Pakl mediates the stimulatory effect of insulin and curcumin on hepatic ChREBP expression

Insulin can stimulate hepatic expression of carbohydrate-responsive element-binding protein （ChREBP）. As recent studies revealed potential metabolic beneficial effects of ChREBP, we asked whether its expression can also be regulated by the dietary polyphenoi curcumin. We also aimed to determine mechanisms underlying ChREBP stimulation by insulin and curcumin. the effect of insulin on ChREBP expression was assessed in mouse hepatocytes, while the effect of curcumin was assessed in mouse hepatocytes and with curcumin garage in mice. Chemical inhibitors for insulin signaling molecules were utilized to identify involved signaling molecules, and the involvement of p21-activated protein kinase 1 （Pakl） was determined with its chemical inhibitor and Pokl-/- hepatocytes. We found that both insulin and curcumin-stimulated ChREBP expression in Akt-independent but MEK/ERK-dependent manner, involving the inactivation of the transcriptional repressor Oct-1. Aged Pokl-/- mice showed reduced body fat volume. Pakl inhibition or its genetic deletion attenuated the stimulatory effect of insulin or curcumin on ChREBP expression. Our study hence suggests the existence of a novel signaling cascade Pakl/MEK/ERK/Oct-1 for both insulin and curcumin in exerting their glucose-lowering effect via promoting hepatic ChREBP production, supports the recognition of beneficial functions of ChREBP, and brings us a new overview on dietary polyphenols.

Solution-processed n-type Bi_2te_(3-x)Se_x nanocomposites with enhanced thermoelectric performance via liquid-phase sintering

the much slower progress in enhancing the thermoelectric performance of n-type Bi2te3 than that of p-type Bi2te3 based materials in the past decade hinders the widespread use in power generation and refrigeration. Here, a facile bottom-up solution-synthesis with spark plasma sintering(SPS) process has been developed to build n-type Bi2te3-xSex bulk nanocomposites, which substantially improves the power factor and decreases the lattice thermal conductivity by tuning the interface scattering of phonons and electrons. the stoichiometric composition in ternary Bi2te3-xSex nanocomposites is also tuned to optimize the carrier concentration and lattice thermal conductivity. the optimized bulk nanocomposite Bi2te2.7Se0.3 exhibits a Zt of 1.1 at^371 K, which is comparable to the corresponding commercially available ingots. Our results demonstrate the great potential of the solution-processed n-type Bi2te3-xSex nanocomposites for cost-effective thermoelectric applications.

＜正＞the rotational/translational actuator （RtAC） system has been served as a testbed for evaluating various nonlinear control methodologies. this paper will study the asymptotic disturbance rejection problem of the RtAC system which aims to design a feedback controller such that the closed-loop system is stable and the output of the closed-loop system can asymptotically approach zero in spite of a sinusoidal disturbance. this problem cannot be solved by conventional inversion based control methods since RtAC is a nonminimum phase system. this paper will formulate this problem as an output regulation problem, and solve it approximately by applying a neural network based approach developed recently by the authors. BACKGROUND tumor necrosis factor-alpha inhibitors,including infliximab and adalimumab,are effective medical treatments for perianal fistulising Crohn’s disease(CD),but not all patients achieve fistula *** to determine the correlation between perianal fistula healing and closure with infliximab and adalimumab trough *** In this multicentre retrospective study conducted across four tertiary inflammatory bowel disease centres in Australia,we identified CD patients with perianal fistulae on maintenance infliximab or adalimumab who had a trough level within twelve weeks of clinical *** collected included demographics,serum infliximab and adalimumab trough levels(mg/L)within 12 wk before or after their most recent clinical assessment and concomitant medical or surgical *** primary outcome was fistula healing,defined as cessation in fistula *** secondary outcome was fistula closure,defined as healing and closure of all external fistula *** between patients who did or did not achieve fistula healing were compared using the chi-square test,t test or Mann-Whitney U *** One hundred and fourteen patients(66 infliximab,48 adalimumab)were ***-eight(72.7%)patients on maintenance infliximab achieved fistula healing and 18(27.3%)achieved fistula ***-seven(77%)patients on maintenance adalimumab achieved fistula healing and 17(35.4%)achieved fistula *** who achieved fistula healing had significantly higher infliximab and adalimumab trough levels than patients who did not[infliximab:6.4(3.8-9.5)vs 3.0(0.3-6.2)mg/L,P=0.003;adalimumab:9.2(6.5-12.0)vs 5.4(2.5-8.3)mg/L,P=0.004].For patients on infliximab,fistula healing was associated with lower rates of detectable anti-infliximab antibodies and younger *** patients on adalimumab,fistula healing was associated with higher rates of combination therapy with an *** trough levels for patients with and without fistu

