Metal-dependent anaerobic methane oxidation in marine sediment:insights from marine settings and other systems

Anaerobic oxidation of methane(AOM) plays a crucial role in controlling global methane emission. This is a microbial process that relies on the reduction of external electron acceptors such as sulfate, nitrate/nitrite, and transient metal ions. In marine settings, the dominant electron acceptor for AOM is sulfate, while other known electron acceptors are transient metal ions such as iron and manganese oxides. Despite the AOM process coupled with sulfate reduction being relatively well characterized,researches on metal-dependent AOM process are few, and no microorganism has to date been identified as being responsible for this reaction in natural marine environments. In this review, geochemical evidences of metal-dependent AOM from sediment cores in various marine environments are summarized. Studies have showed that iron and manganese are reduced in accordance with methane oxidation in seeps or diffusive profiles below the methanogenesis zone. The potential biochemical basis and mechanisms for metal-dependent AOM processes are here presented and discussed. Future research will shed light on the microbes involved in this process and also on the molecular basis of the electron transfer between these microbes and metals in natural marine environments.

AIM： TO assess the effect of iron reduction after phlebotomy in rats with ＂normal＂ hepatic iron concentration （HIC） on the progression of hepatic fibrosis, as a result of bile duct ligation （BDL）. METHODS： Rats underwent phlebotomy before or after sham operation or BDL. Animals undergone only BDL or sham operation served as controls. Two weeks after surgery, indices of hepatic damage and fibrosis were evaluated. RESULTS： Phlebotomy lowered HIC. Phlebotomy after BDL was associated with body weight increase, lower hepatic weight, less portal hypertension, less periportal necrosis, less portal inflammation, lower hepatic activity index score and higher albumin levels. On the other hand, phlebotomy before BDL was associated with body weight decrease and hepatic activity index score increase. Phlebotomy after sham operation was not associated with any hepatic or systemic adverse effects. CONCLUSION： Reduction of HIC after induction of liver damage may have beneficial effects in BDL rats. However, iron deficiency could induce impairment of liver function and may make the liver more susceptible to insults like BDL.

Celiac disease(CD)affects at least 1%of the Western population but remains largely *** our laboratory,we utilize a novel algorithm to diagnose pediatric CD that offers both high sensitivity and high specificity for diagnosis in an outpatient *** aim of the present study was to challenge this algorithm and to test its performance in children and adults suspected of having *** a three-assay algorithm,screening with the most sensitive tissue transglutaminase(tTG)complexed with deamidated gliadin peptide neoepitope immunoglobulin A(IgA)1IgG assay and confirming with the two specific tTG IgA and tTG IgA1IgG assays,we examined the serological results from 112 children aged 0–17 years old and 60 adults in comparison to their respective biopsy *** algorithm performance was calculated by statistical *** use of the new algorithm enabled us to diagnose CD with 98%sensitivity,93%specificity and 95%accuracy in the pediatric group and 94%sensitivity,92%specificity and 93%accuracy in the total population *** false-negative cases in the adult group were attributed to previous adherence to a gluten-free diet,and the single false-negative result in a young child became a true positive after 6 *** have also monitored three celiac patients before and after diagnosis and found that the algorithm may be suitable for disease *** newly proposed three-assay algorithm for celiac detection is very reliable in both children and *** to the high performance of this assay,the further need for confirmatory intestinal biopsies will be reassessed.

AIM： TO assess the safety and efficacy of antioxidant therapy for patients with chronic hepatitis C virus （HCV） infection.METHODS： One hundred chronic HCV infection patients failed in interferon treatment were enrolled and randomly assigned to receive combined intravenous and oral antioxidants or placebo, or oral treatment alone, Primary end points were liver enzymes, HCV-RNA levels and histology.RESULTS： Combined oral and intravenous antioxidant therapy was associated with a significant decline in ALT levels in 52% of patients who received antioxidant therapy vs 20% of patients who received placebo （P = 0.05）. Histology activity index （HAI） score at the end of treatment was reduced in 48% of patients who received antioxidant therapy vs 26% of patients who received placebo （P = 0.21）. HCV-RNA levels decreased by l-log or more in 28% of patients who received antioxidant therapy vs 12% who received placebo （P = NS）. In part 11 of the trial, oral administration of antioxidants was not associated with significant alterations in any of the end points.CONCLUSION： Antioxidant therapy has a mild beneficial effect on the inflammatory response of chronic HCV infection patients who are non-responders to interferon. Combined antiviral and antioxidant therapy may be beneficial for these patients.

Wilson’s disease(WD)is an autosomal recessive inherited disorder of hepatic copper *** can be present in different clinical conditions,with the most common ones being liver disease and neuropsychiatric *** cases present symptoms at<40years of ***,few reports exist in the literature on patients in whom the disease presented beyond this *** this report,we present a case of late onset fulminant WD in a 58-year-old patient in whom the diagnosis was established clinically,by genetic analysis of the ATP7B gene disclosing rare mutations(G1099S and c.1707+3ins T)as well as by high hepatic copper *** also reviewed the relevant *** diagnosis of WD with late onset presentation is easily *** diagnostic features and the geneticbackground in patients with late onset WD are not different from those in patients with early onset WD,except for the *** treatments for this disorder that can be fatal are available and will prevent or reverse many manifestations if the disease is discovered early.

