Exploring the bioactive potential of Serriatia marcescens VITAPI （Acc： 1933637） isolated from soil

BACKGROUND： Serratia is one of the most important groups of bacteria which produces proteolytic enzymes effectively and known to possess anti-inflammatory properties. The main focus of the current study was to extract the enzyme serratiopeptidase and pigment prodigiosin from Serratia mascescens. Prodigiosin is a red colored pigment produced by the bacterium Serratia marcescens. It is emerging as a valuable molecule because of its large applications. It has already been proved that pigmented strain of Serratia marcescens is less virulent than non-pigmented strains. Moreover the strain we have obtained is from farm soil which indicates that prodigiosin production can be carded safely using this strain. METHODS： In the present study, the isolate VITASP strain was confirmed by morphological, biochemical and molecular studies. The enzyme and pigment were analyzed for anti-oxidant, anti-inflammatory and cytotoxic properties. RESULTS： The isolate was further confirmed and identified as Serratia marcescens with 99% similarity. The extracted pigment showed potent radical scavenging effect with 86% and the enzyme was found to inhibit 83%, which was significant in comparison to ascorbic acid standard. The in vitro anti-inflammatory effect of pigment in controlled experimental conditions revealed its protection at 88% and the enzyme with 90%. Aspirin was used as the reference drug. The present findings exhibited a concentration dependent inhibition. The cytotoxic bioassay of pigment showed the IC5o value as （50） ~tg/mL with 63% cytotoxicity which was statistically significant compared to positive control. CONCLUSIONS： Therefore, it appears to be an essential remedial and application research. It may turn out to be highly beneficial to mankind in solving many problems associated with human health.

Cardiac papillary fibroelastoma:The need for a timely diagnosis

Cardiac papillary fibroelastomas(CPFs) are the second most common primary cardiac tumors and the most common cardiac valvular tumors. Although they are histologically benign and usually asymptomatic, CPFs can lead to serious and life-threatening complications like myocardial infarction, stroke, pulmonary embolus, cardiac arrest etc. CPFs represent a rare entity in clinical medicine and literature regarding their management is limited. We report two cases which illustrate such complications arising from undiagnosed CPFs on the aortic valve. We further stress on the importance of identifying CPFs early so that they can be managed appropriately based on recommendations from the available literature.

Rapid Tranquillisation: An AGREEable Ground?

Objective: Evidence base for rapid tranquillisation is an under researched area. Guidelines on rapid tranquilisation from English speaking countries were appraised using AGREE (Appraisal of Guidelines Research and Evaluation) and differences in their recommendations were analysed. Methods: Four independent psychiatrists appraised the guidelines using the AGREE tool. AGREE is a validated instrument used to assess the quality of guideline and recommendations using six domains of which each domain captures a specific aspect of the guideline development. The content was analysed manually. Results: Seven guidelines from five English speaking countries met the inclusion criteria. All the guidelines scored well on the domain of “scope and purpose”. NICE guidelines from the UK consistently scored well on all domains with the maximum possible score of 100 on the “applicability” domain. APA from the USA did well on the domain of “editorial independence”. AGREE could only examine the guideline development process and not the content. The guidelines differed in their recommendations of choice of drug for rapid tranquillisation. Discussion: All guidelines scored reasonably well on AGREE. National Institute of Clinical Excellence (NICE) has used robust strategies in developing the guidelines. Guidelines failed to achieve consensus in recommendations despite using a common pool of evidence. Haloperidol-promethazine combination is not recommended by any with the exception of NICE. This suggests data is selectively interpreted depending on locally prevalent customs.

