The LysR Transcription Factor, HexS, Is Required for Glucose Inhibition of Prodigiosin Production by <i>Serratia marcescens</i>

Generation of many useful microbe-derived secondary metabolites, including the red pigment prodigiosin of the bacterium Serratia marcescens, is inhibited by glucose. In a previous report, a genetic approach was used to determine that glucose dehydrogenase activity (GDH) is required for inhibiting prodigiosin production and transcription of the prodigiosin biosynthetic operon (pigA-N). However, the transcription factor(s) that regulate this process were not characterized. Here we tested the hypothesis that HexS, a LysR-family transcription factor similar to LrhA of Escherichia coli, is required for inhibition of prodigiosin by growth in glucose. We observed that mutation of the hexS gene in S. marcescens allowed the precocious production of prodigiosin in glucose-rich medium conditions that completely inhibited prodigiosin production by the wild type. Unlike previously described mutants able to generate prodigiosin in glucoserich medium, hexS mutants exhibited GDH activity and medium acidification similar to the wild type. Glucose inhibittion of pigA expression was shown to be dependent upon HexS, suggesting that HexS is a key transcription factor in secondary metabolite regulation in response to medium pH. These data give insight into the prodigiosin regulatory pathway and could be used to enhance the production of secondary metabolites.

This thesis presents an exploratory research by analyzing the welfare effects resulted from initial stages of implementation of South North Water Transfer Project （SNWTP）, the middle Route plan, water source in Danjiangkou and Bai Long town in Hubei Province in China. The effects to Land, Supporting Projects, Local Government budget and Income distribution among people of the surveyed area and proposition over the means to resolve these effects have been pointed *** many researches have been done on this project, which is going to alleviate water scarcity for 300-325m people in the northern China and said to be economically accepted by some important organs, such as World Bank and the Chinese government. The question of welfare satisfaction has not been fully explored in the course of implementation of this project. This study points out the effects that have faced similar projects inside China and outside, and describing the impacts that are already experienced in Danjiangkou and Bai Long town on the aforementioned *** studies, of selected cases of Danjiangkou and Bai Long town have been studied in this research. The selected sample was restricted to those who have somewhat be affected by the initial stages of implementation in Danjiangkou City. Formal and informal interviews ware conducted to gather perception of the people and experiences regarding the project execution. more data have been obtained from Hubei Provincial, and respective local government statistics bureau. The information processed by SPSS （Statistical Package for Social Sciences） and results used in the descriptive analysis to indicate the extent of the *** result from descriptive analysis indicates that 61.4% of the respondents agreed that, there exist negative effects among the independent variables;hence negative welfare effects resulted from implementing this gigantic project. Development of an Integrated Tool for Small-Scale maize Farming in Uganda

About 62% of the maize produced in developing countries is cultivated manually owing to limited access to appropriate technology. Available technologies perform a single operation, necessitating farmers to buy multiple implements, which is expensive. In this study, an ox-drawn tool integrating a plough, planter and cultivator for small-scale maize farming was developed and its performance evaluated. Planting and cultivating units were designed and fabricated for assembly onto an existing standard ox-plough beam. The integrated tool was tested in the field to assess the effectiveness and efficiency of cultivation and planting of maize. Results showed that the planting unit had an average seed rate of 35.4 kg/ha with a seed damage of 4% and an effective field capacity of 0.15 ha/h at a field efficiency of 87.3%. The cultivating unit had an effective field capacity of 0.1 ha/h and a weeding efficiency of 86%. The efficiency, effectiveness and reliability for field operations are superior to manual operations currently used by farmers and are comparable to single-unit prototypes developed in other countries. This demonstrates the technical feasibility of integrating planting and cultivation units onto an existing mould board plough instead of buying multiple tools.

