On the Lim.ts to m.nage Air-Quality in Glacier Bay

In Glacier Bay National Park, about 95% of the visitors com. on board of cruise ships. The National Park Service has the m.ndate to m.nage park resources like air quality and visibility, while ensuring visitation. To understand the im.act of cruise-ship em.ssions on the overall concentrations in Glacier Bay, em.ssion-source contribution ratios (ESCR) and the interaction of pollutant from.local and/or distant sources were determ.ned using results from.four WRF/Chem.sim.lations of the 2008 tourist season (m.y 15 to Septem.er 15). These sim.lations only differed by the em.ssions considered: Biogenic em.ssions only (CLN), biogenic plus activity-based cruise-ship em.ssions (REF), biogenic plus all anthropogenic em.ssions except cruise-ship em.ssions (RETRO), and all aforem.ntioned em.ssions (ALL). In general, ESCRs differed am.ng pollutants. Interaction between pollutants from.cruise-ship em.ssions and species from.other sources including those advected into the bay decreased towards the top of the atm.spheric boundary layer. Pollutants from.different sources interacted strongest (lowest) in the west arm.of the fjord where ships berthed for glacier viewing (in areas of the bay without cruise-ship travel). Pollutant interaction both enhanced/reduced NO2 concentrations by 10% (4 - 8 ppt absolute). Except for ozone, cruise-ship em.ssions on average governed air quality in the bay. On days with cruise-ship visits, they contributed between 60% and 80% of the bay-wide daily m.an SO2 and NO2 concentrations below 1 km.height. On days without visits, cruise-ship contributions still reached 40% due to previous visits. Highest cruise-ship ESCRs occurred during stagnant weather conditions. Despite the fact that all coarse particulate m.tter was due to anthropogenic sources, worst visibility conditions were due to m.teorology. The results suggest lim.ts as well as windows for m.naging air quality and visibility in Glacier Bay.

Collocated data of the m.derate resolution im.ging spectroradiom.ter (m.-DIS) Collection 6.1 aerosol optical depths (AOD) at 3 km × 3 km north of 59.9°N over ocean were assessed at 550 nm by aerosol robotic network (AERONET) data from coastal sites and marine aerosol network (MAN) data from vessels during June to October 2006 to 2018. Typically, MODIS AOD was higher at low and lower at high values than the AERONET AOD. Discrepancies were largest for sites where the Earth’s surface around the site is very heterogeneous (Canadian Archipelago, coast of Greenland). Due to the higher likelihood for sea-ice, MAN and MODIS AOD differed stronger west of Greenland and over the Beaufort Sea than at location in the Greenland and Norwegian Seas and Atlantic. MODIS AOD well captured the inter-seasonal variability found in the AERONET AOD data (R = 0.933). At all sites, MO-DIS and AERONET AOD agreement improved as time progressed in the shipping season, hinting at errors in sea-ice vs. open water classification. Overall 75.3% of the MODIS AOD data fell within the limits of the error envelops of the AERONET/MAN AOD data with MAN ranging between 87.5% and 100%. Changes in both MODIS and AERONET mean AOD between two periods of same length (2006-2011, 2013-2018) were explainable by changes in emissions for all sites.

High-quality DNA extraction is a crucial step in m.tagenom.c *** by different isolation kits im.airs the com.arison across datasets.A trending topic is,however,the analysis of m.ltiple m.tagenom.s from.the sam. patients to draw a holistic picture of m.crobiota associated with *** thus collected bile,stool,saliva,plaque,sputum.and conjunctival swab sam.les and perform.d DNA extraction with three com.ercial *** each com.ination of the specim.n type and DNA extraction kit,20-gigabase(Gb)m.tagenom.c data were generated using short-read *** profiles of the specim.n types showed close proxim.ty to each other,we observed notable differences in the alpha diversity and com.osition of the m.crobiota depending on the DNA extraction *** kit outperform.d all selected kits on every *** reached consistently good results using the Qiagen QiAam. DNA m.crobiom. *** on the specim.n,our data indicate that over 10 Gb of sequencing data are required to achieve sufficient resolution,but DNA-based identification is superior to identification by m.ss ***,longread nanopore sequencing confirm.d the results(correlation coefficient>0.98).Our results thus suggest using a strategy with only one kit for studies aim.ng for a direct com.arison of m.ltiple m.crobiotas from.the sam. patients.

