Aldolase B attenuates clear cell renal cell carcinoma progression by inhibiting CtBP2

Aldolase B(ALDOB),a glycolytic enzyme,is uniformly depleted in clear cell renal cell carcinoma(ccRCC)tissues.We previously showed that ALDOB inhibited proliferation through a mechanism independent of its enzymatic activity in ccRCC,but the mechanism was not unequivocally identified.We showed that the corepressor C-terminal-binding protein 2(CtBP2)is a novel ALDOB-interacting protein in ccRCC.The CtBP2-to-ALDOB expression ratio in clinical samples was correlated with the expression of CtBP2 target genes and was associated with shorter survival.ALDOB inhibited CtBP2-mediated repression of multiple cell cycle inhibitor,proapoptotic,and epithelial marker genes.Furthermore,ALDOB overexpression decreased the proliferation and migration of ccRCC cells in an ALDOB-CtBP2 interaction-dependent manner.Mechanistically,our findings showed that ALDOB recruited acireductone dioxygenase 1,which catalyzes the synthesis of an endogenous inhibitor of CtBP2,4-methylthio 2-oxobutyric acid.ALDOB functions as a scaffold to bring acireductone dioxygenase and CtBP2 in close proximity to potentiate acireductone dioxygenase-mediated inhibition of CtBP2,and this scaffolding effect was independent of ALDOB enzymatic activity.Moreover,increased ALDOB expression inhibited tumor growth in a xenograft model and decreased lung metastasis in vivo.Our findings reveal that ALDOB is a negative regulator of CtBP2 and inhibits tumor growth and metastasis in ccRCC.

Atherosclerosis(AS)is characterized by impairment and apoptosis of endothelial cells,continuous systemic and focal inflammation and dysfunction of vascular smooth muscle cells,which is documented as the traditional cellular paradigm.However,the mechanisms appear much more complicated than we thought since a bulk of studies on efferocytosis,transdifferentiation and novel cell death forms such as ferroptosis,pyroptosis,and extracellular trap were reported.Discovery of novel pathological cellular landscapes provides a large number of therapeutic targets.On the other side,the unsatisfactory therapeutic effects of current treatment with lipid-lowering drugs as the cornerstone also restricts the efforts to reduce global AS burden.Stem cell-or nanoparticle-based strategies spurred a lot of attention due to the attractive therapeutic effects and minimized adverse effects.Given the complexity of pathological changes of AS,attempts to develop an almighty medicine based on single mechanisms could be theoretically challenging.In this review,the top stories in the cellular landscapes during the initiation and progression of AS and the therapies were summarized in an integrated perspective to facilitate efforts to develop a multi-targets strategy and fill the gap between mechanism research and clinical translation.The future challenges and improvements were also discussed.

Drug-induced liver injury(DILI)is a common adverse drug reaction,which can even result in liver failure[1,2].The Chinese Medical Association issued the Guidelines for the Diagnosis and Treatment of DILI based on the Roussel Uclaf Causality Assessment Method(RUCAM)in 2015[3].A previous study reported that traditional Chinese medicines(TCMs),herbal and dietary supplements,and antituberculosis drugs were the main causes of DILI in China[4].Herb-induced liver injury(HILI)refers to liver injury caused by TCMs,natural drugs,and their related preparations[5].

General variational solution for seismic and static active earth pressure on rigid walls considering soil tensile strength cut-off

According to the limit equilibrium state of soils behind rigid walls and the pseudo-static approach,a general closed-form solution to seismic and static active earth pressure on the walls,which considers shear and tension failure of the retained soil,is put forward using a variational calculus method.The application point of the active resultant force specified in the proposed method is explained with a clear physical meaning related to possible movement modes of the walls.In respect of the derived nine dependent equations reflecting the functional characteristics of the earth pressure,the proposed method can be performed easily via an implicit strategy.There are 13 basic factors related to the retained soils,walls,and external loads to be involved in the proposed method.The tension crack segment of the slip surface is obviously influenced by these parameters,apart from vertical seismic coefficient and geometric bounds of the surcharge,but the shear slip segment maintains an approximately planar shape almost uninfluenced by these parameters.Noticeably,the proposed method quantitatively reflects that the resultant of the active earth pressure is always within a limited range under different possible movements of the same wall.

Research on diagnosis-related group grouping of inpatient medical expenditure in colorectal cancer patients based on a decision tree model

BACKGROUND In 2018,the diagnosis-related groups prospective payment system(DRGs-PPS)was introduced in a trial operation in Beijing according to the requirements of medical and health reform.The implementation of the system requires that more than 300 disease types pay through the DRGs-PPS for medical insurance.Colorectal cancer(CRC),as a common malignant tumor with high prevalence in recent years,was among the 300 disease types.AIM To investigate the composition and factors related to inpatient medical expenditure in CRC patients based on disease DRGs,and to provide a basis for the rational economic control of hospitalization expenses for the diagnosis and treatment of CRC.METHODS The basic material and cost data for 1026 CRC inpatients in a Grade-A tertiary hospital in Beijing during 2014-2018 were collected using the medical record system.A variance analysis of the composition of medical expenditure was carried out,and a multivariate linear regression model was used to select influencing factors with the greatest statistical significance.A decision tree model based on the exhaustiveχ^2 automatic interaction detector(E-CHAID)algorithm for DRG grouping was built by setting chosen factors as separation nodes,and the payment standard of each diagnostic group and upper limit cost were calculated.The correctness and rationality of the data were re-evaluated and verified by clinical practice.RESULTS The average hospital stay of the 1026 CRC patients investigated was 18.5 d,and the average hospitalization cost was 57872.4 RMB yuan.Factors including age,gender,length of hospital stay,diagnosis and treatment,as well as clinical operations had significant influence on inpatient expenditure(P<0.05).By adopting age,diagnosis,treatment,and surgery as the grouping nodes,a decision tree model based on the E-CHAID algorithm was established,and the CRC patients were divided into 12 DRG cost groups.Among these 12 groups,the number of patients aged≤67 years,and underwent surgery and chemotherapy or radiotherapy was largest;while p