Grain boundaries in organic-inorganic halide perovskite solar cells(PSCs)have been found to be detrimental to the photovoltaic performance of ***,we develop a unique approach to overcome this problem by modifying the edges of perovskite grain boundaries with flakes of high-mobility two-dimensional(2D)materials via a convenient solution process.A synergistic effect between the 2D flakes and perovskite grain boundaries is observed for the first time,which can significantly enhance the performance of *** find that the 2D flakes can conduct holes from the grain boundaries to the hole transport layers in PSCs,thereby making hole channels in the grain boundaries of the ***,2D flakes with high carrier mobilities and short distances to grain boundaries can induce a more pronounced performance enhancement of the *** work presents a cost-effective strategy for improving the performance of PSCs by using high-mobility 2D materials.

Modified triphenylmethane-based Polyimides with Improved Optical,Dielectric and Solubility Properties via Post-Polymerization Modification

A family of new triphenylmethane(tPM)-based polyimides(PIs)containing bulky tert-butyldimethylsiloxy(tBS)side-groups(PI-tPMOSis)has been prepared by a post-polymerization modification via a simple silyl ether reaction of tPM-based PIs containing hydroxyl(OH)groups(PI-tPMOHs).the attachment of tBS side-groups in PI-tPMOSis can be achieved up to 100%,as confirmed by the 1H-NMR and IR *** to the presence of the tPM structure,PI-tPMOSi films still display the excellent thermal stability with high glass transition temperature(tg)of 314–351°C and high degradation temperature(td5%)of 480–501°*** is quite remarkable that the introduction of tBS side-groups into PI-tPMOSi chains results in more superior optical,dielectric and solubility properties in comparison with the precursor PI-tPMOH films,probably due to the reductions of the packing density and charge-transfer complexes(CtCs)*** optical transmittance at 400 nm(t400)of PI-tPMOSi films is significantly increased from 45.3%–68.8%to 75.4%–81.6%of the precursor PI-tPMOH *** dielectric constant(Dk)and dissipation factor(Df)at 1 MHz of PI-tPMOSi films are reduced from 4.11–4.40 and 0.00159–0.00235 to 2.61–2.92 and 0.00125–0.00171 of the precursor PI-tPMOH films,*** the molecular design and simple preparation method,this study provides an effective approach for enhancement of various properties of PI films for microelectronic and photoelectric engineering applications.

Clinical results of radical prostatectomy for patients with prostate cancer in Macao

Background Incidence of prostate cancer has been increasing in recent decades. In the year 2005, prostate cancer became the second most common cancer in males in Macao. the purpose of this report was to review and summarize the clinical features and prognosis of the 54 patients undergoing radical prostatectomy in Macao Special Administrative Region （SAR）, China.Methods From November 2000 to November 2006, retropubic radical prostatectomy were performed in 54 cases for the treatment of prostate cancer. the mean age of patients was 69.8 years （range from 54 to 79）. the preoperative prostate specific antigen （PSA） level, postoperative pathologic stage and Gleason＇s score, operation duration, intraoperative bleeding and intraoperative and postoperative complications were reported. the follow-up duration was 3 months to 6.25 years with a mean of 2.1 years. Postoperative parameters including PSA alteration, biochemical recurrence, local recurrence, distant metastasis and mortality were observed.Results Most of the patients in our study were diagnosed as localized prostate cancer. the patients＇ preoperative serum PSA was 0-4.0 ng/ml （16.7%）, 4.0-10.0 ng/ml （51.8%）, 10.1-20.0 ng/ml （24.1%） and above 20.0 ng/ml （7.4%）. the tNM stage t1a ＋ t1b comprised 7.6% of patients, stage t2a ＋ t2b comprised 20.3%, stage t2c 38.9%, stage t3a 20.3% and over t3a only 12.9%. there were 9.5% cases with Gleason scores of 2-4, 41.5% with scores of 5-6, 30.2% with scores of 7 and 18.8% with scores of 8-10. the average operative duration was 216 minutes and the average intraoperative bleeding was 760 ml. Intraoperative complications included one massive hemorrhage （1.9%）, one rectal injury （1.9%） and one obturator nerve injury （1.9%）. Early postoperative complications consisted of urinary incontinence （14 cases, 25.9%）, bladder neck stricture （5 cases, 9.3%）, acute urinary retention （4 cases, 7.4%）, pelvic effusion （2 cases, 3.8%）, lymphocele （1 case, 1.9%） and vesicorectal fistula （only 1 c
