Background： Recent studies suggest that dietary transfatty acids （TFAs） at relative high levels （i） increase the risk of coronary heart disease, （ii） inhibit the metabolism of linoleic acid and consequently increase requirement for essential fatty acids （EFAs）. The aim of this prospective study was to estimate TFAs the placental transfer of TFAs to the foetus cord blood and cord tissues in France. Material： TFAs consumption was measured in 59 mother-foetus couples. TFAs transferring across the placenta were estimated by comparative measurement （Capillary Gaz Chromatography） of TFAs in mother＇s blood lipids （n = 59）, cord blood lipids and cord tissue plasma lipids （n = 25） cord red blood tissue （n = 25） and umbilical vessels （n = 15）. Results-Discussion： TFA deposition in cord blood is 0.58% slightly lower than mother level. TFAs incorporation in total lips of cord show selectivity of transfer with lower 18：1 t in cord blood and preferential transfer of diene 18：2 tc in cord blood. There is competition between the 18：2 tc in cholesterol esters （CE） with the linolenic acid and a negative correlation in PLT of arterial tissues with the 18：2 tc and C20：4 n-6 or arachidonic acid （AA）, then there is the same competition in PE of venous tissue. But there is no effect on growth in our population of term newborn. Conclusions： Our results confirm the placental TFAs transfer, a better incorporation in the fetus CE; we demonstrated a selective transfer for the 18：2 9 trans 12 cis （18：2 tc） and a negative correlation （r = -0.76） with the linoleic acid and AA （r = 0.98）. This competition, and the presence of these TFAs in cord tissues, even at a low TFAs consumption, remind us to be a potential risk for the fetus concerning EFA metabolism and growth.

Metyrapone (2-methyl-1,2-di-3-pyridyl-1-propanone) is a drug largely used as inhibitor of glucocorticoid synthesis. Although its binding to various proteins has been well indentified, its accurate molecular mechanism of action remains unknown. Therefore, the interactions of metyrapone (MET) with various membrane components such as phospholipids, cholesterol, their corresponding polar heads and a model serine containing peptide have been investigated by NMR and ESR methods. It was found that neither cholesterol nor most of the phospholipids tested, nor dimyristin exhibit any interaction with MET, except phosphatidylserine (DMPS). Furthermore, only serine bearing polar head (O-phosphoserine) showed an association with MET (stoechiometry 1:1, Kd = 3200M-1). As similar observations were also performed on serine alone and in the presence of the serine containing model peptide, (NASDSDGQDL), a possible implication of these interactions in the binding recognition of MET on serine-containing active site was finally tested and discussed.

The combined influence of maturation,ripening,and climate on the profile of bioactive compounds was studied in banana(Musa acuminata,AAA,Cavendish,*** Naine).Their bioactive compounds were determined by the Folin-Ciocalteu assay and high-performance thin layer chromatographic(HPTLC) *** polyphenol content of bananas harvested after 400 degree days remained unchanged during ripening,while bananas harvested after 600 and 900 degree days exhibited a significant polyphenol *** dopamine was the polyphenol with the highest concentration in banana peels during the green developmental stage and ripening,its kinetics differed from the total polyphenol *** results showed that this matrix of choice(maturation,ripening,and climate) may allow selection of the banana(***,AAA,Cavendish,*** Naine) status that will produce optimal concentrations of identified compounds with human health relevance.

Treatment-resistant schizophrenia has an extremely negative impact on mental health and social life. If clozapine, the gold standard treatment, fails, there are very few options left. The literature suggests that high-dose olanzapine (20 - 60 mg/day) is a possible alternative. We report two cases in which very high doses of olanzapine were administered, with significant clinical improvements above 60 mg/day. Clinical, metabolic and cardiac tolerance was good. This report highlights the usefulness of very-high-dose olanzapine in treatment-resistant schizophrenia. The main hypotheses concerning the psychopharmacological mechanisms of very-high-dose olanzapine are discussed.

A recent technique, using to potentially replace the conventional impregnating one in low voltage electric motors, has been studied in this communication. The conventional technique that uses?both primary and secondary insulations;?i.e.: enamel and varnish, has been compared to this recent technique which only uses one component,?i.e.: self-bonding wires. Self-bonding wires polymerization is very quick compared to that of the conventional technique. Environmental impact and both dielectric and mechanical properties of these two techniques have been estimated and compared. The goal is to estimate if this recent technique has not only a better environmental footprint but also good technical properties. The dielectric properties that have been measured are the dielectric strength and?the lifetime under pulse?voltage while mechanical consists in measuring the bonding strength that is crucial for impregnation.?For that purpose, a Life Cycle Assessment (LCA) that can simultaneously evaluatenumerous impacts on ground, water and air,?has been performed. The results show clearly that the recent technique significantly reduces the environmental footprint. Both mechanical and dielectric properties are then compared and analyzed.

Background and Aims: To better understand nonalcoholic steatohepatitis (NASH) disease progression and to evaluate drug targets and compound activity, we undertook the devel-opment of an in vitro 3D model to mimic liver architecture and the NASH environment. Methods:We have developed an in vitro preclinical 3D NASH model by coculturing primary hu-man hepatocytes, human stellate cells, liver endothelial cells and Kupffer cells embedded in a hydrogel of rat collagen on a 96-well plate. A NASH-like environment was induced by ad-dition of medium containing free fatty acids and tumor ne-crosis factor-α. This model was then characterized by biochemical, imaging and transcriptomics analyses. Results:We succeeded in defining suitable culture conditions to main-tain the 3D coculture for up to 10 days in vitro, with the lowest level of steatosis and reproducible low level of inflammation and fibrosis. NASH disease was induced with a custom me-dium mimicking NASH features. The cell model exhibited the key NASH disease phenotypes of hepatocyte injury, steatosis, inflammation, and fibrosis. Hepatocyte injury was highlighted by a decrease of CYP3A4 expression and activity, without loss of viability up to day 10. Moreover, the model was able to stimulate a stable inflammatory and early fibrotic environ-ment, with expression and secretion of several cytokines. A global gene expression analysis confirmed the NASH induc-tion. Conclusions:This is a new in vitro model of NASH dis-ease consisting of four human primary cell-types that exhibits most features of the disease. The 10-day cell viability and cost effectiveness of the model make it suitable for medium throughput drug screening and provide attractive avenues to better understand disease physiology and to identify and characterize new drug targets.