Cyclic Sedimentation of the Barakar Formation, Singrauli Coalfield, India: Statistical Assessment from Borehole Logs

The succession of lithofacies of a part of the Barakar Formation of the Singrauli coalfield has been studied by statistical techniques. The lithologies have been grouped under five facies states viz. coarse-, medium-, and fine-grained sandstones, shale and coal for statistical analyses. Markov chain analysis indicates the arrangement of Barakar lithofacies in the form of fining-upward cycles. A complete cycle consists of conglomerate or coarse-grained sandstone at the base sequentially succeeded by medium- and fine-grained sandstones, shales and coal seam at the top. The entropy analysis puts the Barakar cycles in A-4 type of cyclicity, which consists of different proportions of lower-, side- and upper-truncated cycles of lithologic states. Regression analysis indicates a sympathetic relationship between total thickness of strata (net subsidence) and number of coal cycles and an antipathic relationship between average thickness and number of coal cycles. The cyclic sedimentation of the Barakar Formation was controlled by autocyclic process, which occurred due to the lateral migration of streams triggered by intrabasinal differential subsidence. In many instances, the clastic sediments issued from the laterally migrating rivers interrupted the sedimentation resulting in thinner cycles in areas where the numbers of cycles are more. Principal component and multivariate regression analyses suggest that the net subsidence of the basin is mostly controlled by number and thickness of sandstone beds and coal seams.

TAL effectors delivered by phytopathogenic Xanthomonas species are DNA-sequence-specific transcrip- tional activators of host susceptibility genes and sometimes resistance genes. The modularity of DNA recognition by TAL effectors makes them important also as tools for gene targeting and genome editing. Effector binding elements （EBEs） recognized by native TAL effectors in plants have been identified only on the forward strand of target promoters. Here, we demonstrate that TAL effectors can drive plant tran- scription from EBEs on either strand and in both directions. Furthermore, we show that a native TAL effector from Xanthomonas oryzae pv. oryzicola drives expression of a target with an EBE on each strand of its promoter. By inserting that promoter and derivatives between two reporter genes oriented head to head, we show that the TAL effector drives expression from either EBE in the respective orientations, and that activity at the reverse-strand EBE also potentiates forward transcription driven by activity at the forward-strand EBE. Our results reveal new modes of action for TAL effectors, suggesting the possibility of yet unrecognized targets important in plant disease, expanding the search space for off-targets of custom TAL effectors, and highlighting the potential of TAL effectors for probing fundamental aspects of plant transcription.

Deep neck space infection e A retrospective study of 270 cases at tertiary care center

Objective:To review the clinical findings in deep neck infections and identification predisposing factors of these complications. Methods:In this study, 270 patients with deep neck infections were studied retrospectively, study conducted in the Department of Ear, Nose and Throat, Dr. Shankarrao Chavan Govern-ment Medical College, Nanded, Maharashtra, India, from March 2013 to March 2016. Results:Analysis showed that males are most likely to have deep neck space infections (DNSI). Odontogenic and tonsillar causes were the more frequent ones. Staphylococcus aureus and Streptococcus species were the microorganisms more commonly isolated. Conclusion:DNSI remains a common and challenging disease for otorhinolaryngologists, and should be treated on emergency basis. In developing countries, lack of adequate nutrition, poor oral hygiene, tobacco chewing, smoking and beetle nut chewing has led to an increased prevalence of dental and periodontal diseases. In present study, Odontogenic infections were the most common etiological factor for DNSI.

Dual Artificial Neural Network for Rainfall-Runoff Forecasting

One of the principal issues related to hydrologic models for prediction of runoff is the estimation of extreme values (floods). It is well understood that unless the models capture the dynamics of rainfall-runoff process, the improvement in prediction of such extremes is far from reality. In this paper, it is proposed to develop a dual (combined and paralleled) artificial neural network (D-ANN), which aims to improve the models performance, especially in terms of extreme values. The performance of the proposed dual-ANN model is compared with that of feed forward ANN (FF-ANN) model, the later being the most common ANN model used in hydrologic literature. The forecasting exercise is carried out for hourly river flow data of Kolar Basin, India. The results of the comparison indicate that the D-ANN model performs better than the FF-ANN model.
