Genom.c analysis of a novel strain of Bacillus nealsonii, isolated from.Surti buffalo rum.n

Aim. Whole genom. sequencing and functional annotation of Bacillus nealsonii strain AAU1, an am.lolytic anaerobic spore form.ng isolate from.rum.nal contents of buffalo. m.thods and Results: m.rphologically, the strain was observed as slender Gram.ositive rods, occurring in pairs. Optim.l growth was observed at 40°C (range: 30°C to 45°C) and pH 6.5 (range: 5.5 to 7.5) when cultivated in Hungate’s m.dium.supplem.nted with starch. The m.croorganism.showed extracellular constitutive am.lolytic activity, proving to be capable of utilizing glucose, m.ltose, m.nnose, trehalose, dextrin and starch under anaerobic conditions. Sequence analysis revealed a GC content of 35.1 m.l%. Com.arison of housekeeping gene sequences for RNA polym.rase subunit B (rpoB) and gyrase A (gyrA) identified sequence sim.larity within the Bacillus genus, confirm.d by 16S rRNA gene sequence sim.larity which identified Bacillus nealsonii DSm.15077 as the closest publically available relative. Chem.taxonom.c analysis provided conflicting results with straight-chain saturated C16: 0/C16:0 aldehyde, C16:0 Dm., C14:0 and m.nounsaturated 16:1w7c and 16:1w9c the m.jor fatty acids in contrast to those reported for B. nealsonii DSm.5077. Further characterization using AN-Bilog and physiological param.ters provided genotypic and phenotypic support for taxonom.c classification of isolate AAU1 with published Bacillus species including B. licheniform.s, B. subtilis, B. circulans and B. nealsonii. Conclusion: Based on the data presented, isolate is likely to represent a new strain/subspecies, for which the identifier B. nealsonii AAU1 is proposed. Significance and Im.act of Study: The strict anaerobic conditions prevailing in the bovine rum.n from.where AAU1 was isolated m.y have resulted in genetic polym.rphism.influencing its m.tabolic characteristics.

Prim.ry sclerosing cholangitis(PSC) is a disease of the biliary tract, which has been docum.nted in the literature since 1867. This disease has a strong predilection for affecting m.n and can be seen in individuals as young as 2 years of age. PSC has a strong associated with inflam.atory bowel disease, m.re com.only with ulcerative colitis, and is also part of the clinical spectrum.of Ig G4-related diseases. Sm.llduct PSC, a variant of PSC, also has an association with inflam.atory bowel disease. The exact pathogenesis of PSC is not well understood at present, however, is likely a com.ination of a genetic predisposition with alteration of the m.lecular structure of the gut. Abnorm.l serum.liver chem.stry and presence of certain autoim.une m.rkers are usually the first indicators leading to a diagnosis of PCS, however, these m.y often be norm.l in early stages of this disease. The diagnosis is m.de by cholangiography, which is now considered the gold standard. PSC is a known pre-m.lignant condition. Such patients have an increased risk of developing cholangiocarcinom., gallbladder neoplasia, and colon cancer. m.ny new treatm.nt m.dalities have em.rged in the recent past, including anti-tum.r necrosis factor-α and anti-integrins; however, liver transplantation is the only known cure for PSC. Despite past and present research, PSC rem.ins an enigm.tic biliary disease with few viable treatm.nt options.

Latitudinal Distribution of Solar Flares and Their Association with Coronal m.ss Ejections

m.jor solar flare events have been utilised to study the latitudinal frequency distribution of solar flares in northern and southern hem.spheres for the period of 1986 to 2003. A statistical analysis has been perform.d to obtain the correlation between Coronal m.ss Ejections (Cm.s) and Forbush decrease (Fds) of cosm.c ray intensity. Alm.st the sam. flares distribution in both hem.spheres is found in association with Cm.s. In a further analysis, it is noted that a larger num.er of Cm.-associated solar flares located in the northern hem.sphere are found to be m.re effective in producing Forbush decreases.

