Bulbosum to Go： A Toolbox to Utilize Hordeum vulgarelbulbosum Introgressions for Breeding and Beyond

Hordeum bulbosum L., a wild relative of barley （Hordeum vulgare L.）, has been considered as a valuable source of genetic diversity for barley improvement. Since the 1990s, a considerable number of barley/H, bulbosum introgression lines （IL）s has been generated, with segments introgressed from H. bulbosum harboring a diverse set of desirable traits. However, the efficient utilization of these ILs has been hampered, largely due to the lack of suitable molecular tools for their genetic characterization and highly reduced interspecific recombination frequencies in the region of the introgression. In the present study, we utilized genotyping-by-sequencing for the detailed molecular characterization of 145 ILs. Genotypic information allows the genetic diversity within the set of ILs to be determined and a strategy was outlined to tackle the obstacle of reduced recombination frequencies. Furthermore, we compiled exome capture re-sequencing information of barley and H. bulbosum and designed an integrated barley/ H. bulbosum sequence resource with polymorphism information on interspecific and intraspecific sequence variations of both species. The integrated sequence will be valuable for marker development in barley/H, bulbosum ILs derived from any barley and H. bulbosum donors. This study provides the tools for the widespread utilization of barley/H, bulbosum ILs in applied barley breeding and academic research.

Tumor Volume Associated with Recurrence in Prostate Cancer Patients with Seminal Vesicle Invasion

Objectives: To examine predictors of progression-free survival in men with seminal vesicle invasion (SVI) following radical prostatectomy (RP) for clinically localized prostate cancer. Methods and Materials: Between 1999 and 2009, 1383 men underwent RP at Indiana University. Among them, 115 men were identified with SVI. Disease progression was defined by a rise in PSA ≥ 0.2 ng/ml, receipt of salvage therapy, progression to metastatic disease, or death. After excluding 13 patients for receiving adjuvant therapy, 102 were stratified according to surgical margin (SM) and lymph node (LN) status for Kaplan-Meier analysis of disease progression. Cox proportional hazards analyses of biochemical progression-free survival were undertaken with respect to margin status, pre-operative prostate specific antigen (PSA), tumor volume, age, and post-operative Gleason sum. Stem and leaf plot was undertaken for tumor volume by biochemical PFS. Results: Mean age was 61 years, median Gleason sum was 7, mean tumor volume was 9.7 ml, and mean pre-operative PSA was 13.6 ng/ml. Mean time to disease progression was 17 months. Mean follow-up was 37 months. Kaplan-Meier analysis revealed statistically insignificant differences in progression-free survival stratified by SM and LN status (p = 0.12). Cox univariate analyses revealed tumor volume to be a statistically significant predictor of progression free survival (p = 0.02). Stem and Leaf plot revealed tumor volume to be statistically significantly larger in patients who experienced biochemical recurrence, compared to those who did not. Conclusion: Tumor volume was associated progression-free survival in this cohort of SVI patients, while pathologic Gleason sum, PSA, margin and nodal status were not.

An electro-hydrostatic actuator for hybrid active-passive vibration isolation

A novel electro-hydrostatic actuator （EHA） for active vibration isolation has been designed, modelled and tested. The EHA consists of a brushless DC motor running in oil and integrated with a bidirectional gear pump, driving a hydraulic cylinder. The actuator is designed to be integrated into a flexible strut connecting a helicopter rotor hub and fuselage, to provide isolation at the dominant rotor vibration frequency of around 20 Hz. The resonant frequency of the EHA is tuned to provide some passive vibration isolation. Active control increases the isolation performance by compensating for damping losses, and provides isolation over a broader range of frequencies. Tests on a prototype demonstrated a four-fold reduction of the root-mean-square transmitted force and a near elimination at the fundamental frequency. The advantages of the resonant EHA are a wider range of operating frequencies than a purely passive system, and lower power consumption than a purely active system.

