Objective:To identify the residue determinants of the serospecificity and sero-cross-reactivity of dengue virus(DENV)envelope protein domain 3(ED3),which contains two major putative epitopes of ***:We constructed ED3 from DENV3(3ED3)and DENV4(4ED3),and six epitope-grafted variants,where we transferred epitope 1(L304I,K305D,V309M,and S310A)and/or epitope 2(D383N,K384S,K387T,and N389H)of 4ED3 onto 3ED3 and *** albino mice aged 3-4 weeks were immunized against wildtype and epitope-grafted ED3 variants and anti-ED3 IgG antibody responses were determined using ***:Mouse immunization using 3ED3 and 4ED3 generated serotype-specific antisera,as ***,most epitope-grafted ED3s produced antisera serospecific to the template ED3 with little or no cross-recognition of ED3 of the serotype from which the epitopes were *** indicated that a mere grafting of the epitope was not sufficient to transfer serospecificity,contrary to our ***,one epitope-grafted ED3 mutant,where epitope 1 of 3ED3 was grafted onto 4ED3(4ED3^(epi1)),generated antisera that was serospecific to both 4ED3 and ***:The 4ED3^(epi1)is a chimeric ED3 that produces antisera possessing serospecificity to both 3ED3 and 4ED3 onto a common 4ED3 *** 4ED3^(epi1),therefore,provides a unique tool for analyzing serospecificity and sero-cross-reactivity in *** believe that chimeric ED3 may provide a template for future recombinant ED3 possessing serospecificity of multiple DENVs onto a single scaffold and may pave a way developing tri-and/or tetravalent anti-DENV antisera.

Background and aims:Several bile acids-based monotherapies have been developed for non-alcoholic steatohepatitis(NASH)treatment but clinical trial findings suggest that they do not satisfactorily improve NASH and liver fibrosis in many ***,we have shown that combining a gut-restricted apical sodium-bile acid transporter(ASBT)inhibitor GSK2330672(GSK)with adeno-associated virus(AAV)-mediated liver fibroblast growth factor 15(FGF15)overexpression provides significantly improved efficacy than either single treatment against NASH and liver fibrosis in a high fat,cholesterol,and fructose(HFCFr)diet-induced NASH mouse *** beneficial effects of the com-bined treatment can be attributed to the markedly reduced bile acid pool that reduces liver bile acid burden and intestinal lipid *** aim of this study is to further investigate if combining GSK treatment with the orally bioavailable obeticholic acid(OCA),which induces endogenous FGF15 and inhibits hepatic bile acid synthesis,can achieve similar anti-NASH effect as the GSKþAAV-FGF15 co-treatment in HFCFr-diet-fed *** and methods:Male C57BL/6J mice were fed HFCFr diet to induce NASH and liver *** effect of GSK,OCA,and GSKþOCA treatments on NASH development was compared and contrasted among all ***:Findings from this study showed that the GSKþOCA co-treatment did not cause persistent reduction of obesity over a 12-week treatment *** single treatment nor the GSKþOCA co-treatment reduce hepatic steatosis,but all three treatments reduced hepatic inflammatory cytokines and fibrosis by a similar *** GSKþOCA co-treatment caused a higher degree of total bile acid pool reduction(~55%)than either GSK or OCA treatment ***,such bile acid pool reduction was insufficient to cause increased fecal lipid *** GSKþOCA co-treatment prevented GSK-mediated induction of hepatic cholesterol 7alpha-hydroxylase but failed to induce ileal FGF15 expression.

Objective: To reveal the presence of methicillin resistant Staphylococcus aureus(S. aureus)(MRSA) in poultry samples and to determine the antibiogram pattern against five ***: Samples from different poultry farm of Chittagong city, Bangladesh were examined for S. aureus by different biochemical tests and confirmed as MRSA by identifying the presence of mec A gene using PCR. Antibiotic resistance pattern in S. aureus was determined by antibiotic disk diffusion ***: In this study, a total of 60 samples(30 from nasal swabs and 30 from cloacal swabs) were used, of which 54 were confirmed as S. aureus by different biochemical tests. Among these, 12 were confirmed as MRSA by detecting mec A gene using *** antibiogram study, both nasal and cloacal samples showed the highest resistance against penicillin-G and the lowest resistance was observed against ***: Based on the present study, it can be said that different antibiotics are used extensively in poultry that leads to MRSA and is alarming for human health.