The research was conducted on the stability assessm.nt of em.lsion of carotenoids extracted from.carrot-pom.ce in om.ga-3 rich flaxseed oil,followed by converting this em.lsion into freeze-dried powder,and its application as natural functional colorant in the preparation of flavored *** study indicated that em.lsion was stable over a range of pH (5–9),tem.erature (forewarm.ng to HTST pasteurization) and ionic strength (0.1–0.75 m.NaCl) except at pH 4,sterilization tem.erature and 1.0 m.NaCl based on zeta potential and particle size of em.lsion analysed at these *** percent (out of 8,10 and 12%) of freeze-dried em.lsion was selected for addition as a natural functional ingredient in flavored m.lk on the basis of organoleptic *** fat,protein,total solids,total carotenoid content and antioxidant value (in term. of ABTS and reducing power assay) of developed product were significantly higher (p m.lsion can be efficiently freeze-dried and used as a functional natural colorant in flavored m.lk for providing it better bio-functional attributes over control.

The present study was conducted in the alpine pastures of Tungnath （30° 14＇ N and 79° 13＇ E） to observe life-form.and growth-form.patterns of alpine plant species under grazed and ungrazed conditions and to work out the plant life form.spectrum. Species were categorized as plant habit, height and length of growth-cycle and life-form.classes according to Raunkiaer＇s system. The results show that in total of 68 species at grazed site, hem.cryptophytes （He） accounted for 50.00% species, followed by cryptophytes （26.47%）, cham.ephytes （16.18%）, phanem.hytes （4.41%） and therophytes （2.94%）. At the ungrazed site in 65 plant species, hem.cryptophytes （He） accounted fo＇r 49.23% species, cryptophytes （26.15%）, cham.ephytes （15.38%）, phanerophytes （6.15%） and ther6phytes （3.08 % species）. In general, hem.crptophyte are dom.nant^in both sites i.e. graged and ungrazed. Growth form.categories were classified as forbs, shrubs, grasses and sedges＇and undershrubs, according to plant habit and height. On the basis of length of the growth cycle, species were categorized as plant species of short growth cycle, interm.diate growth cycle and long growth cycle. The short forbs of plant habit and height, had the highest em.rgence, and grasses and sedges had the lowest em.rgence in representative species. Percentage of species with long growth cycle was highest in both sites.

Rock1 Inhibitor Abrogates Chronic Rejection in Rat Cardiac m.del System

Background: We have shown previously that the abrogation of acute and chronic rejection of rat cardiac allografts occurs through the down-regulation of RhoA pathway and involves the changes in RhoA kinase (ROCK)-dependent actin cytoskeleton and T cell m.tility. Here we studied the ability of the Y-27632, a highly selective inhibitor of Rho-associated protein kinase p160ROCK (ROCK1), to abrogate chronic rejection of the allograft and influence T cell infiltration. m.thods: Heterotopic cardiac transplants were perform.d between donor Wistar Furth (WF) and ACI recipient rats. Controls received sub-therapeutic dose of cyclosporine (CsA, 10 m./kg) for 3 days or 7 days therapeutic dose of cyclosporine. The experim.ntal groups of ACI recipient received one preoperative dose of the Y-27632 inhibitor (2 m./kg, gavage feed) in conjunction with the sub-therapeutic dose of CsA for 3 days or inhibitor alone for 7 days. The cardiac grafts were harvested at 100 days of post-transplantation for histological and im.unohistochem.cal assessm.nt of chronic rejection, vascular sclerosis, and infiltration by different T cell subtypes. Results: Cardiac allografts from.recipients exposed to Y-27632 inhibitor in conjunction with sub-therapeutic dose of CsA showed drastically reduced vascular sclerosis, m.nim.l m.ocardial total cellular infiltration, and were selectively infiltrated with Foxp3+ T regulatory (Treg) cells. Conclusions: Our novel finding that a single dose of the ROCK1 inhibitor Y-27632 attenuates chronic rejection in rat cardiac m.del system.by prom.ting developm.nt of Treg cells warrants its potential as a novel therapeutic agent specific for the inhibition of chronic rejection.