We report a significantly-enhanced bioinformatics suite and database for proteomics research called Yale Protein Expression Database（YPED） that is used by investigators at more than 300 institutions worldwide. YPED meets the data management, archival, and analysis needs of a high-throughput mass spectrometry-based proteomics research ranging from a singlelaboratory, group of laboratories within and beyond an institution, to the entire proteomics community. The current version is a significant improvement over the first version in that it contains new modules for liquid chromatography–tandem mass spectrometry（LC–mS/mS） database search results, label and label-free quantitative proteomic analysis, and several scoring outputs for phosphopeptide site localization. In addition, we have added both peptide and protein comparative analysis tools to enable pairwise analysis of distinct peptides/proteins in each sample and of overlapping peptides/proteins between all samples in multiple datasets. We have also implemented a targeted proteomics module for automated multiple reaction monitoring（mRm）/selective reaction monitoring（SRm） assay development. We have linked YPED＇s database search results and both label-based and label-free fold-change analysis to the Skyline Panorama repository for online spectra visualization. In addition, we have built enhanced functionality to curate peptide identifications into an mS/mS peptide spectral library for all of our protein database search identification results.

人的肠道微生物群落由细菌、真菌、病毒等微生物共同组成,影响人体多种生理或病理过程。相较于细菌组分,目前对于人体肠道菌群中的真菌组分认识严重不足。我们通过将高精度分析算法引入真菌ITS序列分析,结合培养组学和比较基因组学分析,建立了一套可靠的真菌菌群分析平台,并以此为基础对造血干细胞移植初期的患者肠道真菌动态变化进行时序性分析。结合临床数据,我们发现移植前后肠道真菌DNA的载量和多样性并无显著区别,但其组分发生了巨大变化,其中有近20%的患者在移植后免疫抑制期出现明显的近平滑念珠菌定植扩增,同时具有特殊的细菌菌群组分变化。尤为重要的是,具有近平滑念珠菌肠道定植的患者比其他患者具有显著恶化的总存活率,以及更高的真菌菌血症发生风险。通过比较念珠菌菌血症患者血液分离株和肠道分离株的全基因组序列,我们发现血液感染发生前一周左右,造成感染的菌株就已经在肠道中发生定植。综上所述,本项研究系统分析了造血干细胞移植过程中人体肠道真菌的动态变化,预示着精准干预肠道病原真菌的定植扩增可以有效预防血液感染,并提高患者总存活率。

Hemorrhagic lumbar synovial facet cyst secondary to transforaminal epidural injection:A case report and review of the literature

A 64-year-old-female presented with progressive left foot weakness, low back and radicular pain after a left sided S1 transforaminal epidural steroid injection(ESI). magnetic resonance imaging revealed left side L5-S1 large extradural heterogeneous mass with layering areas suggesting different stages of hematoma formation. Past medical history was significant for peripheral vascular disease and transient ischemic attacks, for which she took aspirin and clopidogrel(antiplatelet agent). These medications were discontinued one week prior to ESI. Although synovial cysts associated with facet arthropathy are common, hemorrhagic cyst is not. To the best of the authors' knowledge, this is the first reported case of symptomatic hemorrhagic lumbar facet synovial cyst following ESI on a patient taking antiplatelet medications.

Effect of preoperative biliary drainage on outcome of classical pancreaticoduodenectomy

AIm: To investigate the role of preoperative biliary drainage (PBD) in the outcome of classical pancreaticodu odenectomy. mETHODS: A 10-year retrospective data analysis was performed on patients (n = 48) undergoing pancreaticoduodenectomy from march 1994 to march 2004 in department of surgery at SmS medical college, Jaipur, India. Demographic variables, details of preoperative stenting, operative procedure and post operative complications were noted. RESULTS: Preoperative biliary drainage was performed in 21 patients (43.5%). The incidence of septic complications was significantly higher in patients with biliary stent placement (P minor biliary leak rate. mortality and hospital stay in each group was comparable. CONCLUSION: Within this study population the use of PBD by endoscopic stenting was associated with a high incidence of infective complications. These findings do not support the routine use of biliary stenting in patients prior to pancreatico-duodenectomy.

