Platelets are small, versatile blood cells that are critical for hemostasis/thrombosis. Local platelet accumulation is a known contributor to proinflammation in various disease states. However, the anti-inflammatory/immunosuppressive potential of platelets has been poorly explored. Here, we uncovered, unexpectedly, desialylated platelets (dPLTs) down-regulated immune responses against both platelet-associated and -independent antigen challenges. Utilizing multispectral photoacoustic tomography, we tracked dPLT trafficking to gut vasculature and an exclusive Kupffer cell-mediated dPLT clearance in the liver, a process that we identified to be synergistically dependent on platelet glycoprotein Ibα and hepatic Ashwell–Morell receptor. Mechanistically, Kupffer cell clearance of dPLT potentiated a systemic immunosuppressive state with increased anti-inflammatory cytokines and circulating CD4+ regulatory T cells, abolishable by Kupffer cell depletion. Last, in a clinically relevant model of hemophilia A, presensitization with dPLT attenuated anti-factor VIII antibody production after factor VIII ( infusion. As platelet desialylation commonly occurs in daily-aged and activated platelets, these findings open new avenues toward understanding immune homeostasis and potentiate the therapeutic potential of dPLT and engineered dPLT transfusions in controlling autoimmune and alloimmune diseases.

Unexpected Exposure Risks to Emerging Aromatic Amine Antioxidants and p‑Phenylenediamine quinones to Residents:Evidence from External and Internal Exposure as Well as Hepatotoxicity Evaluation

Aromatic amine antioxidants(AAs),extensively used in rubber products,and their ozone-photolyzed product pphenylenediamine quinones(PPD-qs)were found to be pervasive in various environments and with potential ecological and health *** relationship between external and internal exposure to these contaminants and their exposure risks to the general population remain poorly ***,dust−handwipe−urine samples(n=97 pairs)from families in Tianjin,China was collected for analyzing 20 AAs and 6 PPD-qs(including self-synthesized emerging PPD-qs).The median concentration ofΣ_(20)AAs(1310 ng/g,354 ng/m^(2),and 18.5 ng/mL,respectively)andΣ_(6)PPD-qs(1645 ng/g,3913 ng/m^(2)and 3.76 ng/mL,respectively)in dust,handwipe,and urine samples indicated a non-negligible contamination of AAs and PPD-qs in indoor ***,emerging DPPD-q and DNPD-q(oxidation products of DPPD and DNPD,respectively)were prominent,accounting for>90%of *** correlation analysis and estimated daily intake(EDI)assessments indicated that neither dust ingestion nor dermal exposure may be the primary source of internal exposure to AAs and PPD-qs in the general population,and other exposure pathways(e.g.,diet and inhalation)may *** vitro HepG2 cell experiments demonstrated that both PPD-qs and PPDs may have potential exposure risk of hepatotoxicity.

Organic solar cells(OSCs)have emerged as a promising solution for sustainable energy production,offering advantages such as a low carbon footprint,short energy payback period,and compatibility with ***,the use of hazardous solvents continues to dominate the best-performing OSCs,mainly because of the challenges of controlling phase separation and domain crystallinity in *** this study,we combined the solvent vapor treatment of CS2 and thermal annealing to precisely control the phase separation and domain crystallinity in PM6:M-Cl and PM6:O-Cl systems processed with the eco-solvent *** method resulted in a maximum power conversion efficiency(PCE)of 18.4%,which is among the highest values reported for sustainable binary ***,the fabrication techniques were transferred from spin coating in a nitrogen environment to blade printing in ambient air,retaining a PCE of 16.0%,showing its potential for high-throughput and scalable *** addition,a comparative analysis of OSCs processed with hazardous and green solvents was conducted to reveal the differences in phase *** work not only underscores the significance of sustainability in OSCs but also lays the groundwork for unlocking the full potential of open-air-printable sustainable OSCs for commercialization.

The clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated gene(Cas) system is continually optimized to achieve the most efficient gene editing effect. The Cas12i^(Max), a Cas12i variant, exhibits powerful DNA editing activity and enriches the gene editing toolbox. However, the application of Cas12i^(Max)in large domestic animals has not yet been reported. To verify the efficiency and feasibility of multiple gene editing in large animals, we generated porcine fibroblasts with simultaneous knockouts of IGF2, ANPEP, CD163,and MSTN via Cas12i^(Max)in one step. Phenotypically stable pigs were created through somatic cell nuclear transfer technology. They exhibited improved growth performance and muscle quality. Furthermore, we simultaneously edited three genes in bovine fibroblasts. A knockout of MSTN and PRNP was created and the amino acid q-G in CD18 was precisely substituted. Meanwhile, no off-target phenomenon was observed by sum-type analysis or off-target detection. These results verified the effectiveness of Cas12i^(Max)for gene editing in livestock animals and demonstrated the potential application of Cas12i^(Max)in the field of animal trait improvement for agricultural production.

