AIM: To explore the role of the adaptor molecule in liver regeneration after partial hepatectomy (PH). METHODS: We used transgenic mice expressing an N-terminal truncated form of MOrT1/FADD under the control of the albumin promoter. As previously shown, this transgenic protein abrogated CD95- and CD120a-mediated apoptosis in the liver. Cyclin A expression was detected using Western blotting. ELISA and rT-PCr were used to detect IL-6 and IL-6 mrNA, respectively. DNA synthesis in liver tissue was measured by BrdU staining. rESULTS: resection of 70% of the liver was followed by a reduced early regenerative response in the transgenic group at 36 h. Accordingly, 36 h after hepatectomy, cyclin A expression was only detectable in wild-type animals. Consequently, the onset of liver mass restoration was retarded as measured by MrI volumetry and mortality was significantly higher in the transgenic group. CONCLUSION: Our data demonstrate for the first time an involvement of the death receptor molecule MOrT1/ FADD in liver regeneration, beyond its well described role as part of the intracellular death signaling pathway.

Micro-recanalization in a biodegradable graft for reconstruction of the vas deferens is enhanced by sildenafil citrate

This study investigated the effect of sildenafil citrate on micro-recanalization and neovascularization, which were previously demonstrated in a rat model using biodegradable grafts （BGs） for vas deferens reconstruction. A total of 24 male rats underwent bilateral vasectomy with removal of a 0.5-cm vasal segment and were randomly assigned to four groups. Groups 1 and 2 underwent immediate vasovasostomy. Groups 3 and 4 underwent interposition of a 0.5-cm BG in the vasal gap. Groups 1 and 3 were given 5 mg kg-1 day-1 oral sildenafil. Other groups were given placebo. rats were housed with females 12 weeks postoperatively. reconstructed vasal segments were harvested 16 weeks postoperatively and analyzed histologically. Fluid from the distal vasal stump was analyzed for motile sperm. Urine samples obtained 16 weeks postoperatively were analyzed for cGMP levels. cGMP levels in rats treated with sildenafil were signifcantly higher than in control rats. No pregnancies were sired by grafted groups. In all, 5/6 rats in group 1 and 3/6 rats in group 2 sired litters. No motile sperm were noted in the vasal fluid of the grafted groups. Motile sperm were noted in all rats in group 1 and in 5/6 rats in group 2. In addition, 29 and 4 microcanals were detected in the sildenafil and placebo groups, respectively （P = 0.023）. No microcanal exceeded 3 mm in length. An average of 12 and 28 blood vessels per graft were noted in the placebo and sildenafil groups, respectively （P 〈 0.0001）. In conclusion, sildenafil enhances micro-recanalization and neovascularization in BG used for vas deferens reconstruction, but does not increase the microcanal length after 16 weeks.

A data-driven framework to predict the morphology of interfacial Cu6Sn5 IMC in SAC/Cu system during laser soldering

A data-driven approach combining together the experimental laser soldering,finite element analysis and machine learning,has been utilized to predict the morphology of interracial intermetallic compound(IMC) in Sn-xAg-yCu/Cu(SAC/Cu) *** types of SAC solders with varying weight proportion of Ag and Cu,have been processed with fiber laser at different magnitudes of power(30-50 W) and scan speed(10-240 mm/min),and the resultant IMC morphologies characterized through scanning electron microscope are categorized as prismatic and scalloped *** the different alloy composition and laser parameters,finite element method(FEM) is employed to compute the transient distribution of temperature at the interface of solder and *** FEM-generated datasets are supplied to a neural network that predicts the IMC morphology through the quantified values of temperature dependent Jackson parameter(αJ).The numerical value of αJ predicted from neural network is validated with experimental IMC *** critical scan speed for the morphology transition between prismatic and scalloped IMC is estimated for each solder composition at a given ***-0.7 Cu having the largest critical scan speed at 30 W and Sn-3.5 Ag alloy having the largest critical scan speed at input power values of 40 W and 50 W,thus possessing the greatest likelihood of forming prismatic interfacial IMC during laser soldering,can be inferred as most suitable SAC solders in applications exposed to shear loads.

Health Status Inequality among Immigrants in Switzerland

Objective: To assess self-rated health and impairments for six large immigrant groups (Germany, Italy, Kosovo, Portugal, Serbia, Turkey) in Switzerland. Methods: We used population-based survey data from the Swiss Migrant Health Survey 2010 and the Swiss Health Survey 2007. The sample comprised permanent residents aged 17 - 64 years (n = 14,637). Multivariate logistic regressions have been used to estimate odds ratios (Or). results: Ill health and activities of daily living (ADL) impairments were associated with older age in all groups. However, nationals from Turkey and nationals from Kosovo were substantially more likely than Swiss to report ill health (Or = 1.05;CI = 1.02 - 1.09;P = 0.001 and Or = 1.05;CI = 1.01 - 1.10;P = 0.016) and ADL impairments (Or = 1.06;CI = 1.03 - 1.09;P = 0.000 and Or = 1.04;CI = 1.01 - 1.07;P = 0.004) with increasing age. Furthermore, Portuguese women were more likely (Or = 2.65;CI = 1.40 - 5.03;P = 0.003) to report ill health than Swiss women. Conclusions: Immigrant-specific preventive and health promotion initiatives should target vulnerable immigrants from Turkey, Portugal, and Kosovo. Furthermore, groups with few economic and psychosocial resources in the general population of Switzerland should be more involved in interventions to reduce health risk.

