Pseudospondias microcarpa is used in ethnomedicine to manage central nervous system *** hydroethanolic extract(PME)from the leaves of the plant has shown anxiolytic-like properties in mice anxiety ***,its effects in chronic anxiety models and possible mechanism(s)of action were not ***,the current study evaluated the anxiolytic-like mechanisms of PME in zebrafish models of *** zebrafish light dark test(LDT)and novel tank test(NTT)were employed to assess the anxiolytic-like effects of PME(0.1,0.3,1.0 mg mL^(−1)),fluox-etine(3×10^(−5)mg mL^(−1))and diazepam(1.5×10^(−7)mg mL^(−1)).The chronic unpredictable stress(CUS)test was used to further evaluate the extract’s anxiolytic-like *** potential mechanisms of anxiolytic action of the extract was evaluated after pre-treated with flumazenil,granisetron,methysergide,or pizotifen,all at 1×10^(−3)mg mL^(−1).The extract significantly decreased anxiety behaviours in the NT and LD *** observed effects of the extract were however counteracted by flumazenil,granisetron,methysergide and pizotifen *** addition,PME treatment significantly reversed CUS-induced anxiety behaviours in *** show that PME possesses anxiolytic-like effects possibly through interaction with serotonergic and gamma-aminobutyric acid mediated pathways.

背景:近年来对于退变腰椎间盘组织蛋白质成分改变的研究取得了很大进展,但是对于正常和退变腰椎间盘整体蛋白质成分改变的分离和差异鉴定却鲜见报道。目的:建立正常和退变腰椎间盘蛋白质的双向电泳图谱,鉴定差异表达蛋白,探讨腰椎间盘退变的生物学机制。设计、时间及地点:对比观察,于2007-06/2008-12在南方医科大学病理生理实验室完成。材料:由自愿者提供的正常和退变腰椎间盘组织各6例,年龄21～45岁,其中男9例,女3例;取材范围L3～L5。方法:氯化铯梯度离心去除正常和退变腰椎间盘组织蛋白多糖,通过固相pH梯度(IPG)等电聚焦和梯度聚丙烯酰胺双向电泳分离蛋白质,分析电泳图像,使用基质辅助激光解吸附飞行时间质谱仪对差异蛋白质点进行分析鉴定。主要观察指标:正常腰椎间盘及退变腰椎间盘组织IPG等电聚焦双向凝胶电泳结果。结果:双向电泳显示正常和退变腰椎间盘蛋白质图谱差异显著,从中选取16个差异明显的位点进行质谱仪分析,确定了6种具有显著性意义的差异蛋白质。结论:双向电泳可有效地分离正常和退变腰椎间盘组织蛋白质,通过质谱仪鉴定可以确定其之间差异蛋白质的性质。

Grey Crowned Crane and Shoebills Conservation Status and Linkage to Livelihoods of Lake Victoria Shores Adjacent Communities in Mayuge District, Eastern Uganda

This research was done among communities living adjacent to Lake Victoria Shores in Mayuge District to generate knowledge on Shoebill, Grey Crowned Crane conservation status and linkage to livelihoods. This knowledge was necessary to guide design and implementation of interventions that would deliver on the species conservation and adjacent communities’ livelihood benefits at the same time. We obtained information through individual interviews with 101 members of households from targeted communities and key informers-technical officials from the district, and analysed this data using Statistical Package for the Social Sciences (SPSS) by Bryman and Cramer, (2011) Version 20. The results were then subjected to validation using focus group discussions with 10 groups of between 8 - 15 individuals representing the targeted communities and other stakeholders and the results from this further validated with ≥250 representatives of these stakeholders in a one day workshop and with evidences collected during field observations, before synthesis. The findings show: Grey Crowned Crane habitats declining due to mining, agriculture, fishing activities by adjacent communities for their livelihoods (food and income from sale of produce obtained from the species’ habitats). The communities say they need to be engaged in development and implementation of integrated interventions that will improve their livelihoods while securing conservation for Grey Crowned Cranes. They among others propose ecotourism enterprises, training and sensitization on environment and wildlife conservation practices, policies and laws to enhance their knowledge and compliance in management. The fisher folk say fish is often found within Grey Crowned Cranes’ habitats and breeding grounds in the Lake and to avoid destroying the habitats, they need engine boats and standard fishing nets to enable fishing far away in the deeper waters. Dirichlet Process Gaussian Mixture Models:Choice of the Base Distribution

In the Bayesian mixture modeling framework it is possible to infer the necessary number of components to model the data and therefore it is unnecessary to explicitly restrict the number of components. Nonparametric mixture models sidestep the problem of finding the ＂correct＂ number of mixture components by assuming infinitely many components. In this paper Dirichlet process mixture （DPM） models are cast as infinite mixture models and inference using Markov chain Monte Carlo is described. The specification of the priors on the model parameters is often guided by mathematical and practical convenience. The primary goal of this paper is to compare the choice of conjugate and non-conjugate base distributions on a particular class of DPM models which is widely used in applications, the Dirichlet process Gaussian mixture model （DPGMM）. We compare computational efficiency and modeling performance of DPGMM defined using a conjugate and a conditionally conjugate base distribution. We show that better density models can result from using a wider class of priors with no or only a modest increase in computational effort.

recent advances in scanning transmission electron microscopy(STEM)have led to increased development of multidimensional STEM imaging modalities and novel image reconstruction *** interest arises because the main electron lens in a modern transmission electron microscope usually has a diffraction-space information limit that is significantly better than the real-space resolution of the same *** state-of-affairs is sometimes shared by other scattering methods in modern physics and contributes to a broader excitement surrounding multidimensional techniques that scan a probe while recording diffraction-space images,such as ptychography and scanning nano-beam ***,the contrasting resolution in the two spaces raises the question as to what is limiting their effective ***,we examine this paradox by considering the effects of aberrations in both image and diffraction planes,and likewise separate the contributions of pre-and post-sample *** consideration provides insight into aberration-measurement techniques and might also indicate improvements for super-resolution techniques.

The pathophysiology of preeclampsia(PE)remains *** spiral artery remodeling dysfunction and PE offspring cardiovascular future development has been a worldwide *** collected placental and umbilical artery samples from normotensive and PE *** receptor(Mr)and its alternative splicing variant(ASV)expression and their biological effects on PE were *** Mr ASV was found to be highly expressed in all PE samples and slightly expressed in about half of the normotensive samples(umbilical artery,~57.58%;placenta,~36.84%).The Mr ASV expression was positively associated with blood pressure in both *** Mr ASV protein changed the aldosterone-induced expression pattern of Mr target genes related to ion exchanges and cell signaling *** Mr ASV can also impair the proliferation,migration,and tube formation ability of endothelial *** findings indicate that Mr ASV in PE placenta plays a pathogenic role in PE pathophysiology,especially in endothelial dysfunction,and the existence of the Mr ASV in PE umbilical artery provides a new direction in the study of PE offspring with increased risk of cardiovascular diseases.
