The cytokine granulocyte-macrophage-colony stimulating factor (GM-CSF) possesses the capacity to differentiate monocytes into macrophages (MØs) with opposing functions, namely, proinflammatory M1-like MØs and immunosuppressive M2-like MØs. despite the importance of these opposing biological outcomes, the intrinsic mechanism that regulates the functional polarization of MØs under GM-CSF signaling remains elusive. Here, we showed that GM-CSF-induced MØ polarization resulted in the expression of cytokine-inducible SH2-containing protein (CIS) and that CIS deficiency skewed the differentiation of monocytes toward immunosuppressive M2-like MØs. CIS deficiency resulted in hyperactivation of the JAK-STAT5 signaling pathway, consequently promoting downregulation of the transcription factor Interferon Regulatory Factor 8 (IRF8). Loss- and gain-of-function approaches highlighted IRF8 as a critical regulator of the M1-like polarization program. In vivo, CIS deficiency induced the differentiation of M2-like macrophages, which promoted strong Th2 immune responses characterized by the development of severe experimental asthma. Collectively, our results reveal a CIS-modulated mechanism that clarifies the opposing actions of GM-CSF in MØ differentiation and uncovers the role of GM-CSF in controlling allergic inflammation.

Impact of pre- and peri-operative risk factors on length of stay and hospital readmission following minimally-invasive partial nephrectomy

HdPairFinder:A data processing platform for hydrogen/deuterium isotopic labeling-based nontargeted analysis of trace-level amino-containing chemicals in environmental water

The combination of hydrogen/deuterium(H/d)formaldehyde-based isotopic methyl labeling with solid-phase extraction and high-performance liquid chromatography–high resolution mass spectrometry(HPLC-HRMS)is a powerful analytical solution for nontargeted analysis of trace-level amino-containing chemicals in water *** the huge amount of chemical information generated in HPLC-HRMS analysis,identifying all possible H/dlabeled amino chemicals presents a significant challenge in data *** address this,we designed a streamlined data processing pipeline that can automatically extract H/d-labeled amino chemicals from the raw HPLC-HRMS data with high accuracy and ***,we developed a cross-correlation algorithm to correct the retention time shift resulting from deuterium isotopic effects,which enables reliable pairing of H-and d-labeled ***,we implemented several bioinformatic solutions to remove false chemical features generated by in-source fragmentation,salt adduction,and natural13C ***,we used a data mining strategy to construct the AMINES library that consists of over 38,000 structure-disjointed primary and secondary amines to facilitate putative compound ***,we integrated these modules into a freely available R program,*** rationale of each module was justified and its performance tested using experimental H/d-labeled chemical standards and authentic water *** further demonstrated the application of HdPairFinder to effectively extract N-containing contaminants,thus enabling the monitoring of changes of primary and secondary N-compounds in authentic water *** is a reliable bioinformatic tool for rapid processing of H/d isotopic methyl labeling-based nontargeted analysis of water samples,and will facilitate a better understanding of N-containing chemical compounds in water.

Influence of cardiorespiratory fitness and body composition on resting and post-exercise indices of vascular health in young adults

Poor cardiorespiratory fitness may mediate vascular impairments at rest and following an acute bout of exercise in young healthy *** study aimed to compare flow mediated dilation(FMd)and vascular augmentation index(AIx75)between young adults with low,moderate,and high levels of cardiorespiratory fitness before and after an acute bout of aerobic ***-three participants(22 men;21 women)between 18 and 29 years of age completed the *** were classified into low,moderate,and high health-related cardiorespiratory fitness groups according to age-and sex-based relative maximal oxygen consumption(V_O2 max)percentile *** was performed using doppler ultrasound and AIx75 was performed using pulse wave analysis at baseline and 60-min after a 30-min bout of treadmill running at 70%V_O2 max.A significant interaction(p=0.047;ηp 2=0.142)was observed,with the moderate fitness group exhibiting a higher FMd post-exercise compared with baseline([6.7%3.1%]vs.[8.5%2.8%],p=0.028;d=0.598).We found a significant main effect of group for AIx75(p=0.023;ηp 2=0.168),with the high fitness group exhibiting lower AIx75 compared to low fitness group([10%10%]vs.[2%10%],respectively,p=0.019;g=1.07).This was eliminated after covarying for body fat percentage(p=0.489).Our findings suggest that resting FMd and AIx75 responses are not significantly influenced by cardiorespiratory fitness,but FMd recovery responses to exercise may be enhanced in individuals with moderate cardiorespiratory fitness levels.