The phylogenetic relationships of introduced Aphelinus （Hymenoptera： Aphelinidae）, biological control agents of the Russian wheat aphid （Homoptera： Aphididae）

Several species of Aphelinus have been introduced to the US from the Old World for biological control of the Russian wheat aphid, Diuraphis noxia （Modvilko）. Reproductive incompatibility has been observed among populations collected from different geographic areas. We examined whether or not the reproductive incompatibility between strains of A. asychis was caused by distant phylogenetic relationships. Ribosomal DNA sequences of internal transcribed spacers 2 （ITS2） were analyzed in several species of Aphelinus collected from multiple sites of Europe and Asia. Yhe phylogenetic analysis showed that strains within the species A. albipodus and A. asychis are not monophyletic, and two clearly divergent clades were revealed among sequenced samples. Our results suggest that the reproductive incompatibility between three exotic strains ofA. asychis was more likely caused by divergence of phylogeny than by symbiotic bacteria.

地震高分辨率处理或反演的目的是获得精确的反射系数或弹性参数模型。然而地层滤波效应模糊了地震记录中的地层反射信息,因此有必要消除这种滤波效应。现有的大部分提高分辨率方法并不能完全摒弃关于地震子波和q模型的一些假设。为了获得更为切合实际的地震子波,同时自适应获取q模型,文中将地震子波相位估计与q模型估计相结合,提出了基于遗传算法的非稳态地震数据混合相位子波及q值估计方法。首先通过井旁道记录拟合得到初始地震子波的振幅信息,然后依据子波Z变换的根关于单位圆移动与否构建用于遗传算法的编码链条。另一方面,十进制q模型对应的二进制表示形式同样能够利用编码链条表征,因此利用全局优化算法能够同时估计地震混合相位子波以及q模型。结合根变换和遗传算法不断改变子波相位的同时自适应生成q模型,利用子波和q模型构造的时变子波矩阵与测井反射系数得到合成地震记录,并与井旁道记录进行匹配,最终得到合理的混合相位子波和地层q模型,进而构造时变子波矩阵进行时变反褶积。通过井旁道记录与测井数据拟合得到的混合相位子波,与实际地震子波相位更为接近,理论数据和实际数据处理结果证实了该方法的有效性。

Non-alcoholic fatty liver degree and long-term risk of incident inflammatory bowel disease: A large-scale prospective cohort study

Background: Non-alcoholic fatty liver disease (NAFLD) and inflammatory bowel disease (IBD) have shown similar worsening epidemic patterns globally and shared various overlapping pathophysiological mechanisms. However, evidence on the relationship between NAFLD and IBD risk is lacking. We aimed to investigate the associations between long-term risk of incident IBD and NAFLD in a large prospective ***: Participants from the United Kingdom Biobank cohort (https://***/) who were free of IBD and alcoholic liver disease at baseline were enrolled. Baseline non-alcoholic fatty liver degree was measured by the well-established fatty liver index (FLI). The outcomes of interest included incident IBD, ulcerative colitis (UC), and Crohn’s disease (CD). Multivariable Cox proportional hazard regression was used to calculate hazard ratios (HRs) and 95% confidence intervals (CIs).Results: Among 418,721 participants (mean FLI: 48.11 ± 30.11), 160,807 (38.40%) participants were diagnosed as NAFLD at baseline. During a median of 12.4 years’ follow-up, 2346 incident IBD cases (1545 UC, 653 CD, and 148 IBD-unclassified) were identified. Due to limited events, those IBD-unclassified were combined in UC or CD when examining the associated risk of UC or CD, separately. Compared with the lowest quartile of FLI, the highest quartile showed a separately 36.00%, 25.00%, and 58.00% higher risk of incident IBD (HRq4vs. q1 =1.36, 95% CI: 1.19-1.55,Ptrend q4vs. q1 =1.25, 95% CI: 1.07-1.46,Ptrend=0.047), and CD (HRq4vs. q1 =1.58, 95% CI: 1.26-1.97,Ptrend <0.001) after multivariable adjustment. Compared with non-NAFLD, NAFLD participants had a significantly higher risk of incident IBD (HR=1.13, 95% CI: 1.04-1.24) and CD (HR =1.36, 95% CI: 1.17-1.58).Conclusions: Higher degree of non-alcoholic fatty liver is associated with increased risk of incident IBD.