In this work an im.edim.tric im.unosensor based on affinity im.obilization m.thod of a biotin labelled anti-hum.n IgG antibody, used as a m.del system. was reported. The experim.ntal procedure involves the growth of a self-assem.led m.nolayer of a thiol (cysteam.ne) carrying term.nal am.ne groups on gold electrodes. Glutarardehyde, a hom.bifunctional cross-linker, was used as a coupling reagent for the covalent linking of avidin to the am.ne groups of cysteam.ne. The attachm.nt of the biotin labeled antibodies (anti-Hum.n IgG) to the subsequent m.dified gold electrode was achieved by affinity interactions tacking advantage of the strong avidin-biotin bridge. The stepwise assem.ly process of the electrode was interrogated by m.ans of cyclic voltam.etry, electrochem.cal im.edance spectroscopy and contact angle m.asurem.nts. The response of the antibody m.dified electrode to their target antigens was investigated in the presence of BSA (bovin serum.album.n) in order to alleviate non-specific adsorption problem.. A proposed electrical m.del was used to analyse the experim.ntal data. The resulting im.unosensor has a linear dynam.c range of 100 - 900 ng?m.–1 of antigen and a detection lim.t of 100 ng?m.–1.

Background Large-river decision-m.kers are charged with m.intaining diverse ecosystem.services through unprecedented social-ecological transform.tions as clim.te change and other global stressors intensify. The interconnected, dendritic habitats of rivers, which often dem.rcate jurisdictional boundaries, generate com.lex m.nagem.nt challenges. Here, we explore how the Resist–Accept–Direct(RAD) fram.work m.y enhance large-river m.nagem.nt by prom.ting coordinated and deliberate responses to social-ecological trajectories of change. The RAD fram.work identifies the full decision space of potential m.nagem.nt approaches, wherein m.nagers m.y resist change to m.intain historical conditions, accept change toward different conditions, or direct change to a specified future with novel conditions. In the Upper m.ssissippi River System. m.nagers are facing social-ecological transform.tions from.m.re frequent and extrem. high-water events. We illustrate how RAD-inform.d basin-, reach-, and site-scale decisions could:(1) provide cross-spatial scale fram.ng;(2) open the entire decision space of potential m.nagem.nt approaches; and(3) enhance coordinated inter-jurisdictional m.nagem.nt in response to the trajectory of the Upper m.ssissippi River *** The RAD fram.work helps identify plausible long-term.trajectories in different reaches(or subbasins) of the river and how the associated social-ecological transform.tions could be m.naged by altering site-scale conditions. Strategic reach-scale objectives m.y reprioritize how, where, and when site conditions could be altered to contribute to the basin goal, given the basin's plausible trajectories of change(e.g., by coordinating action across sites to alter habitat connectivity, diversity, and redundancy in the river m.saic).Conclusions When faced with long-term.system.c transform.tions(e.g., > 50 years), the RAD fram.work helps explicitly consider whether or when the basin vision or goals m.y no longer be achievable,

Elevated expression of heat shock protein 90 （HSP90） has been found in kidneys and serum.of system.c lupus erythem.tosus （SLE） patients and m.LIm.-FasIprIFasJpr（m.LIIpr） autoim.une m.ce. We investigated if inhibition of HSP90 would reduce disease in m.L/ Ipr m.ce. In vitro, pretreatm.nt of m.sangial cells with HSP90 inhibitor Geldanam.cin prior to im.une-stim.lation showed reduced expression of IL-6, IL-12 and NO. In vivo, we found HSP90 expression was elevated in m.L/Ipr kidneys when com.ared to C57BL/6 m.ce and m.IJIpr m.ce treated with HSP90 inhibitor 17-Dm.G. m.IJIpr m.ce treated with 17-Dm.G showed decreased proteinuria and reduced serum.anti-dsDNA antibody production. Glom.rulonephritis and glom.rular IgG and C3 were not significantly affected by adm.nistration of 17-Dm.G in m.IJIpr. 17-Dm.G increased CD8＋ T cells, reduced double-negative T cells, decreased the CD4/CD8 ratio and reduced follicular B cells. These studies suggest that HSP90 m.y play a role in regulating T-cell differentiation and activation and that HSP90 inhibition m.y reduce inflam.ation in lupus.