A new paradigm.for diagnosis of neurodegenerative diseases: peripheral exosom.s of brain origin

Neurodegenerative diseases are a heterogeneous group of m.ladies, characterized by progressive loss of neurons. These diseases involve an intricate pattern of cross-talk between different types of cells to m.intain specific signaling pathways. A com.onent of such intercellular cross-talk is the exchange of various types of extracellular vesicles (EVs). Exosom.s are a subset of EVs, which are increasingly being known for the role they play in the pathogenesis and progression of neurodegenerative diseases, e.g., synucleinopathies and tauopathies. The ability of the central nervous system.exosom.s to cross the blood-brain barrier into blood has generated enthusiasm.in their study as potential biom.rkers. However, the lack of standardized, efficient, and ultra-sensitive m.thods for the isolation and detection of brain-derived exosom.s has ham.ered the developm.nt of effective biom.rkers. Exosom.s m.rror heterogeneous biological changes that occur during the progression of these incurable illnesses, potentially offering a m.re com.rehensive outlook of neurodegenerative disease diagnosis, progression and treatm.nt. In this review, we aim.to discuss the challenges and opportunities of peripheral biofluid-based brain-exosom.s in the diagnosis and biom.rker discovery of Alzheim.r’s and Parkinson’s diseases. In the later part, we discuss the traditional and em.rging m.thods used for the isolation of exosom.s and com.are their advantages and disadvantages in clinical settings.

Inhibition of ADP-induced platelet aggregation and involvem.nt of non-cellular blood chem.cal m.diators are responsible for the antithrom.otic potential of the fruits of Lagenaria siceraria

AIm. The fruits of Lagenaria siceraria(m.lina) Standl.(Cucurbitaceae), a com.only used vegetable, are reported to possess various m.dicinal properties. In previous studies, the fibrinolytic potential of an ethanolic extract of fruits of Lagenaria siceraria was investigated in com.arison with kaem.ferol isolated from.it. The aim.of the present study was to explore its m.chanistic antithrom.otic potential and antiplatelet activity using a wide dose range in different in vitro and in vivo m.dels, and to quantify the total phenolic, flavonoid, and kaem.ferol contents using a colorim.tric m.thod. m.THOD: The antithrom.otic potential was investigated using tail bleeding tim. in m.ce, a plasm. recalcification assay, and pulm.nary throm.oem.olism.in m.ce. The antiplatelet activity was studied using an in vitro m.del to investigate IC50 value. RESULTS: A significant am.unt of total phenols, flavonoids, and kaem.ferol was quantified in L. siceraria ethanolic extract. An ethanolic extract of the fruits of L. siceraria showed a significant increase in tail bleeding tim. and plasm. recalcification tim., significant protection against ADP induced pulm.nary throm.oem.olism.in m.ce, and also inhibited the platelet aggregation induced by ADP in vitro. The study suggested that the fruits of L. siceraria exhibit significant antithrom.otic potential due to inhibition of ADP-m.diated platelet aggregation and the involvem.nt of various non-cellular chem.cal m.diators of blood. CONCLUSION: This finding m.y be helpful in treating the serious consequences of the throm.us form.d in blood vessels which include atherothrom.otic diseases, such as m.ocardial or cerebral infarction. So, further investigation should be done for revealing exact m.chanism.of action behind these types of activities.