长江下游镇江江段是鱼类种群密集、资源丰富、种类多样、珍稀水生动物广布的江段,大量经济鱼类在此集中产卵,同时也是我国主要经济和珍稀洄游鱼类在生活史过程中产卵、索饵和越冬洄游的必经之道,因此其渔业地位十分重要,历来是我国主要的天然渔业场所。20世纪90年代后,随着经济的快速发展,人类涉水活动趋于频繁,长江下游水文情势、水位、水温和泥沙等发生改变,水利枢纽工程建设导致鱼类等水生生物栖息环境改变,自然的洄游通道被阻断,面对诸多不利于种群的因素,鱼类群落将会如何响应有待研究,因此亟待加强对长江鱼类群落结构及其多样性的系统研究。本研究对长江镇江水域鱼类群落结构进行周年调查研究,同时对水域环境因子进行同步采样,以探究影响鱼类群落结构的关键环境因子;同时对两种重要经济鱼类（鳊和刀轿）的个体生物学进行研究,以期掌握长江下游镇江江段渔业资源现状及重要经济鱼类的生长繁殖特性,为该江段鱼类资源的合理利用与渔业资源恢复提供科学依据。分别于2018年1月、3月、5月、7月、9月和11月,在长江镇江段和畅洲北岔附近水域设置了3个采样点放置渔具（6个大插网、3个小插网和18个虾笼网箱）进行24小时常规监测,此外还放置6条刺网约12小时,用以调查鱼类多样性和优势种。水环境参数监测与鱼类调查同时进行,主要测定的是水体理化参数,包括水温、水深、溶解氧、浊度等物理参数,水体营养盐包括TN、TDN、TP、TDP、N02--N、NH4+-N和P043--P等,生物环境主要包括浮游植物、浮游动物和底栖动物等。水环境调查结果显示,调查水域水温变化范围为9.47-23.1℃,平均温度为16.91±5.62℃;水深在9.17-23.58 m之间变化,平均水深为15.13±4.79 m。总氮变化范围为1.98-2.44 mg/L,平均TN为2.26±0.16 mg/L,总体上超过Ⅲ类水标准。水体氨氮变化范围为0.04-0.35 mg/L,平均氨氮为0.15±0.11 mg/L。水体卡尔森营养状态指数变幅为47.46-51.28,均值49.04±1.07,水体为中度富营养化。浮游植物58种,隶属于6门19科29属,其中硅藻门物种最多,为31种,占物种总数的53.45%;绿藻门次之,共13种,占22.41%,裸藻门、隐藻门、甲藻门和蓝藻门物种较少,分别为3种、2种、1种和8种。浮游动物4门16属20种,原生动物最多,共4属6种,占物种总数的比例为30%;其次为桡足类,6属8种占40%;枝角类4属4种占20%;轮虫类2属2种,占10%。底栖动物2门8种,环节动物门最多为6种,占总物种数的75%;节足动物门2种,占总物种数的25%。鱼类调查共采集渔获物498.35kg,2249尾,长江镇江段鱼类区系主要有四种:中国平原区系复合体、南方热带平原区系复合体、晚第三纪早期区系复合体和南黄海、东海近海类群。渔获物共52种,隶属于8目13科38属,其中包含鱼类49种,甲壳类3种。鲤形目物种的渔获物种数、渔获尾数和渔获重量均极显著高于其他各目鱼类,占据绝对优势,分别达69.23%、81.68%、93.42%。IRI指数大于1000的优势种共5种,分别为鲢、鳊、鳙、似鳊和草鱼;IRI指数介于100至1000之间的常见种共10种,IRI指数介于10至100的偶见种共计16种,IRI指数小于10的罕见种共计18种。长江下游镇江江段共采集到鳊共327尾,体长范围是101.93-245.84 mm,平均体长为 160.22±33.15 mm;雌性群体共体长范围为 101.93-245.8 mm,平均体长为155.50±34.02 mm,优势组体长为101.93-189.77 mm,占雌性群体82.32%,体长范围220-250 mm的个体最少,仅占雌性群体的6.08%;雄性群体体长范围为105.81-24.79 mm,平均体长166.07±31.6 mm,优势体长为130.77-189.58 mm,占雄性群体67.12%,体长范围221.6-244.79mm的个体最少,仅占雄性群体的6.85%。刀鲚88尾,体长范围是221.38-391 mm,平均体长 288.99±33.56 mm 优势组为 243.84-318.68 mm,占群体73.86%,体长范围大于360 mm的个体最少,仅占雌性群体的1.14%。体重范围为38.83-198.03 g,平均体重 93.92±33.79 g,优势体重组为51.44-146.85 g,占群体 76.14%,体重范围38.83-47.84 g的个体最少,仅占雌性群体的9.1%。根据调查结果,长江下游镇江江段鱼类多样性、密度及资源量较高,水域环境条件良好,饵料资源丰富且分布量较大的区域,鱼类群落结构复杂,群落多样性较大。长江镇江段鱼类群落结构与水域生态环境间的相关关系,还