Objective:To investigate the therapeutic effects of methanol extract of Citrus macroptera Montr,fruit inα-amylase inhibitory activity(in vitro)and hypoglycemic activity in normal and glucose induced hyperglycemic rats(in vivo).Methods:Fruits of Citrus macroptera without rind was extracted with pure methanol following cold extraction and tested for presence of phytochemical constituents,α-amylase inhibitory activity,and hypoglycemic effect in normal rats and glucose induced hyperglycemic ***:Presence of saponin,steroid and terpenoid were identified in the *** results showed that fruit extract had moderateα-amylase inhibitory activity[IC_(50)value=(3.638±0.190)mg/mL]as compared to *** at 500 mg/kg and 1000 mg/kg doses fruit extract significantly(P<0.05 and P<0.01 respectively)reduced fasting blood glucose level in normal rats as compared to glibenclamide(5 mg/kg).In oral glucose tolerance test,500 mg/kg dose significantly reduced blood glucose level(P<0.05)at 2 h but 1000 mg/kg dose significantly reduced blood glucose level at 2 h and 3 h(P<0.05 and P<0.01 respectively)whereas glibenclamide(5 mg/kg)significantly reduced glucose level at every hour after *** time effect is also considered extremely significant with F value=23.83 and P value=0.0001 in oral glucose tolerance ***:These findings suggest that the plant may be a potential source for the development of new oral hypoglycemic agent.

The current experiment was conducted for the simultaneous determination of several water-soluble vitamins like ribo- flavin (vitamin B2), niacin (vitamin B3), pantothenic acid (vitamin B5) and pyridoxine (vitamin B6) in five highly con- sumed local leafy vegetables named as bottle gourd leaves (Lagenaria vulgaris) (local name Lau shak), green amaranth leaves (Amaranthus viridis) (local name Data shak), red amaranth leaves (Amaranthus gangeticuss) (local name Lal shak), Indian spinach (Basella alba) (local name Pui shak) and bitter gourd leaves (Momordica charantia) (local name Korola shak). The analyses were performed by HPLC using an analytical reversed phase C-18 (ODS column, 250 × 4.6 mm, 5 μm, Phenomenex, Inc.) column with the mobile phase consisting of a mixture of buffer (hexane sulphonic acid sodium, potassium dihydrogen phosphate and triethylamine, pH 3.0) and methanol in the ratio of 96:4 (v/v) at a flow rate of 1 mL/min with UV detection at 210 nm. The retention times for the vitamins were obtained as 3.61 min, 6.37 min, 9.51 min and 11.51 min for Vitamins B2, B3, B5 and B6, respectively. These obtained values of the vitamins were compared with the values available in published literatures of Deshio Khaddar Pustiman (DKPM), Indian food value (IFV) and United States Department of Agriculture (USDA).

Development of generic drug product by pharmaceutical industry is a scientific and technical approach which is totally different from developing a reference or innovator product. Most of the developing countries focus on developing the generic drug products because huge amount of investment is required for innovation and to develop reference product. The generic medicine has to be bioequivalent to the innovator drug and ensure the same biological effect with proper safety and efficacy. Nowadays, the pharmaceutical industries focus on the development of generic product as this does not require that much time and cost compared to the innovator company. But development of generic product is also difficult as it contains the same therapeutic efficacy as innovator. The development approach is based on the target market, i.e. US market, EU market. If a manufacturer targets the US market, then all excipients should be USP grade, analysis should be conducted by USP method or in-house method and stability studies as well. Prior and during the development of generic drug product API selection, dosage form selection, reference product selection and characterization, formulation development, analytical method development, tech transfer or submission batch are prime concern. Then again, bioequivalence study, drug registration procedure and commercialization of the generic product considering regulatory guidance of respective regulatory agencies and the approaches taken by the regulatory agencies for the development of registration of generic medicines are also crucial as well for the development of generic drug product. The aim of this study was to review the entire stage of a generic drug development by a generic pharmaceutical company.

Dhaka is one of the most densely-populated megacities in the world. Water supply problem is acute in this city, but the quality of drinking water has tremendous importance as potability. This study was aimed at comparing various physicochemical and microbiological parameters which are very much relevant for the drinking water quality of Dhaka city of Bangladesh. A total of 80 samples from 11 collection point of different places were collected over ten months. After that, the samples were examined by a different measuring device for physicochemical parameter testing. Considering physicochemical parameters, all water samples were within the limit for pH. However, 53.75% water had unsatisfactory for chlorine level. Total Dissolve Solid (TDS), hardness, iron, and alkalinity based unsatisfactory were 28.75%, 15.0%, 8.75%, and 3.75% respectively. Five parameters had taken into consideration to measure the microbiological quality. The cultural and biochemical methods showed that 80 water samples have a different range of contamination. The total aerobic count was unsatisfactory for 53.75% samples. Sixty-one samples were found coliform contaminated that is 76.25% of the total samples and 58.75% sample was carried fecal substances. E. coli and Vibrio was unsatisfactory as 61.25% and13.75% respectively. This study revealed that how much safe is supplied water of a municipal water supplier.