The Fate of Aflatoxin in Corn Fermentation

A lab-scale ethanol fermentation was investigated to determine where aflatoxin concentrated during each phase of production. Four corn samples with high levels of aflatoxin (ranging from 7750 – 17,208 parts per billion) and their replicates were compared with a replicated negative control. Fractions were taken from the fermented mash, distilled ethanol, stillage, and dried corn solids (DCS). These fractions were analyzed using two different immunoassay methods and liquid chromatography tandem mass spectrometry (LC-MS/MS). Results indicated no aflatoxin was found in the distilled ethanol. Some aflatoxin (13%) was detected in the stillage, but most of the toxin was recovered in the DCSs ranging from 31% to 58%. A second series of experiments were conducted to investigate the effect of binders on dried distillers grains (DDGs). A brewers dried yeast anti-caking binder that contains glucomannon (MTB-100?), was mixed with contaminated DDGs. Addition of the binder showed a significant reduction in aflatoxin levels in comparison to a positive control. Aflatoxin binding at 2% binder w/w reached 72.5% and showed a minimal binding percentage increase of 80% at 6% binder w/w. Testing was also conducted to determine if environmental variables such as pH and temperature had any effect on the binding capabilities. Temperature near 0?C resulted in binding at 19.7% at a pH range of 6 to 8. Additionally, at a temperature of 40?C resulted in binding of 36%, 47%, and 45% at pHs 6, 7, and 8, respectively. These findings suggest that the addition of sorbents may be an effective way of salvaging contaminated DDGs.

Addition of tert-butylhydroquinone(TBHQ)to maize oil reduces lipid oxidation but does not prevent reductions in serum vitamin E in nursery pigs

Background: Maize oil is abundantly used in foods and feeds and is highly susceptible to oxidation. Consequently,commercially available antioxidants should be evaluated for effectiveness against lipid oxidation in swine diets. Our study was conducted to evaluate growth performance of nursery pigs fed oxidized maize oil and to determine effects of using antioxidants on oxidative status in a 2 × 2 factorial design. Two hundred eight weaned pigs were blocked by initial BW into 13 blocks, resulting in 4 pigs per pen and 13 pens per treatment. Dietary treatments included 6% unoxidized or oxidized maize oil, and 0 or 60 mg/kg of tert-butylhydroquinone(TBHQ), which was added after lipid oxidation. Data for growth performance were collected from 5 time periods of a two-phase feeding program(Phase 1 = d 0 to 12 and Phase 2 = d 13 to 34). Serum and liver samples were collected from one pig per pen, which had initial BW closest to average BW to determine oxidative status on d ***: Oxidized maize oil was heated for 12 h at 185 °C with 12 L/min of air, yielding a peroxide value(PV) of 5.98 m Eq O2/kg and TBARS of 0.11 mg MDA eq/g. Addition of TBHQ to diets containing oxidized maize oil decreased PV by 37% and increased the oil stability index by 69%. Final BW, ADG, ADFI, and G:F of pigs were not different among the four dietary treatments. However, pigs fed oxidized maize oil tended(P < 0.08) to increase hepatosomatic index by 5% compared with those fed unoxidized oil, and this was not affected by adding *** serum vitamin E concentration of pigs fed oxidized maize oil was less(P < 0.03) than pigs fed unoxidized oil,but this reduction was not reversed by adding TBHQ. The Tribbles(TRIB) family of pseudokinase proteins has been shown to play key roles in cell cycle, metabolic diseases, chronic inflammatory disease, and cancer development. A better understanding of the mechanisms of TRIB pseudokinases could provide new insights for disease development and help promote TRIB proteins as novel therapeutic targets for drug discovery. At the 2 nd International Symposium on Tribbles and Diseases held on May 7–9, 2018 in Beijing, China, a group of leading Tribbles scientists reported their findings and ongoing studies about the effects of the different TRIB proteins in the areas of immunity, metabolism, fundamental cell biology and cancer. Here, we summarize important and insightful overviews from 4 keynote lectures, 13 plenary lectures and 8 short talks that took place during this meeting. These findings may offer new insights for the understanding of the roles of TRIB pseudokinases in the development of various diseases.
