Estrogen, estrogen receptors, and hepatocellular carcinoma: Are we there yet?

A protective role of the sex steroid hormone estrogenin hepatocellular carcinoma(Hcc) was suggested a few decades ago according to clinical data showing higher Hcc morbidity and mortality among males. Several recent studies further confirmed the anti-cancer effects of estrogen in the liver. However, it remains to be identified how to exploit estrogen signalling within clinical settings for Hcc treatment. There are several unresolved issues related to the estrogen pathway in liver cells. The main problems include the absence of a clear understanding of which estrogen receptor(ER) isoform is predominantly expressed in normal and malignant liver cells, the ER isoform expression difference between males and females, and which ER isoform should be targeted when designing Hcc therapy. Some of those questions were recently addressed by Iyer and coauthors. The current editorial review critically analyses the study by Iyer et al(WJG, 2017) that investigated the expression of ER subtypes in liver samples collected from patients with a healthy liver, hepatitis c virus cirrhosis, and Hcc. ER presence was evaluated in association with gender, intracellular localization, inflammation marker NF-kB, and proliferation-related effector cyclin D1. The study limitations and advantages are discussed in light of recent advances in the Hcc and estrogen signalling areas.

cONSPEcTUS: Future medicine is primarily aiming at the development of novel approachesfor an early diagnosis of diseases and a personalized therapy for patients. For achieving theseobjectives, a key role is played by medical imaging. Among available noninvasive imagingtechniques, Fluorine-19 (^(19)F) Magnetic Resonance Imaging (MRI) is emerging as a powerfulquantitative detection modality for clinical use both for molecular imaging and for cell *** strength of using ^(19)F-MRI is mainly related to the lack of endogenous organic fluorine intissues, with no background, enabling the visualization of fluorinated tracers as hot-spot images,adding secondary independent information to the anatomical features provided by thegrayscale ^(1)H-MRI. The main challenge for ^(19)F-MRI clinical application is the intrinsic reducedsensitivity of MRI. To improve sensitivity, undoubtedly the use of a high field MRI scanner andcryogenic radiofrequency probes is advantageous, but there is a clear need of developingincreasingly effective fluorinated *** ideal tracer should bear as many as possible magnetically equivalent fluorine atoms andshow optimal magnetic resonance relaxivity properties (i.e., T_(1) and T_(2)), which enable reduced acquisition time with the possibility toapply fast imaging methods. Moreover, it should be biocompatible with reduced tendency to bioaccumulate in tissues, which is oneof the main drawbacks in using perfluorocarbons (PFcs), together with their difficulty to be chemically modified with functionalgroups. In fact, PFcs such as perfluorooctyl bromide (PFOB), perfluoro-15-crown-5-ether (PFcE), and linear perfluoropolyethers(PFPE) are currently the most used tracers in ^(19)F-MRI preclinical and clinical studies, with the above-mentioned limitations. Influence of polyolefin fibers on the strength and deformability properties of road pavement concrete

Influence of the type and quantity of polyolefin fibers on the strength properties(compression strength, tensile strength in bending, strength in uniaxial tension), the deformation properties(elastic modulus, Poisson’s coefficient) and the abrasion resistance of cement concrete with water-to-cement ratio within 0.31 e0.55 were stated in the *** ways of fibers introduction into fresh concrete were investigated. It was shown that the fibers introduction method and procedure of mixing are the ways to improve the fibers uniform distribution in fresh concrete. The increase of the bending tensile strength and the uniaxial tensile strength of concrete with fibers reinforcement in comparison with the reference concrete was observed with the water-to-cement ratio decrease. The increase of uniaxial tensile strength at age of 28 days for concrete with macrofibers in amount of4.5 kg/m^3 was 23% and 29%;for macrofibers in quantity of 3 kg/m^3 was 19% and 26% with water-cement ratio equal to 0.49 and 0.31, respectively. The maximum reduction of abrasion in the range of 7.5%e10% was observed in concrete with water-to-cement ratio within0.44 e0.55 for all investigated types of fibers. The influence of fibers on the concrete abrasion with lower W/c ratio was negligible. The results can contribute to the rational use of modified polyolefin fibers in road pavement concrete.

Performance of low temperature Microbial Fuel cells(MFcs)catalyzed by mixed bacterial consortia

Microbial Fuel cells（MFcs） are a promising technology for treating wastewater in a sustainable manner. In potential applications, low temperatures substantially reduce MFc performance. To better understand the effect of temperature and particularly how bioanodes respond to changes in temperature, we investigated the current generation of mixed-culture and pure-culture MFcs at two low temperatures, 10°c and 5°c. The results implied that the mixed-culture MFc sustainably performed better than the pure-culture（Shewanella） MFc at 10°c, but the electrogenic activity of anodic bacteria was substantially reduced at the lower temperature of 5°c. At 10°c, the maximum output voltage generated with the mixed-culture was 540–560 m V, which was 10%–15% higher than that of Shewanella MFcs. The maximum power density reached 465.3 ± 5.8 m W/m^2 for the mixed-culture at10°c, while only 68.7 ± 3.7 m W/m^2 was achieved with the pure-culture. It was shown that the anodic biofilm of the mixed-culture MFc had a lower overpotential and resistance than the pure-culture MFc. Phylogenetic analysis disclosed the prevalence of Geobacter and Pseudomonas rather than Shewanella in the mixed-culture anodic biofilm, which mitigated the increase of resistance or overpotential at low temperatures.