Jerusalem is holy to the three main monotheistic religions. Hence Jerusalem is, from a touristic perspective, a world city. Jerusalem is also a highly segmented city. While Jerusalem may seem as an extreme case, it is not unique. Many cities attract large number of tourists to various widely renowned sites within them, in many cases with religious facets. Public transport systems are largely designed to address the demand of local residents. Yet, tourist flows often differ from the travel patterns of local residents, both temporally and spatially. This paper evaluates how Jerusalem’s public transport system serves the global tourist city. To this end, the study focuses on independent tourists, not package-tour tourists. Specifically, we analyze to what extent are the main tourist sites served by the public transportation system. To evaluate accessibility to tourist destinations we use GIS to conduct Least-Cost-Modeling. This methodology measures the effective, rather than the Euclidian distance to tourist destination. We find that the accessibility by public transportation to tourist destinations with high global importance, specifically Christian-religious sites is relatively low. In the final part of the paper we propose a number of explanations for that.

Mapping What They Know: Concept Maps as an Effective Tool for Assessing Students’ Systems Thinking

In 2006 anew biology curriculum called “Human Biology: Emphasizing the Role of Homeostasis” was introduced into the Israeli high school system. Complex systems like those that make up the human body have become increasingly important as a focus of high school education. This study examines the effectiveness of the concept map as a assessment tool for students’ systems thinking, a tool that provides researchers with a detailed picture of the systems thinking development taking place within individual students. The content of the students’ concept maps was translated into information about students’ system thinking using the Systems Thinking Hierarchy (STH) model, a model in which system thinking is categorized according to eight hierarchical characteristics or abilities. The goal was to use the maps to characterize Israeli high school students’ understanding of the body’s systemic nature. To do this, we identified the extent to which the students understand three central elements of systems, namely hierarchy, homeostasis, and dynamism, and then analyzed this understanding according to its place within the hierarchical stages of the STH model. The extensive qualitative data analysis of 48 concept maps made by 11th grade biology majors suggest that the strength of the concept map is in its ability to describe the first two levels of system thinking (analysis and synthesis). However, it proved less successful in eliciting evidence of the third and highest level, particularly of students’ understanding of patterns, of homeostasis and their capacity for temporal thinking.

The recently domesticated species, Cephalaria joppensis (CJ), is emerging as a new alternative forage crop in Israel. It has high biomass potential and nutritional values that are comparable to forage wheat. However, CJ emerges slowly under cold conditions, which hinders its development as a major winter crop. Additional tolerance for abiotic stress would improve its performance as a forage crop. We examined the effects of several abiotic factors (i.e., cold, salinity, drought and pH) on CJ germination under controlled conditions. The effect of temperature was studied by incubating seeds at different temperatures between 7°C and 35°C. The effects of salinity, osmotic potential and pH were tested by incubating seeds at different NaCl, PEG and pH levels, respectively. Temperature, salinity and osmotic potential significantly affected germination;whereas pH did not. Temperature did not affect the final proportion of germinated seeds, but did affect other germination-rate variables, indicating that germination rate might be the limiting factor under field conditions. Salinity also affected germination-rate variables, but not the proportion of seeds that germinated. Notably, CJ was found to be relatively resistant to high salt concentrations, with a 273 mM NaCl threshold for germination, indicating its potential as a relatively salt-tolerant forage crop. Both the proportion of germinated seeds and the germination rate were highly sensitive to the osmotic-potential treatments, indicating that drought resistance will remain the biggest challenge for CJ. This study provides baseline data for a rapid and efficient system for further screening for abiotic-stress tolerance among wild and cultivated lines of CJ.

Effect of autologous mesenchymal stem cells induced by low level laser therapy on cardiogenesis in the infarcted area following myocardial infarction in rats

In this study, we investigated the hypothesis that photobiostimulation by low-energy laser therapy (LLLT) applied to the bone marrow (BM) of myocardial infarcted rats may attenuate the scarring processes that follow myocardial infarction (MI). Wistar rats underwent experimental MI. LLLT (Ga-Al-As diode laser) was applied to the BM of the exposed tibia at different time intervals post-MI (4 hrs, 48 hrs and 5 days). Sham-operated infarcted rats served as control. Infarct size was significantly reduced (55%) in the laser-treated rats as compared to the control non-treated rats, at 2 weeks post-MI. A significant 3-fold increase was observed in the density of desmin immunopositive stained cells 14 days post-MI in the infarcted area of the laser-treated rats as compared to the non-laser-treated controls. The electron microscopy from the control infarcted rat hearts revealed a typical interphase area between the intact myocardium and the infarcted area, with conspicuous fibroblasts with collagen deposition dispersed among them. In rats that were laser treated (to BM), the interphase zone demonstrated cells with different intracellular structures. There was also a significant increase in the percentage of c-kit positive cells and macrophages in the circulating blood of the laser treated rats as compared to control non treated ones. In the majority of the cells clusters of myofibrils anchored to well-developed Z-lines and structures resembling the morphological characteristics of mature intact cardiomyocytes were evident. In conclusion, LLLT to the BM of rats post-MI induces cardiogenesis mainly at the borders of the infarcted area in the heart.
