Dissecting the cell microenvironment of ovarian endometrioma through single-cell RNA sequencing

Ovarian endometrioma（OE）, also known as “chocolate cysts,” is a cystic mass that develops in the ovaries due to endometriosis and is a common gynecological condition characterized by the growth of endometrial tissue outside the uterus, leading to symptoms such as dysmenorrhea, pelvic pain, and infertility. However, the precise molecular and cellular mechanisms driving this pathophysiology remain largely unknown, posing challenges for diagnosis and treatment. Here, we employed integrated single-cell transcriptomic profiling of over 52,000 individual cells from endometrial tissues of OE patients and healthy donors and identified twelve major cell populations. We identified notable alterations in cell type-specific proportions and molecular signatures associated with OE. Notably, the activation of IGFBP5+macrophages with pro-inflammatory properties, NK cell exhaustion, and aberrant proliferation of IQCG+and KLF2+epithelium are key features and may be the potential mechanisms underlying the pathogenesis of OE. Collectively, our data contribute to a better understanding of OE at the single cell level and may pave the way for the development of novel therapeutic strategies.

BACKGROUND When herpes zoster is complicated with paralytic ileus,this mostly involves acute intestinal pseudo-obstruction of Ogilvie’s syndrome manifesting as obvious dilatation of the cecum and right colon;small intestinal obstruction is ***,we present a patient with a very rare case of small bowel *** SUMMARY A 76-year-old female patient complained of right upper quadrant *** days later,a blistering,right-sided rash of the thoracoabdominal dermatome(T5-T10) emerged in conjunction with small intestinal dilatation and the inability to *** tomography of the abdomen confirmed small bowel *** therapy,gastrointestinal decompression,and enemas proved *** 4 d of stagnation,an epidural block was performed for pain relief and prompted the passage of gas and stool,resolving the obstructive *** days later,the rash appeared dry and crusted,and the pain *** 5 d,no abnormality was visible by gastroenteroscopy,and the patient was discharged on day *** This case shows that herpes zoster may induce small bowel pseudo-obstruction in addition to colonic *** block can not only treat intercostal neuralgia but also resolve small bowel pseudo-obstruction caused by herpes zoster.

POD analysis on vortical structures in MVG wake by Liutex core line identification

The Liutex core line method, first combined with the snapshot proper orthogonal decomposition (POD), is utilized in a supersonic micro-vortex generator (MVG) wake flow at Ma = 2.5 and Reθ = 5 760 to reveal the physical significance of each POD mode of the flow field. Compared with other scalar-based vortex identification methods, the Liutex core line identification is verified to be the most appropriate approach that is threshold-free and provides full information of a fluid rotation motion. Meanwhile, the Liutex integration is employed to quantitatively track the evolution of the vortices in MVG wake and is applied to the determination of the effective control section of the MVG wake for the optimization study of MVG design. The physical mechanism of each POD mode for multi-scale and multi-frequency vortical structures is investigated by using Liutex core line identification to give some revelations. For the mean mode (mode 0) indicating the time-averaged velocity flowfield of the MVG wake flow, a pair of primary counter-rotating streamwise vortices and another pair of secondary vortices is uniquely identified by two pairs of Liutex core lines with Liutex magnitude. In contrast, mode 1 is featured by a fluctuated roll-up motion of streamwise vortex, and the streamwise component of the MVG wake is demonstrated to be dominant in terms of the total kinetic energy contribution. Meanwhile, a dominant shedding frequency of St = 0.072 is detected from the temporal behavior of mode 2, which has the organized arc-shaped vortex structures shedding from MVG induced by the K-H instability. Additionally, mode 4 subjects to low-frequency oscillations of the wall vortices and thus takes a relatively lower frequency of St = 0.044.

Design of Fault Diagnosis Observer for HAGC System on Strip Rolling Mill

By building mathematical model for HAGC （hydraulic automation gauge control） system of strip rolling mill, treating faults as unknown inputs induced by model uncertainty, and analyzing fault direction, an unknown input fault diagnosis observer group was designed. Fault detection and isolation were realized through making ob- server residuals robust to specific faults but sensitive to other faults. Sufficient existence conditions and design of the observers were given in detail. Diagnosis observer parameters for servo-valve, cylinder, roller and body rolling mill were obtained resoectively. The effectiveness of this diagnosis method was oroved bv actual data simulations.

Quantum machine learning based on quantum algorithms may achieve an exponential speedup over classical algorithms in dealing with some problems such as *** this paper,we use the method of training the lower bound of the average log likelihood function on the quantum Boltzmann machine(QBM)to recognize the handwritten number datasets and compare the training results with classical *** find that,when the QBM is semi-restricted,the training results get better with fewer computing *** shows that it is necessary to design a targeted algorithm to speed up computation and save resources.