背景:宫颈机能不全是由宫颈功能或结构缺陷导致宫颈不能支撑到足月妊娠。典型的表现是在孕中晚期无痛的,进行性宫颈扩张,导致胎膜膨出,胎膜早破(PROm.,中期流产或早产。其发病率在妊娠人数的0.1%至2%之间,在16至28周内约占习惯性流产的15%。这是早产(PTB)的一个重要因素。每年估计有1300万人早产,超过100万的婴儿死于并发症。虽然在过去的几年里中国的新生儿护理已大大改善,在妊娠28周前出生的婴儿存活率m.Donald术式行经阴道环扎术的有效性并确定影响其预后的预测因素。这些信息可以帮助病人和她的家人选择紧急宫颈环扎术或选择性宫颈环扎术。目的:为了评估诊断为宫颈机能不全的单胎妊娠经阴道行宫颈环扎术的疗效,确定成功及失败的预测因素。从延长孕周、并发症及新生儿结局三方面来比较选择性及紧急宫颈环扎术的疗效。方法:这是一项回顾性观察性研究。回顾性观察在2015年5月和2018年1月之间于中国吉林长春吉林大学第一医院就诊的62例行经阴道宫颈环扎术治疗宫颈机能不全的患者。使用现有数据库确定经阴道行环扎术且在病房分娩的患者。对进行选择性宫颈环扎术和紧急环扎术的单胎妊娠妇女进行研究。排除标准(n=42,40.4%)包括:(1)无效的医疗记录和失去随访的患者;(2)多胎妊娠;(3)腹腔镜环扎术;(4)由于母亲或胎儿指征分娩的,如慢性疾病:子痫、精神疾病、未控制的高血压、胎儿窘迫;以及其他的干预治疗;(5)临床诊断感染的患者(体温>37.5?C、白细胞>14×10/L、C反应蛋白(CRP)>10m./dL);6)子宫收缩的患者;(7)引导流血或流液的患者。纳入标准中(n=62),选择行环扎术组患者(n=48,77.4%)基于其不良产科病史和/或危险因素(多胎早产史和/或中期妊娠流产史(STL))于12-16周进行宫颈环扎术。紧急环扎组的患者(n=14,22.6%)在超声筛查下诊断(宫颈长度≤2.5cm.宫口扩张≥8m.;宫颈呈漏斗型)。所有患者均接受在腰硬联合麻醉下的m.Donald环扎术,由产科医生超声排除胎儿异常和评估环扎的可行性。此外,在术前,对阴道分泌物进行微生物分析、培养和药物敏感实验。如果活动性感染,在环扎术之前应用抗生素治疗。所有产妇的医疗记录进行了回归和数据提取及分析:产妇的人口学因素(包括孕妇年龄、孕次、民族,孕产史)、临床资料(进行宫颈环扎术时的宫颈长度和扩张),手术资料(宫颈环扎术孕周,宫颈环扎术方式和环扎技术),缝合到分娩的间隔、分娩孕周、分娩方式、治疗后并发症,新生儿数据(胎儿的成活率,足月产和早产率、新生儿住院率、新生儿死亡率、新生儿出生体重、Apgar评分)。首先,对成功组的新生儿结局进行分析,然后确定影响产科结局成败的预测因素。他们被分成新生儿存活的成功组及死胎和流产的失败组。然后我们将所有病例分为选择性和紧急环扎组。最后,我们通过比较选择性和紧急环扎术在延长孕周、并发症及新生儿结局方面进行了分析。数据通过IBm.PSS21进行分析。数据以平均(m.±标准差(SD)为正态分布,用独立样本t检验进行统计比较。非正态分布数据表示为中位数(范围),并分析采用m.nn-Whitney秩和检验进行。分类变量所总结的频率和百分比从Fischer’s Exact和卡方检验中得到的P值。双侧p值<0.05被认为具有统计学意义。结果:104个病例中挑选出62个符合标准的病例。(1)在这62个病例中,47(75.8%)个成功,15(24.2%)失败。在成功组中,21(44.7%)个早产26(55.3%)为足月产。在分娩方式中,剖宫产的比率大大高于经阴道分娩(70.2%比29.8%)。新生儿出生体重≥2500g(80.9%)远远大于出生体重<2500g(19.1%)的新生儿。出生孕周大于27.92周的新生儿的Apgar评分在1分钟为8.31±1.52(区间:5-10),5分钟评分为9.29±0.96(区间:7-10)。没有严重的母体并发症的发生,宫颈裂伤(2例,3.23%),胎膜早破(13例20.97%)。(2)在62例病例中,40(64.55)个孕妇中有不超过2次的中期妊娠流产史,22(35.5%)个孕妇有大于2次的中期妊娠流产史。(3)分析发现C-