Therapeutic advances in neural regeneration for huntington’s disease

huntington’s disease is a neurodegenerative disease caused by the expansion mutation of a cytosine-adenine-guanine triplet in the exon 1 of the HTT gene which is responsible for the production of the huntingtin (Htt) protein. In physiological conditions, Htt is involved in many cellular processes such as cell signaling, transcriptional regulation, energy metabolism regulation, dNA maintenance, axonal trafficking, and antiapoptotic activity. When the genetic alteration is present, the production of a mutant version of Htt (mHtt) occurs, which is characterized by a plethora of pathogenic activities that, finally, lead to cell death. Among all the cells in which mHtt exerts its dangerous activity, the GABAergic Medium Spiny Neurons seem to be the most affected by the mHtt-induced excitotoxicity both in the cortex and in the striatum. However, as the neurodegeneration proceeds ahead the neuronal loss grows also in other brain areas such as the cerebellum, hypothalamus, thalamus, subthalamic nucleus, globus pallidus, and substantia nigra, determining the variety of symptoms that characterize huntington’s disease. From a clinical point of view, huntington’s disease is characterized by a wide spectrum of symptoms spanning from motor impairment to cognitive disorders and dementia. huntington’s disease shows a prevalence of around 3.92 cases every 100,000 worldwide and an incidence of 0.48 new cases every 100,000/year. To date, there is no available cure for huntington’s disease. Several treatments have been developed so far, aiming to reduce the severity of one or more symptoms to slow down the inexorable decline caused by the disease. In this context, the search for reliable strategies to target the different aspects of huntington’s disease become of the utmost interest. Background:Cyclosporine-A(Cs-A),an immunosuppressant drug indicated for organ transplant patients has been associated with toxicities arising from activation of pathological *** Ginkgo biloba supplement(GBS)is an antioxidant-enhancing herbal product widely used for treating neurological and vascular ***:This study investigated the ameliorative effect of GBS on cyclosporine-A induced cardiotoxicity in Wistar ***:The study included 20 Wistar rats(n=5)and they were treated as follows:group 1 received distilled water(10 mL/kg),group 2 received Cs-A(25 mg/kg),group 3 received GBS(50 mg/kg)and group 4 received Cs-A(25 mg/kg)and GBS(50 mg/kg).All treatment were done intraperitoneal for 14 days and thereafter,animals were euthanised and heart tissues were harvested for biochemical assays as well as immunohistochemical *** was also collected for serum biochemical ***:Our results revealed that GBS reduces Cs-A induced increase of AST,ALT,ALP,LdH,and GGT levels in serum relative to Cs-A ***,increased oxido-nitrergic markers like MdA,NO,together with elevated concentrations of cholesterol,triglycerides,LdL,creatine kinase-MB,troponin-I,as well as pro-inflammatory cytokines(IL-6,and TNF-α)and heart weight index were reduced by *** Cs-A induced suppression of low-density lipoprotein,antioxidant(GSH,SOd,CAT),Na-K ATPase activities and antiapoptotic element,Bcl-2 were increased by ***,signalling pathways including ERK1/2,and mTOR as well as ventricular thickness were significantly alleviated by GBS ***:The findings suggest that GBS ameliorated Cs-A-induced cardiotoxicity by activating mTOR/ERK1/2 signalling pathways.

Two-dimensional(2d)inorganic/organic hybrids provide a versatile platform for diverse applications,including electronic,catalysis,and energy storage *** recent surge in 2d covalent organic frameworks(COFs)has introduced an organic counterpart for the development of advanced 2d organic/inorganic hybrids with improved electronic coupling,charge separation,and carrier ***,existing synthesis methods have primarily focused on few-layered film structures,which limits scalability for practical ***,we present a general synthesis approach for a range of COF/inorganic 2d material hybrids,utilizing 2d inorganic materials as both catalysts and inorganic building *** leveraging the intrinsic Lewis acid sites on the inorganic 2d materials such as hexagonal boron nitride(hBN)and transition metal dichalcogenides,COFs with diverse functional groups and topologies can grow on the surface of inorganic 2d *** controlled 2d morphology and excellent solution dispersibility of the resulting hybrids allow for easy processing into films through vacuum *** proof of concept,hBN/COF films were employed as filters for Rhodamine 6G removal under flow-through conditions,achieving a removal rate exceeding 93%.The present work provides a simple and versatile synthesis method for the scalable fabrication of COF/inorganic 2d hybrids,offering exciting opportunities for practical applications such as water treatment and energy storage.