A self-adaptive inorganic in-situ separator by particle crosslinking for nonflammable lithium-ion batteries

All-safe liquid-state lithium-ion batteries(ASLS-LIBs) is of great interest as they can potentially combine the safety of all-solid-state batteries with the high performance and low manufacturing cost of traditional liquid-state LIBs. However, the practical success of ASLS-LIBs is bottlenecked by the lack of advanced separator technology that can simultaneously realize high performances in puncturing-tolerability,fire-resistance, and importantly, wetting-capability with non-flammable liquid-electrolytes. Here, we propose a concept of inorganic in-situ separator(IISS) by hybrid-sol physical crosslinking directly onto the electrode surface to address the above challenges. Particularly, the hybrid-sol is designed with silica nanoparticles as the building block and poly(vinylidene difluoride) nanoparticles as the crosslinking agent. The critical factors for controlling the IISS microstructures and properties have been systematically investigated. The advantages of the IISS have been confirmed by its fast wetting with various fireresistant liquid-electrolytes, customizable thickness and porous structures, robust interface with planar or three-dimensional(3D)-structured electrodes, and importantly, unexpected self-adaptability against puncturing. Enabled by the above merits, a fire-resistant ASLS-LIB is successfully assembled and demonstrated with stable electrochemical performance. This sol-crosslinked IISS may open an avenue for the studies on the next-generation separator technology, cell assembling, solid electrolyte processing as well as non-flammable secondary batteries.

Polymer microbottle with flexible tunability for ultrasensitive ultrasound sensing

Optical whispering-gallery-mode microcavities have attracted significant attention for their potential in ultrasensitive ultrasound sensing, despite always relying on expensive tunable lasers in applications. In this study, we integrated an electrothermal tuning function inside the microcavity, enabling fast scanning of modes by applying voltages, which helps provide real-time searching and tracking of the optimal mode. Our device demonstrated a quality factor exceeding 10～6with a broad tuning range over 33 GHz. This structure achieved high sensitivity in ultrasound detection, with a noise equivalent pressure（NEP） of 3.35 mPa/Hz1/2at 20 MHz. We further reported the advantages of the mode thermal broadening effect for ultrasound detection, with successfully obtaining high-contrast photoacoustic images. Our research introduces an innovative approach for cost-effective, high-stability ultrasound detection with microcavity, showing great value for application in photoacoustic imaging.

Insufficient MIRO1 contributes to declined oocyte quality during reproductive aging

Mitochondrial Rho-GTPase 1 (MIRO1) is an outer mitochondrial membrane protein which regulates mitochondrial transport and mitophagy in mitosis. In present study, we reported the crucial roles of MIRO1 in mammalian oocyte meiosis and its potential relationship with aging. We found that MIRO1 expressed in mouse and porcine oocytes, and its expression decreased in aged mice. MIRO1 deficiency caused the failure of meiotic resumption and polar body extrusion in both mouse and porcine oocytes, which could be rescued by exogenous MIRO1supplementation. Mass spectrometry data indicated that MIRO1 associated with several cytoskeleton and cell cycle-related proteins, and MIRO1 regulated motor protein Dynein for microtubule-organizing centers (MTOCs) dynamics at germinal vesicle (GV) stage, which determined meiotic resumption. Furthermore, we found that MIRO1 regulated Aurora A and kinesin family member 11 (KIF11) for meiotic spindle assembly in oocytes. Besides, MIRO1 associated with several mitochondria-related proteins dynamic-related protein 1 (DRP1),Parkin and lysosomal-associated membrane protein 2 (LAMP2) for mitochondrial dynamics and mitophagy during oocyte meiosis. Taken together, our results suggested that MIRO1 played pivotal roles in meiotic resumption, spindle assembly and mitochondrial function in mouse and porcine oocytes, and its insufficiency might contribute to the oocyte maturation defects during aging.

Bioremediation of Hexavalent Chromium Pollution by Sporosarcina saromensis M52 Isolated from Offshore Sediments in Xiamen, China

Objective Cr（VI） removal from industrial effluents and sediments has attracted the attention of environmental researchers. In the present study, we aimed to isolate bacteria for Cr（VI） bioremediation from sediment samples and to optimize parameters of biodegradation. Methods Strains with the ability to tolerate Cr（VI） were obtained by serial dilution and spread plate methods and characterized by morphology, 16 S r DNA identification, and phylogenetic analysis. Cr（VI） was determined using the 1,5-diphenylcarbazide method, and the optimum p H and temperature for degradation were studied using a multiple-factor mixed experimental design. Statistical analysis methods were used to analyze the results. Results Fifty-five strains were obtained, and one strain（Sporosarcina saromensis M52; patent application number： 201410819443.3） having the ability to tolerate 500 mg Cr（VI）/L was selected to optimize the degradation conditions. M52 was found be able to efficiently remove 50-200 mg Cr（VI）/L in 24 h, achieving the highest removal efficiency at p H 7.0-8.5 and 35 °C. Moreover, M52 could completely degrade 100 mg Cr（VI）/L at p H 8.0 and 35 °C in 24 h. The mechanism involved in the reduction of Cr（VI） was considered to be bioreduction rather than absorption. Conclusion The strong degradation ability of S. saromensis M52 and its advantageous functional characteristics support the potential use of this organism for bioremediation of heavy metal pollution.