Introduction:Bats are threatened by agricultural intensification,and although bat ecology in agricultural landscapes is in the focus of current research,the effects of interacting spatiotemporal factors on species-specific bat activity above farmland remain *** aim was to identify spatiotemporal factors and their interactions relevant for the activity of bat species above conventionally managed arable ***:We repeatedly monitored relative bat activity above open arable fields in Germany using acoustic *** used site-related biotic and abiotic factors and landscape characteristics across five spatial scales,their combinations,and interactions to identify those factors which best explain variation in bat ***:Numerous interactions between landscape characteristics and the insect abundance affected bat activity above *** instance,Pipistrellus pipistrellus became more active with increasing insect abundance,but only above fields with a low proportion of woody vegetation cover in the ***,the level of bat activity in summer depended on landscape *** example,the activity of Pipistrellus nathusii was relatively low in summer above fields that were surrounded by vegetation patches with a high degree of edge complexity(e.g.,hedgerow).However,the activity remained at a relatively high level and did not differ between seasons above fields that were surrounded by vegetation patches with a low degree of edge complexity(e.g.,roundly shaped forest patch).conclusions:Our results revealed that landscape characteristics and their interactions with insect abundance affected bat activity above conventionally managed fields and highlighted the opportunistic foraging behavior of *** improve the conditions for bats in agricultural landscapes,we recommend re-establishing landscape heterogeneity to protect aquatic habitats and to increase arthropod availability.

Selection of phages and conditions for the safe phage therapy against Pseudomonas aeruginosa infections

The emergence of multidrug-resistant bacterial pathogens forced us to consider the phage therapy as one of the possible alternative approaches to treatment. The purpose of this paper is to consider the conditions for the safe, long-term use of phage therapy against various infections caused by Pseudomonas aeruginosa. We describe the selection of the most suitable phages, their most effective combinations and some approaches for the rapid recognition of phages unsuitable for use in therapy. The benefi ts and disadvantages of the various different approaches to the preparation of phage mixtures are considered, together with the specifi c conditions that are required for the safe application of phage therapy in general hospitals and the possibilities for the development of personalized phage therapy.

Impact of Natural Oil-Based Recycled Polyols on Properties of cast Polyurethanes

In this study, castor oil, rapeseed oil and medium chain triglycerides of coconut oil, were transesterified bymeans of 2-ethyl-2-hydroxymethyl-1,3-propanediol (trimethylolpropane) and consequently used to convertpolycarbonate waste from end-of-life vehicles into liquid polyols. The prepared recycled polyols, composeduniquely of renewable and recycled components, had a hydroxyl number of ca. 250 mg KOH·g−1. They weresuccessfully applied as 100% replacement of a virgin polyol for preparation of solid crosslinked polyurethanes(PU) by solvent-free casting. The produced rigid cast PU exhibited the main transition temperature rangingfrom 44°c to 53°c, the hardness value from 46 to 61 Shore D and the beneficial low water absorption (0.4-0.5wt.%). The PU network structure was highly influenced by the presence of aromatic structures ofpolycarbonate-units and fatty acid compositions. A suitable selection of natural oil thus enabled to adjustthermo-mechanical properties and promote excellent optical transparency of the produced PU.

Is diabetes a causal agent for colorectal cancer? Pathophysiological and molecular mechanisms

The possible relationship between diabetes mellitus (DM) and colorectal cancer (cRc), concerning pathophysiological and molecular mechanisms is highlighted in this review. The most recent and complete articles and developments in this particular field were thoroughly reviewed. common risk factors, such as obesity, sedentary lifestyle, and Western diet between DM and cRc, led to the theory that DM might be a causal agent for cRc development. Various studies have connected type 2 DM and cRc, either proximal or distal, in both sexes. Additionally, chronic insulin treatment has been linked with increased colorectal tumor risk among type 2 diabetic patients. Interestingly, elevated hemoglobin A1c has been proven to be an independent predictor of aggressive clinical behavior in cRc patients. These mechanisms include the insulin-like growth factor-hyperinsulinemia theory and the participation of oncogenic intracellular signaling pathways. Furthermore, it has been proposed that cox-2 inhibitors might have a role in decreasing the incidence of cRc. Finally, the use of statins to reduce the risk for colon cancer in patients with diabetes has remained controversial. Diabetic patients over 50 should receive counseling regarding their elevated risk for cRc, and screening colonoscopy should be recommended before initiating insulin therapy. However, there are no current guidelines, and this strategy is not yet applicable to some countries, as the corresponding risk would not allow screening colonoscopy to be adopted. There is strong evidence to indicate that DM is a causal agent for cRc development. This conclusion provides new impetus for re-evaluating cRc screening worldwide.
