Butein,a rare chalcone found in the toxic plant toxicodendron vernicifluum,has been shown to regulate glucose homeostasis via inhibition of the nuclear factor kappa-B kinase subunit beta(IKKβ)/nuclear factor kappa B(NF-κB)pathway in the ***,we investigated whether the nonpoisonous plant Dahlia pinnata could be a source of butein as a potential treatment for type 2 diabetes(t2D).In mice fed a high-fat diet(HFD)to induce glucose intolerance,an oral *** petal extract improved glucose tolerance at doses of 3.3 mg/kg body weight and 10 mg/kg body ***,this effect was not mediated by butein alone but by butein combined with the closely related flavonoids,sulfuretin and/or ***,the extract improved systemic insulin *** of phosphatidylinositol 3-kinase to block insulin signaling in the brain abrogated the glucoregulatory effect of the orally administered *** extract reinstated central insulin signaling and normalized astrogliosis in the hypothalamus of HFD-fed *** NF-κB reporter zebrafish to determine IKKβ/NF-κB activity,a potent anti-inflammatory action of the extract was found.A randomized controlled crossover clinical trial on participants with predia-betes or t2D confirmed the safety and efficacy of the extract in *** conclusion,we identified an extract from the flower petals of *** as a novel treatment option for t2D,potentially targeting the central regulation of glucose homeostasis as a root cause of the disease.

Clostridium beijerinckii is a potentially important industrial microorganism as it can synthesize valuable chemicals and fuels from various carbon sources. the establishment of convenient to use, effective gene tools with which the organism can be rapidly modified is essential if its full potential is to be realized. Here, we developed a genomic editing tool(pCBEclos) for use in C. beijerinckii based on the fusion of cytidine deaminase(Apobec1), Cas9 D10 A nickase and uracil DNA glycosylase inhibitor(UGI). Apobec1 and UGI are guided to the target site where they introduce specific base-pair substitutions through the conversion of C·G to t·A. By appropriate choice of target sequence, these nucleotide changes are capable of creating missense mutation or null mutations in a gene. through optimization of pCBEclos, the system derived, pCBEclos-opt, has been used to rapidly generate four different mutants in C. beijerinckii, in pyrE, xylR, spo0 A and araR. the efficiency of the system was such that they could sometimes be directly obtained following transformation, otherwise only requiring one single re-streaking step. Whilst CRISPR-Cas9 nickase systems, such as pNICKclos2.0, have previously been reported in C. beijerinckii, pCBEclos-opt does not rely on homologous recombination, a process that is intrinsically inefficient in clostridia such as C. beijerinckii. As a consequence, bulky editing templates do not need to be included in the knock-out plasmids. this both reduces plasmid size and makes their construction simpler, e.g., whereas the assembly of pNICKclos2.0 requires six primers for the assembly of a typical knock-out plasmid, pCBEclos-opt requires just two primers. the pCBEclos-opt plasmid established here represents a powerful new tool for genome editing in C. beijerinckii, which should be readily applicable to other clostridial species.

信号分子NO已被证明在植物生长发育的许多过程中起重要作用。为了发掘影响NO代谢和积累的关键基因,我们利用NO特异性荧光染料DAF-FMDA结合HO处理,建立了一套筛选NO积累缺陷突变体的系统。经HO处理30min后,拟南芥野生型植株根部NO的含量可以增加8倍,而突变体中HO诱导NO积累的功能缺失。对一个突变体的图位克隆结果表明,该突变体是由于抑制蛋白基因PHB3上的一个点突变造成的,使得PHB3蛋白的SPFH区域由保守的甘氨酸-37转变为天冬氨酸。通过对该突变体和t-DNA插入突变体的研究,发现它们的根部受HO诱导的NO积累量明显低于野生型,且在ABA诱导的NO积累、气孔关闭和生长素诱导的侧根形成过程中存在缺陷。但突变体中HO代谢和信号传递过程并无缺陷,HO诱导的基因表达也不受影响。上述结果证明,PHB3是调节NO代谢平衡体系的一个重要因子,而且将抑制基因的功能进一步延伸到对NO积累和NO所介导反应的调控。