In this paper,we give a brief survey of recent results on axially symmetric Navier-Stokes equations(ASNS)in the following categories:regularity criterion,Liouville property for ancient solutions,decay and vanishing of stationary solutions.Some discussions also touch on the full 3 dimensional equations.Two results,closing of the scaling gap for ASNS and vanishing of homogeneous D solutions in 3 dimensional slabs will be described in more detail.In the addendum,two new results in the 3rd category will also be presented,which are generalizations of recently published results by the author and coauthors.

Heavy metal pollution is becoming a prominent stress on plants.Plants contaminated with heavy metals undergo changes in external morphology and internal structure,and heavy metals can accumulate through the food chain,threatening human health.Detecting heavy metal stress on plants quickly,accurately,and nondestructively helps to achieve precise management of plant growth status and accelerate the breeding of heavy metal-resistant plant varieties.Traditional chemical reagent-based detection methods are laborious,destructive,time-consuming,and costly.The internal and external structures of plants can be altered by heavy metal contamination,which can lead to changes in plants'absorption and reflection of light.Visible/near-infrared(V/NIR)spectroscopy can obtain plant spectral information,and hyperspectral imaging(HSI)can obtain spectral and spatial information in simple,speedy,and nondestructive ways.These 2 technologies have been the most widely used high-throughput phenotyping technologies of plants.This review summarizes the application of V/NIR spectroscopy and HSI in plant heavy metal stress phenotype analysis as well as introduces the method of combining spectroscopy with machine learning approaches for high-throughput phenotyping of plant heavy metal stress,including unstressed and stressed identification,stress types identification,stress degrees identification,and heavy metal content estimation.The vegetation indexes,full-range spectra,and feature bands identified by different plant heavy metal stress phenotyping methods are reviewed.The advantages,limitations,challenges,and prospects of V/NIR spectroscopy and HSI for plant heavy metal stress phenotyping are discussed.Further studies are needed to promote the research and application of V/NIR spectroscopy and HSI for plant heavy metal stress phenotyping.

Cucumber is an important vegetable worldwide,and powdery mildew(PM)is a common and serious disease of cucumbers.Breeding disease-resistant cucumber varieties is the most advantageous strategy to control this disease.In recent years,exploration and identification of cucumber PM resistance genes have achieved great advancement,and many genes have been cloned and verified using different methods.However,the resistance mechanism of cucumber PM is still unclear,and many ambiguities need to be elucidated urgently.In this review,we summarized the research advances in PM resistance in cucumbers,including genetic analysis,quantitative trait locus mapping,map-based cloning,transcriptomics,mlo-mediated PM resistance,and mining of noncoding RNAs involved in resistance.Finally,the research directions and the problems that need to be solved in the future were discussed.

Tuning Charge Transport of Oligopeptide Junctions via Interfacial Amino Acids

Comprehensive Summary,The charge transport through peptides can imitate the corresponding processes in more complicated proteins,enabling us to develop high-performance bioelectronic devices and to understand the mechanisms of biomolecular recognition and information transfer.While charge transport modulation through individual peptides has been achieved via various covalent strategies,the intermolecular modulation is still very challenging,which may capture the charge transport between proteins.To tackle this challenge,we used well-defined self-assembled monolayers(SAMs)of oligopeptides as a model to imitate the interface of proteins and explored an interfacial amino acid strategy for charge transport modulation.We showed that non-covalently interfaced charged amino acids(e.g.,arginine)effectively attenuated the charge transport of glutamic acid terminated polyglycine peptide SAMs.By analyzing the relationship of the charge transport with the molecular frontier orbital relative to the Fermi energy level of the electrode,the molecule-electrodes coupling(Γ),and the trends in skewness and kurtosis with voltage and the dielectric constant(εr),we showed that the attenuation was from the decreasedΓand the reduced polarizability.We present an efficient strategy to modulate the charge transport of oligopeptide-SAM junctions by intermolecular interactions,which will advance our understanding of charge transport in biological systems and facilitate developing future electronics.

Multivariable sales prediction for filling stations via GA improved BiLSTM

Accurate sales prediction in filling stations is the basis to fill in the refined oil in time and avoid the outof-stock as much as possible.Considering the defect of great“lag”in the general time series model,this paper summarizes the multiple factors that influence the oil sales and develops a multivariable long short-term memory(LSTM)neural network,with the hyper-parameters being improved by the genetic algorithm(GA).To further improve the prediction accuracy,the proposed LSTM neural network is generalized to bidirectional LSTM(Bi LSTM),in which the impact of future factors on present sales can be taken into account by backward training.Finally,different LSTM structures and genetic algorithm parameters are tested to discuss their impact on prediction accuracy.Results demonstrated that genetic algorithm improved Bi LSTM model is superior to extreme gradient boosting,ARIMA,and artificial neural network,having the highest accuracy of 89%.
