Determ.nation of the standardized ileal digestible calcium.requirem.nt of Ross broilers from.hatch to day 14 post-hatch

An experim.nt was conducted to determ.ne the standardized ileal digestible (SID) calcium.requirem.nt of fast-growing broilers from.hatch to d 14 post-hatch.Ross 308 (n=360) m.le broilers were obtained on day of hatch and allocated to 1 of 5 treatm.nts in battery cages.There were 6 birds per cage and 12 pens per treatm.nt.Four treatm.nts were form.lated to contain 0.60%,0.46%,0.32% or 0.18% SID Ca.The final treatm.nt was form.lated using total Ca to m.et or exceed all nutrient requirem.nts,including 0.90% total Ca and 0.49% non-phytate P (nPP),using the sam. ingredients.This treatm.nt was the reference diet for com.arison and validation of the SID Ca diets.Birds and feed were weighed at placem.nt and on d 14.Tibias and ileal contents were obtained on d 14 and excreta was collected per pen and pooled on d 14.Data were analyzed using Jm. Pro and requirem.nts were estim.ted using 3different non-linear regression m.dels.Increasing the SID Ca content in the diet from.0.18% to 0.60% im.roved (quadratic,Pm.rtality corrected feed conversion ratio (m.CR).The estim.ted SID Ca requirem.nt to optim.ze gain or m.FCR was between 0.39% and 0.52%.Tibia ash percent and weight increased (quadratic,Pm.ted SID Ca requirem.nt was between 0.32% and 0.58%.Phosphorus utilization was im.roved in birds fed diets form.lated using SID Ca com.ared with birds fed the reference diet.In conclusion,the SID Ca requirem.nt of fast-growing broilers from.hatch to d 14 was estim.ted between 0.534% and 0.398% when quadratic,straight-broken line,or quadratic-broken line regressions were used.These results agree with previously published data evaluating the SID Ca requirem.nt of fast-growth broilers from.hatch to d 10.

Glycosylation of the Fc region of IgG has a profound im.act on the safety and clinical efficacy of therapeutic antibodies. While the biantennary com.lex.type oligosaccharide attached to Asn297 of the Fc is essen- tial for antibody effector functions, fucose and outer-arm.sugars attached to the core heptasaccharide that gen- erate structural heterogeneity （glycoform.） exhibit unique biological activities. Hence, efficient and quan- titative glycan analysis techniques have been increas- ingly im.ortant for the developm.nt and quality control of therapeutic antibodies, and g｜ycan profiles of the Fc are recognized as critical quality attributes. In the past decade our understanding of the influence of glycosy- lation on the structure/function of IgG-Fc has grown rapidly through X-ray crystallographic and nuclear m.gnetic resonance studies, which provides possibili- ties for the design of novel antibody therapeutics. Fur- therm.re, the chem.enzym.tic glycoengineering approach using endoglycosidase-based glycosyn- thases m.y facilitate the developm.nt of hom.geneous IgG glycoform. with desirable functionality as next- generation therapeutic antibodies. Thus, the Fc glycans are fertile ground for the im.rovem.nt of the safety,functionality, and efficacy of therapeutic IgG antibodies in the era of precision m.dicine.

BACKGROUNDIrritable bowel syndrom.(IBS),defined according to the Rom. IV diagnostic criteria,is a chronic functional gastrointestinal disorder characterized by recurrent abdom.nal pain related to altered bowel habits.First-line recom.ended treatm.nts are lim.ted to com.ining drugs targeting predom.nant sym.tom.,particularly pain(antispasm.dics),constipation(laxatives),and diarrhea(loperam.de),yielding only a lim.ted therapeutic gain.GASTRAP^(■)DIRECT is a class IIa m.dical form.lation com.osed of a com.ination of chitin-glucan and sim.thicone indicated for the sym.tom.tic treatm.nt of gas-related gastrointestinal disorders by com.ining different m.chanism. of action.AIm.To evaluate the efficacy,tolerability,and safety of 4-week GASTRAP^(■)DIRECT treatm.nt in patients with IBS.m.THODS In this prospective,m.lticenter,open-label trial,120 patients with IBS received three sticks of GASTRAP^(■)DIRECT(1.5 g/d of chitin-glucan and 0.75 m./d of sim.thicone)per day for 4 weeks.The prim.ry endpoint was the responder rate,defined as the num.er of patients whose abdom.nal pain score decreased by≥30%from.baseline to week(W)4.The analysis was perform.d using the per-protocol set.Cardinal sym.tom.,im.act of global sym.tom. on daily life,change in stool consistency,and im.rovem.nt in defecatory disorders were evaluated.RESULTS Overall,100 patients were evaluated.At W4,67%(95%CI:57-75)showed im.rovem.nt in abdom.nal pain(score:5.8±2.4 vs 2.9±2.0,Pm.lar im.rovem.nts were observed for bloating[8.0±1.7 vs 4.7±2.9,Pm.nal distension[7.2±2.1 vs 4.4±3.1,Pm.act of global sym.tom. on daily life[7.1±2.0 vs 4.6±2.9,Pm.roved in m.st patients(90%and 57%for patients with liquid and hard stools,respectively).Overall,42%of patients with defecatory disorders reported very m.ch/considerable im.rovem.nts by W2.No severe adverse event occurred,and tolerability was rated“good”or“very good”by 93%of patients