[背景]评估由于环境空气中细颗粒物（PM2.5）长期暴露引起的疾病负担,需要了解相对风险度（RR）函数的图形和幅度。然而,在世界上很多观察到高PM2.5环境浓度的地方,都缺乏足够的直接证据确定死亡RR函数的图形。[目的]建立可用于全球PM2.5暴露范围内成人死因的RR函数：缺血性心脏病（IHD）、脑血管疾病（脑卒中）、慢性阻塞性肺病（COPD）和肺癌（LC）。建立急性下呼吸道感染（ALRI）发病的RR函数,用于估算〈5岁的儿童中死亡率和健康生活损失年数。[方法]通过整合现有的来自于环境空气污染（AAP）、二手烟草烟雾、家用固体烹饪用燃料和主动吸烟（AS）等研究的RR信息,拟合综合暴露-反应（IER）模型。采用颗粒物质的吸入剂量,通过转化AS暴露来估计全年PM2.5的暴露当量。在估算全球环境PM2.5浓度的基础上,得出各国的人群归因分数（PAFs）。[结果]与以往负担评估中使用的其他7种模型相比,IER模型是一种优越的RR预测方法。在不同国家中,归因于AAP暴露的PAF百分数各不相同,分别为：IHD,2~41;脑卒中,1~43;COPD,〈1~21;LC,〈1~25;ALRI,〈1~38。[结论]我们建立了一种以细颗粒物为基础的RR模型,它通过整合来自不同类型的燃烧所产生颗粒物排放的RR信息,涵盖全球范围的暴露。当获得新的RR信息时,可以对这个模型进行更新。

Societal biosecurity–measures built into everyday society to minimize risks from pests and diseases–is an important aspect of managing epidemics and *** aimed to identify societal options for reducing the transmission and spread of respiratory *** used SARS-CoV-2(severe acute respiratory syndrome coronavirus 2)as a case study to meet the immediate need to manage the COVID-19 pandemic and eventually transition to more normal societal conditions,and to catalog options for managing similar pandemics in the *** used a‘solution scanning’*** read the literature;consulted psychology,public health,medical,and solution scanning experts;crowd-sourced options using social media;and collated comments on a ***,we present a list of 519 possible measures to reduce SARS-CoV-2 transmission and *** provide a long list of options for policymakers and businesses to consider when designing biosecurity plans to combat SARS-CoV-2 and similar pathogens in the *** also developed an online application to help with this *** encourage testing of actions,documentation of outcomes,revisions to the current list,and the addition of further options.

Background and aimsCigarette pack warning labels, especially stronger ones, stimulate quitting-related activity. this study aimed to extend understanding of impact of pack warning changes in Malaysia and thailand, focussing on the 2009 introduction of graphic warnings in Malaysia. In particular, we aimed to:(1) compare the differences in pack warning reactions of smokers within and between these two developing countries over time,(2) examine if warning reactions predict subsequent quitting activities, and(3) examine possible wear-out effects of pack warnings in these two countries. Methods Data came from 6 waves of the International tobacco Control Southeast Asia(the ItC SEA) Survey, a prospective cohort survey conducted among adult smokers in Malaysia and thailand between 2005 and 2013. Between 4706 and 4422 smokers were interviewed at each survey wave. Key measures included salience of cigarette pack warnings, cognitive responses, forgoing cigarettes and avoiding warnings. the outcome measure in predictive analysis was quit attempts between survey *** During the first three survey waves(2005-2008), reaction levels were significantly lower in Malaysia than in thailand except for forgoing cigarettes at Wave 1. However, at Wave 4(shortly after the implementation of graphic warnings) the reaction levels among Malaysia smokers considerably increased from that of early waves;and for some measures the levels reached as high as that of thailand. Compared to the early waves, the warning salience and cognitive reaction levels in Malaysia remained higher at Wave 5. But overall, in Malaysia wear-out of warnings was greater –by Wave 6, most of the reported reaction levels were no longer significantly higher than that of the baseline wave. In contrast, overall, the reaction levels of post-graphic warning waves were higher than those at the baseline wave in *** thailand, Wave 3 warning reactions were predictive of subsequent quit attempts in bivariate analysis

this study examines the effects of different cigarette package warnings in Australia, Canada and the United Kingdom up to 5 years postimplementation. the data came from the International tobacco Control *** included salience of warnings, cognitive responses, forgoing cigarettes and avoiding warnings. Although salience of the UK warnings was higher than the Australian and Canadian pictorial warnings, this did not lead to greater levels of cognitive reactions, forgoing or *** was no difference in ratings between the Australian and UK warnings for cognitive responses and forgoing, but the Canadian warnings were responded to more strongly. Avoidance of the Australian warnings was greater than to UK ones, but less than to the Canadian warnings. the impact of warnings declined over time in all three countries. Declines were comparable between Australia and the United Kingdom on all measures except avoiding, where Australia had a greater rate of decline;and for salience where the decline was slower in Canada.
