observed particular DNA-shape features associated with A/T modules,indicating that the DNA shape may contribute to MYC DNA *** extended this analysis to 20 additional bHLHs and observed correspondence between in vitro binding and protein function,but it could not be attributed to A/T modules as in *** conclude that different bHLHs may have their own codes for DNA binding and specific selection of targets that,at least in the case of MYCs,depend on the TF-DNA interplay.

The presence of genes encoding organellar proteins in different cellular compartments necessitates a tight coordination of expression by the different genomes of the eukaryotic cell. This coordination of gene expression is achieved by organelle-to-nucleus or retrograde communication. Stress-induced perturbations of the tetrapyrrole pathway trigger large changes in nuclear gene expression in plants. recently, we identified HSP90 proteins as ligands of the pu- tative plastid signal Mg-ProtolX. In order to investigate whether the interaction between HSP90 and Mg-ProtolX is bio- logically relevant, we produced transgenic lines with reduced levels of cytosolic HSP90 in wild-type and gun5 backgrounds. our work reveals that HSP90 proteins respond to the tetrapyrrole-mediated plastid signal to control expres- sion of photosynthesis-associated nuclear genes （PhANG） during the response to oxidative stress. We also show that the hy5 mutant is insensitive to tetrapyrrole accumulation and that Mg-ProtolX, cytosolic HSP90, and HY5 are all part of the same signaling pathway. These findings suggest that a regulatory complex controlling gene expression that includes HSP90 proteins and a transcription factor that is modified by tetrapyrroles in response to changes in the environment is evolutionarily conserved between yeast and plants.