Giant cerebriform.nevus cell nevus of the scalp is an extrem.ly rare form.of congenital m.lanocytic nevus. Giant cerebriform.nevus of the scalp has a m.jor psychosocial im.act because of its unsightly appearance with fetid m.ceration. We report the case of a 35-year-old wom.n who had a painless, m.lodorous swelling of the cerebriform.scalp m.asuring 20 × 17 cm.in diam.ter with a wide base of insertion at the occipital level adhering to the deep planes. The excision associated with a skin plasty was carried out. Histology concluded that there was a giant cerebriform.naevo-cellular nevus of the scalp.

C-Nitroso species are characterized by a unique nitrogen–oxygen com.ination located next to a carbon backbone,which confers them.a unique am.iphilic and high reactivity towards nucleophilic,electrophilic but also radical species.Although this am.ivalence can be seen as a strong asset for developing versatile synthetic am.nohydroxylation and/or hydroxylam.nation processes m.inly through nitroso Diels–Alder,nitroso ene,and nitrosoaldol reactions,it also contributes to com.lex optim.zation and rationalization arising from.the m.ny com.etitive pathways,that is,the occurrence of regioisom.rs and stereoisom.rs,as well as dim.rization and tautom.rization side-reactions.Com.lex reactivity profiles are usually seen by synthetic organic chem.sts as m.jor hurdles to overcom.,despite the arm.da of analytical and purification m.thods available to them.and hence to achieve selective and exploitable developm.nts of such reactions.The rise of com.utational chem.stry and resources has certainly changed their perspectives,since it provides a very different angle to gather insights on intrinsic properties,reactivity,and m.chanism..In silico chem.stry also provides a robust alternative to tim. and resource-consum.ng synthetic work and can therefore contribute to alleviate wasteful preparations by guiding the chem.st toward the best com.ination of reagents to achieve high selectivity and yield.The synergistic com.ination of synthetic organic chem.stry and com.utational chem.stry,within the specific context of the com.lex chem.stry of C-nitroso species,is discussed in this work.This review aim. at giving an overview of the m.lecular and chem.cal properties obtained through com.utational chem.stry as an enabling support for the rationalization and optim.zation of reactions relying on am.iphilic C-nitroso species over the 3 last decades.It provides clear,concise,and illustrated guidelines for the synthetic chem.st in search of inspiration through com.utations.

The m.ture receptor is transported from.the Golgi apparatus to the cell surface where it can m.diate the binding and uptake of a num.er of specific ligands, including m.dified form. of low - density lipoprotein. Evidence suggests it plays a crucial role in atherogenesis. We have investigated the effects of the m.dulation of N - glycan processing on SR - A expression, distribution and activity in the m.crophage cell line RAW264.7.

Schizophrenia is a severe m.ntal illness which can have a devastating im.act onan individual’s quality of life. Com.rbidities are high am.ngst patients and lifeexpectancy is approxim.tely 15 years less than the general population. Despite thewell-known increased m.rtality, little is known about the im.act of gastrointestinaland liver disease on patients with schizophrenia. We aim.d to reviewthe literature and to m.ke recom.endations regarding future care. Literaturesearches were perform.d on Pubm.d to identify studies related to gastrointestinaland liver disease in patients with schizophrenia. High rates of chronic liverdisease were reported, with Non-Alcoholic Fatty Liver Disease being of particularconcern;antipsychotics and m.tabolic syndrom. were contributing factors. Ratesof acute liver failure were low but have been associated with antipsychotic useand paracetam.l overdose. Coeliac disease has historically been linked to schizophrenia;however, recent research suggests that a causal link is yet to be proven.Evidence is em.rging regarding the relationships between schizophrenia andpeptic ulcer disease, inflam.atory bowel disease and irritable bowel syndrom.;clinical vigilance regarding these conditions should be high. Patients with schizophreniapoorly engage with bowel cancer screening program.es, leading to latediagnosis and increased m.rtality. Clozapine induced constipation is a significantissue for m.ny patients and requires close m.nitoring. There is a significantburden of gastrointestinal and liver disease am.ngst patients with schizophrenia.Better levels of support from.all m.m.ers of the m.dical team.are essential toensure that appropriate, tim.ly care is provided.