New perspectives on exploitation of incretin peptides for the treatment of diabetes and related disorders

The applicability of stable gut hormones for the treatment of obesity-related diabetes is now undisputable. This is based predominantly on prominent and sustained glucoselowering actions, plus evidence that these peptides can augment insulin secretion and pancreatic islet function over time. This review highlights the therapeutic potential of glucagon-like peptide-1(GLP-1), glucose-dependent insulinotropic polypeptide(GIP), oxyntomodulin(OXM) and cholecystokinin(CCK) for obesity-related *** GLP-1 mimetics have already been successfully adopted into the diabetic clinic, whereas GIP, CCK and OXM molecules offer promise as potential new classes of antidiabetic drugs. Moreover, recent studies have shown improved therapeutic effects following simultaneous modulation of multiple receptor signalling pathways by combination therapy or use of dual/triple agonist peptides. However, timing and composition of injections may be important to permit interludes of beta-cell rest. The review also addresses the possible perils of incretin based drugs for treatment of prediabetes. Finally, the unanticipated utility of stable gut peptides as effective treatments for complications of diabetes, bone disorders, cognitive impairment and cardiovascular dysfunction is considered.

risk of overweight and obesity among migrants in Switzerland

We assessed the risk of overweight and obesity for six large migrant groups in Switzerland. We used population-based survey data form the Swiss Migrant Health Survey 2010 and the Swiss Health Survey 2007. The sample comprised permanent residents aged 17 - 64 years (n = 14,637). Multivariate logistic regressions have been used to estimate odds ratios (Or). Compared with Swiss nationals, the odds of being obese were 1.97 times higher for Turkish nationals (CI-95 1.26, 3.07), 1.73 times higher for Kosovan nationals (CI-95 1.09, 2.74) and 3.42 times higher for Serbs (CI-95 2.28, 5.12). The chance of being obese was not statistically different between Swiss nationals and those from Portugal, Italy, and Germany. The chance of being overweight or obese (BMI ≥ 25) was higher for migrants than for Swiss nationals. The respective odds were 1.54 times higher for nationals from Portugal (CI-95 1.13, 2.11), 2.05 times higher for those from Turkey (CI-95 1.50, 2.78), 2.67 times higher for those from Serbia (CI-95 1.93, 3.68), 2.68 times higher for those from Kosovo (CI-95 2.01, 3.56), and 2.16 times higher for nationals from Italy (CI-95 1.54, 3.02). We suggest that preventive initiatives should take into account the specific needs of migrants as well as the needs of persons with low education from all nationalities.

Update on autoimmune hepatitis

Autoimmune hepatitis (AIH) is a necroinflammatory liver disease of unknown etiology that occurs in children and adults of all ages. Characteristics are its autoimmune features,hyperglobulinemia (IgG),and the presence of circulating autoantibodies,as well as a response to immunosuppressant drugs. Current treatment consists of prednisone and azathioprine and in most patients this disease has become very treatable. Over the past 2 years,a couple of new insights into the genetic aspects,clinical course and treatment of AIH have been reported,which will be the focus of this review. In particular,we concentrate on genome-wide microsatellite analysis,a novel mouse model of AIH,the evaluation of a large AIH cohort for overlap syndromes,suggested novel criteria for the diagnosis of AIH,and the latest studies on treatment of AIH with budenoside and mycophenolate mofetil.

Multiple reaction monitoring for the detection of disease-related synaptic proteins

Synaptic dysfunction occurs early in Alzheimer＇s disease （AD） and is acknowledged as a primary pathologic target for treatment. Synaptic degeneration is the pathological feature most strongly correlated with loss of cognitive function ante mortern （Terry et al., 1991）. Synapses are heavily damaged in hippocampal and neocortical regions of AD brain, whereas motor and occipital cortices are relatively spared （Honer et al., 1992）. Despite extensive work, the molecular mechanisms underlying synaptic degeneration are largely unknown.

Dynamics and origin of cytokinins involved in plant manipulation by a leaf-mining insect

Several herbivorous insects and plant-associated microorganisms control the phytohormonal balance, thus enabling them to successfully exploit the plant by inhibiting plant defenses and withdrawing plant resources for their own benefit. The leaf-mining moth Phyllonorycter blancardella modifies the cytokinin （CK） profile of mined leaf-tissues, and the insect symbiotic bacteria Wolbachia is involved in the plant manipulation to the benefit of the insect host. To gain a deeper understanding into the possible origin and dynamics of CKs, we conducted an extensive characterization of CKs in larvae and in infected apple leaves. Our results show the enhanced CK levels in mines, both on green and yellow leaves, allowing insects to control their nutritional supply under fluctuating environmental conditions. The spatial distribution of CKs within the mined leaves shows that hormone manipulation is strictly limited to the mine suggesting the absence of CK translocation from distant leaf areas toward the insect feeding site. Mass spectrometry analyses reveal that major CK types accumulating in mines and larvae are similar to what is observed for most gall-inducers, suggesting that strategies underlying the plant manipulation may be shared between herbivorous insects with distinct life histories. results further show that CKs are detected in the highest levels in larvae, reinforcing our hypothesis that CKs accumulating in the mines originate from the insect itself. Presence of bacteria- specific methylthio-CKs is consistent with previous results suggesting that insect bacterial symbionts contribute to the observed phenotype. Our study provides key findings toward the understanding of molecular mechanisms underlying this intricate plant-insect-microbe interaction.
