Anterior urethral strictures, where the length is more kthan 2 cm, are best treated by substitution urethroplasty with either preputial/penile skin flaps or free grafts.1 The use of dartos pedicled flaps has many advantages in terms of increased survival thanks to its own vascularization. Recently, buccal mucosa has become increasingly popular among urologists for urethral replacement when local penile skin is unavailable. Both penile skin flaps and buccal mucosa grafts have emerged as reliable urethral substitutes with comparable long-term results. These urethral substitutes are traditionally placed on the ventral aspect of the stricture and have a success rate of about 85%.

In this work,the effect of Re alloying on the phase composition,crystal structure,and mechanical properties of NbMoTaWRe_(x) (x=0,0.27,0.57,0.92,1.33) refractory high-entropy alloys (RHEAs) were systematically investigated by combining the calculation of phase diagram(CALPHAD),first-principle calculations and *** theoretical predictions showed good consistency with the experimental *** the increase in Re content,the theoretical results showed that all considered alloys have a single body-centered cubic (bcc) structure and the lattice constant and ductility were decreased,while the elastic moduli and hardness were *** avoid extreme brittleness,a strategic suggestion was given for the design of Re-containing RHEAs in the future.

Mge3Lie0.4Zr alloys containing RE elements(Gd,La,Nd)(Mge3LieREe0.4Zr alloys)are investigated to reveal the influence of homogenization treatment on microstructures and distributions of RE,Zr *** is found that 300C24 h homogenization treatment shows better improvement on the microstructure including the refinement of grain size,the dispersion of cellular dendrite and low melting point *** treatment,La and Nd segregate effectively at grain boundary and Zr segregates in the form of *** treatment induces the reduction of RE ***,the segregation of Zr in precipitates cannot be abated due to the relatively low diffusion rate compared with RE elements.

Directional axon regeneration and remyelination are crucial for repair of spinal cord injury(SCI),but existing treatments do not effectively promote those ***,we propose a strategy for construction of niche-specific spinal white matter-like tissue(WMLT)using decellularized optic nerve(DON)loaded with neurotrophin-3(NT-3)-overexpressing oligodendrocyte precursor cells.A rat model with a white matter defect in the dorsal spinal cord of the T10 segment was *** WMLT transplantation group showed significant improvement in coordinated motor functions compared with the control *** transplants integrated well with host spinal cord white matter,effectively addressing several barriers to directional axonal regeneration and myelination during SCI *** WMLT,laminin was found to promote development of oligodendroglial lineage(OL)cells by binding to laminin ***,laminin could also guide linear axon regeneration via interactions with specific integrins on the axon *** WMLT developed here utilizes the unique microstructure and bioactive matrix of DON to create a niche rich in laminin,NT-3 and OL cells to achieve significant structural repair of *** protocol can help to promote research on repair of nerve injury and construction of neural tissues and organoids that form specific cell niches.

2022年2—3月,在内蒙古大兴安岭汗马国家级自然保护区开展原麝(Moschus moschiferus)野外调查和粪便样本采集,共收集疑似原麝新鲜粪便样本36份,经分子生物学方法确定原麝粪便样本33份,同时采集14种原麝可采食的植物样本。通过食性分析,测定可采食植物的宏营养和微营养含量,定量食物可获得性和可食植物的营养含量,以探究原麝冬季食性及食物资源可获得性。结果表明:不同种类食物组成比例和食物可获得性存在差异,地衣是原麝冬季大宗食物(50.65%),环境中食物可获得性不是决定食物组成比例的主要因素;原麝冬季首先从环境中摄取碳水化合物和Na含量高的草本类植物,其次有选择地采食能量、蛋白质及脂肪含量较高的灌木类(含低矮乔木)植物,以达到自身营养物质的均衡和满足特定的营养需求。通过转化的冗余分析(tb-RDA)发现,冬季生境中原麝采食的灌木类(含低矮乔木)与草本类植物的营养供应具有显著差异,且生境植物总结构性碳水化合物(TSC)和总能(GE)显著影响原麝冬季食物组成。研究结果表明冬季生境中原麝采食植物的可获得性及营养和能量分配对原麝采食策略的选择具有重要意义。

目的:为解决矿山开采顶板支护材料的短缺及石墨尾砂大量堆积问题。方法:采用碱激发的手段首次激发矿渣粉煤灰的火山灰活性、石墨尾砂作为骨料,制备沿空留巷支护体材料。试验采用响应面及满意度函数法进行配合比优化。结果:随着用碱量的提高,石墨尾砂沿空留巷支护体材料的抗压强度呈现出先增大后减小的趋势,成本逐渐增大;随着粉/矿的提高,沿空留巷支护体材料的抗压强度逐渐下降,流动性逐渐变好,成本减少;随着骨胶比的增加,沿空留巷支护体材料的抗压强度逐渐降低,成本逐渐降低。质量浓度对沿空留巷支护体材料的影响较小。结论:沿空留巷支护体具有最优目标强度且成本为最小,其最优配比为粉/矿为0.501、骨胶比为3.71、质量浓度为85.1%、用碱量为8%。意义:为提高石墨尾砂的工程实际应用提供试验及理论依据。

An efficient and rapid method to detect and verify natural antisense transcripts of animal genes

High-throughput sequencing has identified a large number of sense-antisense transcriptional pairs, which indicates that these genes were transcribed from both directions. Recent reports have demonstrated that many antisense RNAs, especially lnc RNA（long non-coding RNA）, can interact with the sense RNA by forming an RNA duplex. Many methods, such as RNA-sequencing, Northern blotting, RNase protection assays and strand-specific PCR, can be used to detect the antisense transcript and gene transcriptional orientation. However, the applications of these methods have been constrained, to some extent, because of the high cost, difficult operation or inaccuracy, especially regarding the analysis of substantial amounts of data. Thus, we developed an easy method to detect and validate these complicated RNAs. We primarily took advantage of the strand specificity of RT-PCR and the single-strand specificity of S1 endonuclease to analyze sense and antisense transcripts. Four known genes, including mouse β-actin and Tsix（Xist antisense RNA）, chicken LXN（latexin） and GFM1（Gelongation factor, mitochondrial 1）, were used to establish the method. These four genes were well studied and transcribed from positive strand, negative strand or both strands of DNA, respectively, which represented all possible cases. The results indicated that the method can easily distinguish sense, antisense and sense-antisense transcriptional pairs. In addition, it can be used to verify the results of high-throughput sequencing, as well as to analyze the regulatory mechanisms between RNAs. This method can improve the accuracy of detection and can be mainly used in analyzing single gene and was low cost.