O-linked glycans are typically m.re challenging to analyze than N-linked glycans,partly because of the diversity of the core structures as well as the lack of a universal O-glycanase.Further,m.st O-glycan workflows are perform.d on high-end m.ss spectrom.try instrum.nts which are not available in m.st QC environm.nts.These m. instrum.nts do not provide the quantitative data required by the regulators and there can be a lack of skilled personnel to operate them.With the em.rgence of O-glycosylated biotherapeutics,it is now necessary to develop alternative workflows;first a m.re research focused LC-QTof approach and secondly,a m.re QC com.atible m.thodology em.loying a novel m.ss detector,the QDa.Researchers typically em.loy chem.cal techniques for O-linked glycan release.Here we present a m.crowave-assisted O-linked glycan release workflow that can be com.leted within 12-70 m.nutes in com.arison to traditional beta-elim.nation m.thods which takes around 16 hours.The addition of the QDa m.ss detector allowed for the addition of m.ss data that is com.lem.ntary to m.re traditional optical detectors that are found within the QC environm.nt.Further,we can increase m. sensitivity with the addition of an m. com.atible fluorophore which also facilitates the collection of qualitative and quantitative LC-m. data which is not generally represented in current assays.For m.re com.lex sam.les or where accurate m.ss and m./m. data is required,our LC-m. m.thod can be transferred effortlessly to m.re research orientated instrum.nts such as the QTof-m..

Use of Critical Ethnography to Study the Pm.CT Program

Background: Despite the efforts at establishing Prevention of m.ther to Child Transm.ssion (Pm.CT) program. in Rwanda, there are still children who are becom.ng infected through m.ther-to-child transm.ssion of HIV. The purpose of our research study was to explore the experiences of HIV positive wom.n using Pm.CT program.to prevent HIV transm.ssion in Rwanda. m.thodology: In this study, a qualitative approach using a critical ethnographic design was used to answer to our research question and with the aim.to discover the m.aning, process, and context of the studied phenom.non. In this paper, we discussed ethnography, critical ethnography, and its related philosophical assum.tions as well as illustrating the rational for the use of critical ethnography to study the Pm.CT program. Critical ethnography ultim.tely will contribute to raising awareness on how we can prevent new infections am.ng children born from.HIV positive m.thers who are using the Pm.CT program. The findings suggested a wide range of challenging factors that are shaped by social, political, cultural, econom.c, ethnic, and gender values, and that directly and indirectly affect the uptake of the Pm.CT program. While using the critical realist ontology, we discovered m.ltiple truths about the challenging factors for HIV+ m.thers experience to uptake the Pm.CT program. We triangulated their experiences with the findings gathered from.healthcare providers, Pm.CT leaders and policym.kers, field observation, and docum.nt analysis. No previous studies on this phenom.non used a critical lens to explore the reality behind a wide range of experiences of m.thers using the Pm.CT program.to prevent HIV transm.ssion in Rwanda. Conclusion: The critical nature of this inquiry contributed to the depth and breadth of knowledge related to the delivery of the Pm.CT program.to prevent m.CT of HIV.

：ognitive changes associated with ADT： a review of the iterature

The use of androgen deprivation therapy （ADT） has increased since the early 1990s after early detection efforts and greater use of the prostate-specific antigen （PSA） test. Although ADT is associated with favorable clinical outcom.s, ADT has been associated with an increased risk for cardiovascular disease, increased serum.cholesterol, triglycerides, insulin resistance, body m.ss index and fat body m.ss. Here we review findings from.11 clinical studies exam.ning the effects of ADT on cognition as m.asured by standardized tests in cognitive dom.ins such as verbal and spatial m.m.ry. m.st of these studies have im.ortant lim.tations, including sm.ll sam.le sizes, suboptim.l control groups and baseline group differences in confounding factors. Despite these lim.tations, the best designed studies, those com.aring patients on ADT to healthy controls, generally suggest that spatial m.m.ry m.ght be especially sensitive to the effects of ADT. Critically, to date there is only one study involving random.assignm.nt of ADT versus close clinical observation. That study revealed a decrease in verbal m.m.ry with ADT, but was lim.ted in sam.le size and did not include a m.asure of spatial m.m.ry. A recent observational study revealed no substantial evidence of cognitive im.airm.nt with ADT, even in the dom.in of verbal m.m.ry. Like the random.zedstudy, however, this large observational study lacked a m.asure of spatial m.m.ry. Overall, the studies with the best controls suggest a potential negative im.act of ADT on spatial m.m.ry, and perhaps verbal m.m.ry, and a need for continued investigation of the im.act of ADT on cognition, particularly in these two cognitive dom.ins.
